Abstract

The present disclosure generally relates to lyophilized pharmaceutical compositions comprising polymeric nanoparticles which, upon reconstitution, have low levels of greater than 10 micron size particles. Other aspects of the invention include methods of making such nanoparticles.

Claims (19)

1. A method of preventing substantial aggregation of particles in a reconstituted pharmaceutical nanoparticle composition comprising nanoparticles comprised of poly(lactic) acid-block-poly(ethylene)glycol copolymer or poly(lactic)-co-poly(glycolic) acid-block-poly(ethylene)glycol copolymer and a therapeutic agent, the method comprising:

adding a sugar selected from trehalose and sucrose, and a salt to a formulation comprising the nanoparticles such that the formulation has 10 to 500 mM ionic halide and about 5 to about 15 weight percent sucrose or about 10 to about 20 weight percent trehalose;

lyophilizing the formulation to form a lyophilized formulation;

reconstituting the lyophilized formulation, wherein the reconstituted composition does not have substantial aggregation of the nanoparticles.

2. The method of claim 1 , wherein the sugar is trehalose.

3. The method of claim 1 , wherein the sugar is sucrose.

4. The method of claim 1 , wherein the salt is calcium chloride.

5. The method of claim 1 , wherein the salt is sodium chloride.

6. The method of claim 1 , further comprising adding hydroxypropyl β-cyclodextrin to the formulation comprising the nanoparticles such that the formulation has about 5 to about 20 weight percent hydroxypropyl β-cyclodextrin.

7. The method of claim 6 , wherein the formulation has about 7 to about 12 weight percent hydroxypropyl β-cyclodextrin.

8. The method of claim 1 , wherein the poly(lactic) acid-block-poly(ethylene)glycol copolymer comprises poly(lactic acid) having a number average molecular weight of about 15 to about 20 kDa and poly(ethylene)glycol having a number average molecular weight of about 4 to about 6 kDa.

9. The method of claim 8 , wherein the poly(lactic acid) has a number average molecular weight of about 16 kDa and the poly(ethylene)glycol has a number average molecular weight of about 5 kDa.

10. The method of claim 1 , wherein reconstitution of the lyophilized formulation in less than or about 100 mL of an aqueous medium has less than 6000 microparticles of greater than or equal to 10 microns; and less than 600 microparticles of greater than or equal to 25 microns.

11. The method of claim 1 , wherein the lyophilized formulation has a nanoparticle concentration of greater than about 40 mg/mL.

12. The method of claim 1 , wherein the nanoparticles have a diameter of about 60 nm to about 140 nm.

13. The method of claim 1 , wherein the nanoparticles have a diameter of about 70 nm to about 130 nm.

14. The method of claim 1 , wherein the nanoparticles comprise about 0.2 to about 35 weight percent of the therapeutic agent.

15. The method of claim 1 , wherein the nanoparticles comprise about 4 to about 25 weight percent of the therapeutic agent.

16. The method of claim 1 , wherein the therapeutic agent is docetaxel.
