Abstract

Disclosed herein are compounds that form covalent bonds with Bruton's tyrosine kinase (Btk). Also described are irreversible inhibitors of Btk, such as those having the structure of Formula (A) Methods for the preparation of the compounds are disclosed. Also disclosed are pharmaceutical compositions that include the compounds. Methods of using the Btk inhibitors are disclosed, alone or in combination with other therapeutic agents, for the treatment of autoimmune diseases or conditions, heteroimmune diseases or conditions, cancer, including lymphoma, and inflammatory diseases or conditions.

Claims (60)

1. A method of treating a blood cell cancer comprising administering to a subject in need thereof a composition containing a therapeutically effective amount of an inhibitor of a tyrosine kinase, wherein the tyrosine kinase is interleukin-2-inducible tyrosine kinase (ITK) or Bruton's tyrosine kinase wherein the inhibitor has the structure of Formula (A):

wherein:

A is N;

R 1 is L 2 -(substituted or unsubstituted heteroaryl), or L 2 -(substituted or unsubstituted aryl), where L 2 is a bond, O, S, —S(═O), —S(═O) 2 , C(═O), -(substituted or unsubstituted C 1 -C 6 alkylene), or -(substituted or unsubstituted C 2 -C 6 alkenylene);

R 2 and R 3 are independently selected from H, lower alkyl and substituted lower alkyl;

R 4 is L 3 -X-L 4 -G, wherein,

L 3 is optional, and when present is an optionally substituted or unsubstituted alkylene, optionally substituted or unsubstituted cycloalkylene, optionally substituted or unsubstituted alkenylene, optionally substituted or unsubstituted alkynylene;

X is optional, and when present is O, —C(═O), S, —S(═O), —S(═O) 2 , —NH, —NR 9 , —NHC(O), —C(O)NH, —NR 9 C(O), —C(O)NR 9 , —S(═O) 2 NH, —NHS(═O) 2 , —S(═O) 2 NR 9 —, —NR 9 S(═O) 2 , —OC(O)NH—, —NHC(O)O—, —OC(O)NR 9 —, —NR 9 C(O)O—, —CH═NO—, —ON═CH—, —NR 10 C(O)NR 10 —, heteroarylene, arylene, —NR 10 C(═NR 11 )NR 10 —, —NR 10 C(═NR 11 )—, —C(═NR 1 )NR 10 —, —OC(═NR 11 )—, or —C(═NR 11 )O—;

L 4 is optional, and when present is a substituted or unsubstituted alkylene, substituted or unsubstituted cycloalkylene, substituted or unsubstituted alkenylene, substituted or unsubstituted alkynylene, substituted or unsubstituted arylene, substituted or unsubstituted heteroarylene, substituted or unsubstituted heterocycle;

or L 3 , X and L 4 taken together form a nitrogen containing heterocyclic ring;

G is

 wherein,

R 6 , R 7 and R 8 are independently selected from among H, lower alkyl or substituted lower alkyl, lower heteroalkyl or substituted lower heteroalkyl, substituted or unsubstituted lower cycloalkyl, and substituted or unsubstituted lower heterocycloalkyl;

R 9 is selected from among H, substituted or unsubstituted lower alkyl, and substituted or unsubstituted lower cycloalkyl;

each R 10 is independently H, substituted or unsubstituted lower alkyl, or substituted or unsubstituted lower cycloalkyl; or

two R 10 groups can together form a 5-, 6-, 7-, or 8-membered heterocyclic ring; or

R 10 and R 11 can together form a 5-, 6-, 7-, or 8-membered heterocyclic ring; and

R 11 is selected from H, —S(═O) 2 R 8 , —S(═O) 2 NH 2 , —C(O)R 8 , —CN, —NO 2 , heteroaryl, or heteroalkyl; or a pharmaceutically acceptable salt thereof.

2. The method of claim 1 , wherein the inhibitor has the structure of Formula D:

wherein:

L a is CH 2 , O, NH or S;

Ar is a substituted or unsubstituted aryl, or a substituted or unsubstituted heteroaryl;

Y is an optionally substituted group selected from among alkylene, heteroalkylene, cycloalkylene, heterocycloalkylene, arylene, and heteroarylene;

Z is C(═O), OC(═O), NHC(═O), C(═S), S(═O) x , OS(═O) x , NHS(═O) x , where x is 1 or 2;

R 7 and R 8 are independently selected from among H; or

R 7 and R 8 taken together form a bond; and

R 6 is H.

3. The method of claim 1 , wherein the subject is a human.

4. The method of claim 1 , wherein the inhibitor forms a covalent bond to cysteine residue 481 of Bruton's tyrosine kinase (Btk).

5. The method of claim 1 , wherein the inhibitor is an irreversible inhibitor of Bruton's tyrosine kinase (Btk).

6. The method of claim 1 , wherein the Bruton's tyrosine kinase (Btk) is activated.

7. The method of claim 2 , wherein L a is O.

8. The method of claim 2 , wherein Z is C(═O), NHC(═O), or S(═O) 2 .

9. The method of claim 2 , wherein the inhibitor of Formula (D) is selected from among:

1-(3-(4-amino-3-(4-phenoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)piperidin-1-yl)prop-2-en-1-one; 1-(3-(4-amino-3-(4-phenoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)piperidin-1-yl)sulfonylethene; 1-(3-(4-amino-3-(4-phenoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)piperidin-1-yl)prop-2-yn-1-one; 1-(4-(4-amino-3-(4-phenoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)piperidin-1-yl)prop-2-en-1-one; N-((1s,4s)-4-(4-amino-3-(4-phenoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)cyclohexyl)acrylamide; 1-((R)-3-(4-amino-3-(4-phenoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)pyrrolidin-1-yl)prop-2-en-1-one; 1-((S)-3-(4-amino-3-(4-phenoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)pyrrolidin-1-yl)prop-2-en-1-one; 1-((R)-3-(4-amino-3-(4-phenoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)piperidin-1-yl)prop-2-en-1-one; and 1-((S)-3-(4-amino-3-(4-phenoxyphenyl)-1H-pyrazolo[3,4-d]pyrimidin-1-yl)piperidin-1-yl)prop-2-en-1-one.

10. The method of claim 2 , wherein the inhibitor is a compound of Formula (D) or a pharmaceutically acceptable salt thereof:

wherein:

L a is O or S;

Ar is an unsubstituted phenyl;

Y is a 4-, 5-, 6-, or 7-membered cycloalkyl ring, or

Y is azetidinyl, pyrrolidinyl, piperidinyl, or azepanyl;

Z is C(═O), OC(═O), NHC(═O), S(═O) x , or NHS(═O) x , where x is 2;

R 8 is H; R 7 is H; or

R 7 and R 8 taken together form a bond; and

R 6 is H.

11. The method of claim 1 , wherein R 2 and R 3 are each independently H.

12. The method of claim 1 , wherein R 1 is a substituted phenyl.

13. The method of claim 1 , wherein R 4 is L 3 -X-L 4 -G wherein L 3 , X and L 4 taken together form a nitrogen containing heterocyclic ring.

14. The method of claim 1 , wherein G is

15. The method of claim 1 , wherein R 6 , R 7 , and R 8 are each independently H.

16. The method of claim 1 , wherein the compound has the structure

or a pharmaceutically acceptable salt thereof.

17. A method for treating a blood cell cancer comprising administering to a subject in need thereof a composition containing a therapeutically effective amount of an inhibitor of a tyrosine kinase, wherein the tyrosine kinase is interleukin-2-inducible tyrosine kinase (ITK) or Bruton's tyrosine kinase, wherein the inhibitor has the structure

or a pharmaceutically acceptable salt thereof.

18. The method of claim 17 , wherein the blood cell cancer is a mast cell malignancy.

19. The method of claim 18 , wherein the blood cell cancer is a lymphoma.

20. The method of claim 19 , wherein the lymphoma is diffuse large B-cell lymphoma, follicular lymphoma, lymphoplasmacytic lymphoma/Waldenström macroglobulinemia, splenic marginal zone lymphoma, extranodal marginal zone B cell lymphoma, nodal marginal zone B cell lymphoma, mantle cell lymphoma, mediastinal (thymic) large B cell lymphoma, intravascular large B cell lymphoma, or primary effusion lymphoma.

21. The method of claim 1 , wherein the blood cell cancer is a leukemia.

22. The method of claim 21 , wherein the leukemia is chronic lymphocytic leukemia.
