Glycosylation engineering of antibodies for improving antibody-dependent cellular cytotoxicity
View Patent ↗The present invention relates to the field glycosylation engineering of proteins. More particular, the present invention is directed to the glycosylation engineering of proteins to provide proteins with improved therapeutic properties, e.g., antibodies, antibody fragments, or a fusion protein that includes a region equivalent to the Fc region of an immunoglobulin, with enhanced Fc-mediated cellular cytotoxicity.
1. A recombinant antibody having a Fc region with variant glycoforms obtained from a Chinese Hamster Ovary host cell that has altered glycosyltransferase expression, wherein said antibody has at least about 33% increased maximal ADCC activity compared to the corresponding antibody lacking the variant glycoforms produced by a CHO cell that does not have altered glycosyltransferase expression.
2. The recombinant antibody of claim 1 , wherein said antibody is a humanized antibody.
3. The recombinant antibody of claim 1 , wherein said antibody is an antibody fragment that contains the Fc region.
4. The recombinant antibody of claim 1 , wherein said variant glycoforms comprise an increase in the proportion of GlcNAc residues.
5. The recombinant antibody of claim 1 , wherein said variant glycoforms comprise a decrease in the proportion of fucose residues.
6. The recombinant antibody of claim 1 , wherein said variant glycoforms comprise an increase in the proportion of GlcNAc residues relative to the proportion of fucose residues.
7. The recombinant antibody of claim 1 , wherein said variant glycoforms comprise bisected N-linked oligosaccharides.
8. The recombinant antibody of claim 7 , wherein said bisected N-linked oligosaccharides are selected from the group consisting of bisected, complex and bisected, hybrid.
9. The recombinant antibody of claim 1 , wherein said antibody is a therapeutic antibody.
10. The recombinant antibody of claim 1 , wherein said antibody selectively binds to a cancer antigen.

The recombinant antibody of claim 10 , wherein said antibody is a selected from the group consisting of: an anti-CD20 antibody, an anti-human neuroblastoma antibody, an anti-human renal cell carcinoma antibody, an anti-HER2 antibody, an anti-human colon, lung, and breast carcinoma antibody, an anti-human 17-1A antigen antibody, a humanized anti-human colorectal tumor antibody, an anti-human melanoma antibody, and an anti-human squamous-cell carcinoma antibody.
12. The recombinant antibody of claim 1 , wherein said antibody is IgG.
13. The recombinant antibody of claim 7 , wherein the majority of N-linked oligosaccharides are bisected.
14. The recombinant antibody of claim 1 , wherein the majority of N-linked oligosaccharides are nonfucosylated.
15. The recombinant antibody of claim 1 , wherein the majority of N-linked oligosaccharides are bisected, nonfucosylated.
