Abstract

The present invention relates to Pregnane X receptor (PXR) polypeptides and crystals that are useful, for example, for crystallization and in assays for identification of modulators of PXR.

Claims (11)

1. A crystalline composition comprising:

(a) a crystal of a polypeptide comprising the amino acid sequence set forth in SEQ ID NO: 24, wherein the crystal is in space group P2 1 2 1 2 1 and comprises unit cell dimensions a=84.2 Å, b=90.0 Å, c=106.6 Å, α=β=γ=90°;

(b) a crystal of a polypeptide comprising the amino acid sequence set forth in SEQ ID NO: 24, wherein the crystal is in space group P4 3 2 1 2 and comprises unit cell dimensions a=b=94.2 Å, c=88.2 Å, α=β=γ=90°; or

(c) a crystal of a polypeptide comprising the amino acid sequence set forth in SEQ ID NO: 24 complexed with

wherein the crystal is in space group P2 1 2 1 2 1 and comprises unit cell dimensions a=86.0 Å, b=89.3 Å, c=106.0 Å, α=β=γ=90°.

The crystalline composition of claim 1 , wherein crystal (a) comprises a first and a second monomer of the polypeptide, wherein the first monomer of the polypeptide is characterized by the structural coordinates set forth in Table 3a and the second monomer of the polypeptide is characterized by the structural coordinates set forth in Table 3b.

3. The crystalline composition of claim 1 , wherein the polypeptide of crystal (b) is characterized by the structural coordinates of Table 4a.

The crystalline composition of claim 1 , wherein crystal (c) comprises a first and a second monomer of the polypeptide, wherein the first monomer of the polypeptide is characterized by the structural coordinates set forth in Table 5a, the second monomer of the polypeptide is characterized by the structural coordinates set forth in Table 5b, and SR12813 complexed with said polypeptides is characterized by the structural coordinates set forth in Table 5c.

5. The crystalline composition of claim 1 wherein crystal (a) diffracts X-rays to a resolution of 2.3 Å.

6. The crystalline composition of claim 1 wherein crystal (b) diffracts X-rays to a resolution of 2.2 Å.

7. The crystalline composition of claim 1 wherein crystal (c) diffracts X-rays to a resolution of 2.2 Å.
