Abstract

The present invention relates to regulation of cold sensation and pain. More particularly, the present invention is directed to nucleic acids encoding a member of the transient regulatory protein family, CMR1, which is involved in modulation of the perception of cold sensations and pain. The invention further relates to methods for identifying and using agents that modulate cold responses and pain responses stimulated by cold via modulation of CMR1 and CMR1-related signal transduction.

Claims (25)

1. A method for identifying a compound that modulates a cold-and-menthol-sensitive receptor, the method comprising:

(i) contacting a cell comprising a cold- and menthol-sensitive receptor (CMR1) polypeptide with the compound, wherein the CMR1 polypeptide comprises the sequence of SEQ ID NO:1 and forms a cation channel; and

(ii) determining an effect of the compound upon the cell comprising the CMR1 polypeptide, and

(iii) comparing the effect of the compound to that of a CMR1 agonist selected from the group consisting of cold temperature, menthol, menthone, eucalyptol, and icilin,

thereby identifying a compound that modulates the cold- and-menthol-sensitive receptor.

2. The method of claim 1 , wherein the CMR1 polypeptide is encoded by a nucleic acid having at least 95% identity to SEQ ID NO:2.

3. The method of claim 1 , wherein the CMR1 polypeptide is encoded by a nucleic acid having the sequence set forth in SEQ ID NO:2.

4. The method of claim 1 , further comprising a step of stimulating the cell with cold temperature.

5. The method of claim 1 , wherein the identified compound is a CMR1 agonist.

6. The method of claim 1 , wherein the identified compound is a CMR1 antagonist.

7. The method of claim 1 , wherein the ability of said compound to modulate cold sensation is confirmed by contacting a mammal with said compound and assessing whether said compound modulates cold sensation.

8. The method of claim 1 , wherein the cation channel responds to cool or cold temperatures.

9. The method of claim 1 , wherein the cation channel responds to a compound that elicits a cool or cold sensation.

10. A method for identifying a compound that modulates a cold-and-menthol-sensitive receptor, the method comprising the steps of:

(i) contacting a cell comprising a cold- and menthol-sensitive receptor (CMR1) polypeptide with a CMR1 agonist and the compound, wherein the CMR1 polypeptide comprises the sequence of SEQ ID NO:1 and forms a cation channel, and wherein the CMR1 agonist is selected from the group consisting of cold temperature, menthol, menthone, eucalyptol, and icilin; and

(ii) determining the effect of the compound and the CMR1 agonist upon the cell comprising the CMR1 polypeptide by comparing it to the effect of the agonist alone;

wherein a difference between the effect of the compound and the agonist and the effect of the agonist alone indicates that the compound modulates cold- and-menthol-sensitive receptor activity.

11. The method of claim 10 , wherein the CMR1 polypeptide is encoded by a nucleic acid having at least 95% identity to SEQ ID NO:2.

12. The method of claim 10 , wherein the CMR1 polypeptide is encoded by a nucleic acid having the sequence set forth in SEQ ID NO:2.

13. The method of claim 10 , wherein the cation channel responds to cool or cold temperatures.

14. The method of claim 10 , wherein the cation channel responds to a compound that elicits a cool or cold sensation.

15. A recombinant cell for identifying a compound that modulates cold-and menthol-sensitive receptor (CMR1) activity, the cell comprising a CMR1 polypeptide, wherein the CMR1 polypeptide comprises SEQ ID NO:1.

16. The recombinant cell of claim 15 , wherein the CMR1 polypeptide is encoded by a nucleic acid having the sequence set forth in SEQ ID NO:2.

17. The recombinant cell of claim 15 , wherein the cation channel responds to cool or cold temperatures.

18. The recombinant cell of claim 15 , wherein the cation channel responds to a compound that elicits a cool or cold sensation.
