Abstract

The invention provides a biological method of producing isoprenoids.

Claims (19)

1. A recombinant yeast having an endogenous mevalonate pathway comprising: an enzyme that converts acetyl-CoA to acetoacetyl-CoA (thiolase), an enzyme that converts acetoacetyl-CoA to hydroxymethylglutaryl-CoA (HMG Co A synthase; HMGS), an enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid (HMG Co A reductase; HMGR), an enzyme that converts mevalonic acid to phosphomevalonic acid (mevalonate kinase; MK), an enzyme that converts phosphomevalonic acid to phosphomevalonate (phosphomevalonate kinase; PMK), and an enzyme that converts phosphomevalonate to isopentenyl pyrophosphate (phosphomevalonate decarboxylase; PMD) that is capable of making an isoprenoid compound comprising at least one heterologous nucleic acid molecule encoding:

a heterologous HMGR;

a heterologous MK;

a heterologous PMK; and,

a heterologous PMD;

wherein the yeast has more copies of HMGR encoded by the endogenous and heterologous nucleic acid molecules, in total, than the number of copies of each of thiolase and HMGS and the yeast has more copies of each of MK, PMK, and PMD encoded by the endogenous and heterologous nucleic acid molecules, in total, than the number of copies of each of thiolase and HMGS.

2. The recombinant yeast of claim 1 wherein the at least one heterologous nucleic acid molecule is encoded by one recombinant nucleic acid molecule.

3. The recombinant yeast of claim 1 wherein the at least one heterologous nucleic acid molecule is encoded by a plurality of nucleic acid molecules.

4. The recombinant yeast of claim 1 wherein the at least one heterologous nucleic acid molecule is under inducible control.

5. The recombinant yeast of claim 1 wherein the at least one heterologous nucleic acid molecule includes multiple copies of a nucleic acid molecule encoding an enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid.

6. The recombinant yeast of claim 5 wherein the multiple copies are integrated into a chromosome.

7. The recombinant yeast of claim 5 wherein the multiple copies are on a plasmid.

8. The recombinant yeast of claim 1 wherein the at least one heterologous enzyme includes an enzyme that converts acetoacetyl-CoA and acetyl-CoA into hydroxymethylglutaryl-CoA.

9. The recombinant yeast of claim 1 wherein the yeast is Saccharomyces cerevisiae.

10. The recombinant yeast of claim 1 wherein the heterologous enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid is a truncated HMG-Co A reductase.

11. The recombinant yeast of claim 1 wherein the heterologous enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid is a bacterial enzyme.

12. The recombinant yeast of claim 1 wherein the heterologous enzyme that converts hydroxymethylglutaryl-CoA to mevalonic acid is a yeast enzyme.

13. The recombinant yeast of claim 1 wherein each of the heterologous enzymes is a eukaryotic enzyme.

14. The recombinant yeast of claim 1 wherein each of the heterologous enzymes is a yeast enzyme.
