DNA sequence, and recombinant preparation of the grass pollen allergen Lol p4
The present invention relates to the provision of a DNA sequence of the major grass pollen allergen Lol p 4. The invention also encompasses fragments, new combinations of partial sequences and point mutants having a hypoallergenic action. The recombinant DNA molecules and the derived polypeptides, fragments, new combinations of partial sequences and variants can be utilised for the therapy of pollen-allergic diseases. The proteins prepared by recombinant methods can be employed for in vitro and in vivo diagnosis of pollen allergies.
1. An expression vector or a cloning system comprising a polynucleotide comprising the nucleic acid sequence set forth in SEQ ID NO: 1 which is functionally linked to a heterologous expression control sequence.
2. A culture comprising a host organism transformed with the expression vector or the cloning system according to claim 1 .
3. A process for the preparation of a polypeptide comprising the amino acid sequence set forth in SEQ ID NO: 2, comprising
cultivating a host organism of claim 2 under conditions suitable for the expression of said polypeptide; and
isolating the polypeptide from the culture.
4. An isolated DNA comprising the nucleic acid sequence which encodes the polypeptide sequence set forth in SEQ ID NO: 2, wherein said nucleic acid sequence has been codon optimized for the expression of said polypeptide of SEQ ID NO: 2 by recombinant methods in a host cell that is not a cell of Lolium perenne.
5. An human expression vector or an E. coli expression vector which comprises the isolated DNA according to claim 4 and a heterologous expression control sequence.
6. A method for the treatment of an allergy triggered by a group 4 allergen of Poaceae , comprising administering to a subject in need thereof, the coding DNA sequence according to claim 4 .
