Abstract

The present invention relates to isolated polypeptides having endoglucanase activity and isolated polynucleotides encoding the polypeptides. The invention also relates to nucleic acid constructs, vectors, and host cells comprising the polynucleotides as well as methods for producing and using the polypeptides.

Claims (28)

1. An isolated polynucleotide encoding a polypeptide having endoglucanase activity, selected from the group consisting of:

(a) a polynucleotide encoding a polypeptide having endoglucanase activity comprising an amino acid sequence having at least 95% sequence identity with the mature polypeptide of SEQ ID NO: 2;

(b) a polynucleotide encoding a polypeptide having endoglucanase activity which hybridizes under high stringency conditions with (i) the mature polypeptide coding sequence of SEQ ID NO: 1, (ii) the genomic DNA sequence comprising the mature polypeptide coding sequence of SEQ ID NO: 1, or (iii) a full-length complementary strand of (i) or (ii); and

(c) a polynucleotide encoding a polypeptide having endoglucanase activity comprising a nucleotide sequence having at least 95% identity with the mature polypeptide coding sequence of SEQ ID NO: 1.

2. The isolated polynucleotide of claim 1 , which encodes a polypeptide comprising an amino acid sequence having at least 95% sequence identity with the mature polypeptide of SEQ ID NO: 2.

3. The isolated polynucleotide of claim 2 , which encodes a polypeptide comprising an amino acid sequence having at least 97% sequence identity with the mature polypeptide of SEQ ID NO: 2.

4. The isolated polynucleotide of claim 1 , which encodes a polypeptide comprising or consisting of the amino acid sequence of SEQ ID NO: 2.

5. The isolated polynucleotide of claim 1 , which encodes a polypeptide comprising or consisting of the mature polypeptide of SEQ ID NO: 2.

The isolated polynucleotide of claim 1 , which hybridizes under high stringency conditions with (i) the mature polypeptide coding sequence of SEQ ID NO: 1, (ii) the genomic DNA sequence comprising the mature polypeptide coding sequence of SEQ ID NO: 1, or (iii) a full-length complementary strand of (i) or (ii).

The isolated polynucleotide of claim 6 , which hybridizes under very high stringency conditions with (i) the mature polypeptide coding sequence of SEQ ID NO: 1, (ii) the genomic DNA sequence comprising the mature polypeptide coding sequence of SEQ ID NO: 1, or (iii) a full-length complementary strand of (i) or (ii).

8. The isolated polynucleotide of claim 1 , which comprises a nucleotide sequence having at least 95% sequence identity with the mature polypeptide coding sequence of SEQ ID NO: 1.

9. The isolated polynucleotide of claim 8 , which comprises a nucleotide sequence having at least 97% sequence identity with the mature polypeptide coding sequence of SEQ ID NO: 1.

10. The isolated polynucleotide of claim 1 , which is contained in plasmid pTter7F which is contained in E. coli NRRL B-30837.

11. A nucleic acid construct comprising the polynucleotide of claim 1 operably linked to one or more control sequences that direct the production of the polypeptide in an expression host.

12. A recombinant expression vector comprising the polynucleotide of claim 1 .

13. A recombinant host cell comprising the polynucleotide of claim 1 .

14. A method for producing the polypeptide having endoglucanase activity, comprising: (a) cultivating the host cell of claim 13 under conditions conducive for production of the polypeptide; and (b) recovering the polypeptide.

15. A method for producing a mutant of a parent cell, which comprises disrupting or deleting the polynucleotide of claim 1 , which results in the mutant producing less of the polypeptide having endoglucanase activity than the parent cell.

16. A nucleic acid construct comprising a gene encoding a protein operably linked to a nucleotide sequence encoding a signal peptide comprising or consisting of amino acids 1 to 17 of SEQ ID NO: 2, wherein the gene is foreign to the nucleotide sequence.

17. A recombinant expression vector comprising the nucleic acid construct of claim 16 .

18. A recombinant host cell comprising the nucleic acid construct of claim 16 .

19. A method for producing a protein comprising: (a) cultivating the recombinant host cell of claim 18 under conditions conducive for production of the protein; and (b) recovering the protein.

20. The isolated polynucleotide of claim 1 , which encodes a polypeptide comprising an amino acid sequence having at least 98% sequence identity with the mature polypeptide of SEQ ID NO: 2.

21. The isolated polynucleotide of claim 1 , which encodes a polypeptide comprising an amino acid sequence having at least 99% sequence identity with the mature polypeptide of SEQ ID NO: 2.

22. The isolated polynucleotide of claim 1 , which comprises a nucleotide sequence having at least 98% sequence identity with the mature polypeptide coding sequence of SEQ ID NO: 1.

23. The isolated polynucleotide of claim 1 , which comprises a nucleotide sequence having at least 99% sequence identity with the mature polypeptide coding sequence of SEQ ID NO: 1.

24. The isolated polynucleotide of claim 1 , wherein the mature polypeptide is amino acids 18 to 336 of SEQ ID NO: 2.

25. The isolated polynucleotide of claim 1 , wherein the mature polypeptide coding sequence is nucleotides 52 to 1008 of SEQ ID NO: 1.
