Abstract

Compositions and methods for alleviating the symptoms associated with chronic fatigue syndrome and fibromyalgia syndrome are provided. The compositions are based on use of a transdermal gel formulation delivery system for androgens, either alone or in combination with other hormones.

Claims (17)

1. A method of alleviating the symptoms of fibromyalgia in a human female patient suffering from deficient serum testosterone levels, comprising administering daily to the patient suffering from deficient serum testosterone levels, a safe and effective amount of a transdermal testosterone gel composition which is both effective for alleviating the female patient's symptoms of fibromyalgia and for consistently raising the female patient's serum testosterone levels only within limits approximating the reference range for normal premenopausal women, wherein the composition contains a daily unit dose of testosterone and is formulated to provide steady state total testosterone serum levels without raising free testosterone serum levels or twenty-four hour free testosterone AUC above the levels required for both therapeutic efficacy and safety.

A method of alleviating the symptoms of fibromyalgia in a human female patient suffering from deficient serum testosterone levels, comprising administering daily to the female patient a transdermal testosterone gel composition that is safe and effective for raising a human female patient's serum testosterone levels only within limits approximating the reference range for normal premenopausal women, wherein steady state concentrations, defined by mean concentrations through an approximately 24 hour sampling period, established after Day 1 and are within limits approximating the reference range for normal premenopausal women.

A method of increasing serum testosterone levels in a human female patient deficient in serum testosterone levels, comprising administering daily to the female patient a transdermal testosterone gel composition that is safe and effective for raising a human female patient's serum testosterone levels only within limits approximating the reference range for normal premenopausal women, wherein a mean serum concentration of total testosterone 24 hours after application of a first dose of the composition on Day 1 is higher than a mean serum concentration for time zero on Day 1 and, on average, is sustained throughout an approximately 28 day period and remains within limits approximating the reference range for normal premenopausal women.

A method of increasing serum testosterone levels in a human female patient deficient in serum testosterone levels comprising administering daily to the female patient a transdermal testosterone gel composition that is safe and effective for raising a human female patient's serum testosterone levels only within limits approximating the reference range for normal premenopausal women, wherein a mean serum concentration of total testosterone 24 hours after application of a first dose of the composition on Day 1 is higher than a mean serum concentration for time zero on Day 1 and steady state concentrations, defined by mean concentrations throughout an approximately 24 hour sampling period, on average, are established after Day 1 and remain within limits approximating the reference range for normal premenopausal women.

5. The method of claim 4 , wherein steady state concentrations are defined by mean concentrations throughout the day for a full 24 hour sampling period.

6. The method of claim 5 , wherein the steady state concentrations are established by about Day 28.

7. The method of claim 4 , wherein steady state concentrations are statistically significant and remain within limits approximating the reference range for normal premenopausal women.

8. The method of claim 4 , wherein the method treats chronic fatigue syndrome.

9. The method of claim 4 , wherein the method treats fibromyalgia.

10. The method of claim 4 , wherein the method treats decreased libido.

11. The method of claim 4 , wherein the testosterone is selected from the group consisting of testosterone, dihydrotestosterone, androstenedione, methyltestosterone, and a testosterone ester.

12. The method of claim 11 , wherein the testosterone ester is selected from the group consisting of testosterone enanthate and testosterone cypionate.

13. The method of claim 4 , wherein the testosterone is selected from the group consisting of methyltestosterone, testosterone enanthate, and testosterone cypionate.

14. The method of claim 4 , wherein free testosterone serum levels do not rise above about 4.69 pg/ml.

15. The method of claim 4 , wherein 24-hour free testosterone AUC levels do not rise above about 71.38 pg-h/ml.

16. The method of claim 4 , wherein the increase in serum testosterone levels provides steady state total testosterone serum level ranges from about 0.9 ng/ml to about 1.4 ng/ml.

17. A method of increasing serum testosterone levels in a human female patient deficient in serum testosterone levels comprising administering daily to the female patient a transdermal testosterone gel composition that is safe and effective for raising a human female patient's serum testosterone levels only within limits approximating the reference range for normal premenopausal women, wherein steady state concentrations, defined by mean concentrations throughout the day for an approximately 24 hour sampling period, on average, are established after Day 1 and remain within limits approximating the reference range for normal premenopausal women.
