Abstract

The present invention provides methods for determining a putative antibacterial, the methods comprising determining whether the putative antibacterial inhibits GlgE or Rv3032. The present invention also provides the antibacterial, the pharmaceutical composition and the method of making the antibacterial as well as a method of treating a subject infected with a bacterial comprising administration of the antibacterial.

Claims (20)

1. A method for determining whether an agent is a putative antibacterial, the method comprising experimentally determining whether or not the agent inhibits the maltosyltransferase GIgE (GIgE) by measuring GIgE activity in the presence of the agent;

wherein determining whether the agent inhibits GIgE comprises contacting GIgE with the agent in the presence of maltose-1-phosphate and maltooligosaccharide and measuring GIgE activity;

wherein an agent that reduces GIgE activity is determined to inhibit GIgE and is identified as a putative antibacterial.

2. The method of claim 1 , wherein measuring GIgE activity comprises measuring the release of inorganic phosphate.

3. The method of claim 1 , wherein measuring GIgE activity comprises measuring maltooligosaccharide elongation or maltose 1-phosphate consumption.

4. The method of claim 1 , wherein measuring GIgE activity comprises measuring disproportionation activity.

5. The method of claim 1 , wherein determining whether the agent inhibits GIgE activity comprises contacting bacterial cells comprising GIgE with the agent and measuring GIgE activity.

6. The method of claim 1 , further comprising contacting bacterial cells with agent that inhibits GigE activity and measuring bacterial cell viability.

7. The method of claim 1 , wherein the agent is selected from the group consisting of: a chemical, small molecule, polypeptide, saccharide, protein, protein fragment, or aptamer.

8. The method of claim 1 , wherein the antibacterial is effective against mycobacteria.

9. The method of claim 1 , wherein the antibacterial is effective against an opportunistic pathogen.

10. The method of claim 6 , wherein the bacterial cells are in presence of trehalose.

11. The method of claim 6 , the method additionally comprising one or more controls comprising:

measuring bacterial cell viability in the absence of the agent; and/or

contacting bacterial cells with the agent in the absence of trehalose and measuring bacterial cell viability; and/or

contacting bacterial cells with inoperable trehalose synthase (TreS), or pea pathogenicity (Pep2) protein with the agent; and

measuring bacterial cell viability.

12. The method of claim 6 , wherein cell viability is measured after between 24 and 192 hours.

13. The method of claim 8 , wherein the mycobacteria are selected from the group consisting of Mycobacterium tuberculosis, Mycobacterium bovis, Mycobacterium bovis BCG, Mycobacterium smegmatis and δglgE Mycobacterium smegmatis.

14. The method of claim 9 , wherein the opportunistic pathogen is from genus Pseudomonas , or genus Burkholderia .
