Methods and compositions for the repair and/or regeneration of damaged myocardium
Methods, compositions, and kits for repairing damaged myocardium and/or myocardial cells including the administration cytokines are disclosed and claimed. Methods and compositions for the development of large arteries and vessels are also disclosed and claimed. The present application also discloses and claims methods and media for the growth, expansion, and activation of human cardiac stem cells.
1. A pharmaceutical composition comprising isolated adult cardiac stem cells in a therapeutically effective amount for repairing and/or regenerating damaged myocardium and a pharmaceutically acceptable carrier, wherein said adult cardiac stem cells are c-kit positive, lineage negative, and characterized by their ability to differentiate into myocytes, endothelial cells, and smooth muscle cells.
2. The pharmaceutical composition of claim 1 , wherein the cardiac stem cells are isolated from myocardial tissue.
3. The pharmaceutical composition of claim 1 , wherein the cardiac stem cells are human cells.
4. The pharmaceutical composition of claim 3 , wherein the cardiac stem cells are autologous.
5. The pharmaceutical composition of claim 1 , wherein the cardiac stem cells express c-Met and/or IGF-1 receptor.
6. The pharmaceutical composition of claim 1 , wherein the cardiac stem cells are 5-7 μm in diameter.
7. The pharmaceutical composition of claim 1 , wherein the composition comprises about 2×10 4 to about 1×10 5 isolated adult cardiac stem cells.
8. The pharmaceutical composition of claim 1 , wherein the cardiac stem cells are cultured and expanded in vitro.
9. The pharmaceutical composition of claim 1 , further comprising one or more cytokines.
10. The pharmaceutical composition of claim 9 , wherein the one or more cytokines are hepatocyte growth factor and/or insulin-like growth factor-1.
11. The pharmaceutical composition of claim 1 , wherein the composition is formulated for intramyocardial or intraarterial injection.
12. The pharmaceutical composition of claim 1 , wherein the cardiac stem cells do not express lineage markers MyoD, myogenin, Myf5, CD45, CD45RO, CD8, TER-119, MAP1b, neurofilament 200, GFAP, α-sarcomeric actin, cardiac myosin, desmin, α-cardiac actinin, factor VIII, CD31, vimentin, or α-smooth muscle actin.
