Abstract

This invention is related to the use of polyurethane-based polymer as a drug delivery device to deliver biologically active compounds at a constant rate for an extended period of time and methods of manufactures thereof. The device is very biocompatible and biostable, and is useful as an implant in patients (humans and animals) for the delivery of appropriate bioactive substances to tissues or organs.

Claims (13)

1. A method of delivering one or more active ingredients to a subject, comprising: implanting an implantable device into the subject, in which the implantable device comprises a polyurethane-based polymer having polycarbonate-based polyol segments represented by the formula:

—O—[(CH 2 ) 6 —CO 3 ] n —(CH 2 )—O—

whereby an increase in “n” represents a decrease in Flex Modulus, said polyurethane-based polymer configured to provide a cylindrically shaped reservoir, wherein the reservoir is sealed after being charged with an effective amount of a solid formulation comprising one or more active ingredients exhibiting hydrophobic characteristics, such that the release of the one or more active ingredients at a substantially zero order rate in vitro by diffusion through the polyurethane-based polymer generally increases with the selection of polyurethane-based polymers of decreasing Flex Modulus.

2. The method of claim 1 in which the release of the one or more active ingredients is modulated by selecting polyurethane-based polymers with a Flex Modulus ranging from about 620-92,000 psi.

3. The method of claim 1 in which the release of the one or more active ingredients is modulated by selecting polyurethane-based polymers with a Flex Modulus ranging from about 4,500-92,000 psi.

4. The method of claim 1 in which the selected polyurethane-based polymer has a Flex Modulus selected from 620 and 4,500 psi.

5. The method of claim 1 in which the one or more active ingredients exhibit a relative water solubility (Log P) of about 1.9 or greater.

6. The method of claim 5 in which the one or more active ingredients exhibit a relative water solubility (Log P) ranging from about 1.9 to about 3.3.

7. The method of claim 1 in which the one or more active ingredients exhibit a molecular weight (MW) of about 230 amu or greater.

8. The method of claim 7 in which the one or more active ingredients exhibit a molecular weight (MW) ranging from about 230 to about 410 amu.

9. The method of claim 1 in which the solid formulation includes one or more pharmaceutically acceptable carriers that are not required for the substantially zero order release rate in vitro of the one or more active ingredients through the polyurethane-based polymer.

10. The method of claim 9 in which the one or more pharmaceutically acceptable carriers are selected from the group consisting of croscarmellose (CC), hydroxypropylcellulose (HPC), stearic acid (SA), polyethyleneglycol (PEG) and combinations thereof.

The method of claim 1 in which the at least one active ingredient is selected from drugs that can act on the central nervous system, psychic energizers, tranquilizers, anti-convulsants, muscle relaxants, anti-parkinson, analgesic, anti-inflammatory, anesthetic, antispasmodic, muscle contractants, anti-microbials, anti-malarials, hormonal agents, sympathomimetic, cardiovascular, diuretics, and antiparasitic.
