Nanoparticles for protein drug delivery
View Patent ↗The invention discloses the nanoparticles composed of chitosan, poly-γ-glutamic acid, and at least one bioactive agent characterized with a positive surface charge and their enhanced permeability for paracellular drug delivery.
1. A pharmaceutical composition of nanoparticles for vaccine delivery comprising:
(a) a vaccine,
(b) a low molecular weight chitosan, and
(c) a polyglutamic acid or salt thereof, wherein said nanoparticles have a net positively charged surface.
2. The pharmaceutical composition of claim 1 , wherein said polyglutamic acid is γ-polyglutamic acid (γ-PGA) or α-polyglutamic acid (α-PGA).
3. The pharmaceutical composition of claim 1 , wherein said nanoparticles are further encapsulated in a capsule.
4. The pharmaceutical composition of claim 1 , wherein the low molecular weight chitosan has a molecular weight of 80 kDa.
5. The pharmaceutical composition of claim 1 , wherein the low molecular weight chitosan has a molecular weight of 40 kDa.
6. The pharmaceutical composition of claim 1 , wherein the low molecular weight chitosan is trimethyl chitosan, N,O-carboxylmethyl chitosan, or chitosan modified with a side chain selected from the group consisting of alkyl group and polyethylene glycol.
7. The pharmaceutical composition of claim 1 , wherein the composition is in a tablet form with one or more excipients.
8. The pharmaceutical composition of claim 1 , wherein said vaccine is a prophylactic vaccine.
9. The pharmaceutical composition of claim 1 , wherein said vaccine is a therapeutic vaccine.
10. The pharmaceutical composition of claim 1 , wherein the composition is formulated for oral administration.
11. The pharmaceutical composition of claim 1 , wherein the composition is formulated for parenteral administration.
12. The pharmaceutical composition of claim 1 , wherein said nanoparticles are prepared via a simple and mild ionic-gelation method.
13. The pharmaceutical composition of claim 1 , wherein said nanoparticles are crosslinked with a crosslinking agent or with infrared irradiation.
14. The pharmaceutical composition of claim 13 , wherein a degree of crosslinking of said nanoparticles is less than 50%.
15. The pharmaceutical composition of claim 13 , wherein a degree of crosslinking of said nanoparticles is ranged between 1% and 20%.
