Abstract

Compositions and methods comprising “lymphotoxin-β receptor blocking agents” which block lymphotoxin-β receptor signalling and are useful for altering immunological diseases, and particularly antibody mediated immune responses.

Claims (29)

1. A method for inhibiting a humoral immune response in a subject comprising administering to the subject a pharmaceutical composition comprising an effective amount of an antibody directed against surface lymphotoxin (LT) ligand, and a pharmaceutically acceptable carrier, such that a humoral immune response is inhibited.

2. The method according to claim 1 , wherein the antibody directed against surface LT ligand is an antibody that binds to surface LT ligand without binding to secreted LTα.

3. The method according to claim 1 , wherein the antibody directed against surface LT ligand binds to LTβ.

4. The method according to claim 3 , wherein LTβ is human LTβ.

5. The method according to claim 1 , wherein the antibody directed against surface LT ligand functions as an LTβR blocking agent.

6. A method for inhibiting a humoral immune response by inhibiting LT-β receptor signaling without inhibiting TNF-R signaling in a subject comprising administering to a subject a pharmaceutical composition comprising an amount of an antibody directed against surface LT ligand, and a pharmaceutically acceptable carrier, such that a humoral immune response is inhibited by inhibiting LT-β receptor signaling without inhibiting TNF-R signaling.

7. The method according to claim 6 , wherein the antibody directed against surface LT ligand is an antibody that binds to surface LT ligand without binding to secreted LTα.

8. The method according to claim 6 , wherein the antibody directed against surface LT ligand binds to LTβ.

9. The method according to claim 8 , wherein LTβ is human LTβ.

10. The method according to claim 6 , wherein the antibody directed against surface LT ligand functions as an LTβR blocking agent.

11. A method for disrupting the association of immune complexes and B cell follicles in a subject comprising administering to the subject a pharmaceutical composition comprising an amount of an antibody directed against surface LT ligand, and pharmaceutically acceptable carrier, such that the association of immune complexes and B cell follicles is disrupted.

12. The method according to claim 11 , wherein the antibody directed against surface LT ligand is an antibody that binds to surface LT ligand without binding to secreted LTα.

13. The method according to claim 11 , wherein the antibody directed against surface LT ligand binds to LTβ.

14. The method according to claim 13 , wherein LTβ is human LTβ.

15. The method according to claim 11 , wherein the antibody directed against surface LT ligand functions as an LTβR blocking agent.

A method of inhibiting a humoral response in a subject suffering from a hypersensitivity response, comprising administering to the subject a pharmaceutical composition comprising an effective amount of an antibody directed against surface LT ligand, and a pharmaceutically acceptable carrier, such that a humoral response is inhibited.

17. The method according to claim 16 , wherein the antibody directed against surface LT ligand is an antibody that binds to surface LT ligand without binding to secreted LTα.

18. The method according to claim 16 , wherein the antibody directed against surface LT ligand binds to LTβ.

19. The method according to claim 18 , wherein LTβ is human LTβ.

20. The method according to claim 16 , wherein the antibody directed against surface LT ligand functions as an LTβR blocking agent.

21. The method of claim 16 , wherein the hypersensitivity response is a type I response.

22. The method of claim 16 , wherein the hypersensitivity response is a type II or type III response.

A method of inhibiting a humoral response associated with graft rejection in a subject comprising administering a pharmaceutical composition comprising an effective amount of an antibody directed against surface LT ligand, and a pharmaceutically acceptable carrier, such that the humoral immune response associated with graft rejection is inhibited.

24. The method according to claim 23 , wherein the antibody directed against surface LT ligand is an antibody that binds to surface LT ligand without binding to secreted LTα.

25. The method according to claim 23 , wherein the antibody directed against surface LT ligand binds to LTβ.

26. The method according to claim 25 , wherein LTβ is human LTβ.

27. The method according to claim 23 , wherein the antibody directed against surface LT ligand functions as an LTβR blocking agent.

28. A method for inhibiting a humoral immune response in a subject comprising administering a pharmaceutical composition comprising an antibody directed against surface LT ligand, and a pharmaceutically acceptable carrier, such that the humoral immune response is inhibited.

29. A method for inhibiting a humoral immune response by inhibiting LT-β receptor signaling without inhibiting TNF-R signaling in a subject comprising administering a pharmaceutical composition comprising an antibody directed against surface LT ligand, such that the humoral immune response is inhibited by inhibiting LT-β receptor signaling without inhibiting TNF-R signaling.
