Abstract

This invention provides antibodies that interact with or bind to human nerve growth factor (NGF) and neutralize the function of NGF thereby. The invention also provides pharmaceutical compositions of these antibodies and methods for neutralizing NGF function, and particularly for treating NGF-related disorders (e.g., chronic pain) by administering a pharmaceutically effective amount of anti-NGF antibodies. Methods of detecting the amount of NGF in a sample using anti-NGF antibodies are also provided.

Claims (8)

1. An isolated monoclonal antibody that specifically binds to an epitope on human nerve growth factor (NGF), said isolated monoclonal antibody comprising a heavy chain variable region, wherein the heavy chain variable region comprises CDR1 (SEQ ID NO: 22), CDR2 (SEQ ID NO: 18), and CDR3 (SEQ ID NO: 14) of SEQ ID NO: 10, and a light chain variable region, wherein the light chain variable region comprises CDR1 (SEQ ID NO: 24), CDR2 (SEQ ID NO: 20), and CDR3 (SEQ ID NO: 16) of SEQ ID 12.

2. The isolated monoclonal antibody of claim 1 , wherein the isolated monoclonal antibody is fully human.

3. The isolated monoclonal antibody of claim 1 , wherein said epitope comprises a plurality of determinants in regions of human NGF comprising the amino acid sequences of SEQ ID NOs: 56, 65, 71, 72, 74, 75, and 76.

4. An antigen binding fragment of the isolated monoclonal antibody of claim 1 .

5. The antigen binding fragment of claim 4 selected from the group consisting of a F(ab), a F(ab)′, and a (Fab′) 2 .

6. The isolated monoclonal antibody of claim 1 , wherein the isolated monoclonal antibody is a single chain Fv antibody.

7. The isolated monoclonal antibody of claim 1 , wherein the isolated monoclonal antibody is a chimeric or humanized antibody.

An isolated monoclonal antibody that specifically binds to an epitope on human nerve growth factor (NGF), wherein said epitope comprises a plurality of determinants in regions of human NGF comprising the amino acid sequences of SEQ ID NOs: 56, 65, 71, 72, 74, 75, and 76 and wherein said monoclonal antibody competes for binding to NGF with a monoclonal antibody according to claim 1 and inhibits NGF induced survival in an in vitro neutralization assay with an IC 50 of about 1×10 −8 M or less.
