Abstract

Disclosed herein are improved osteogenic devices and methods of use thereof for repair of bone and cartilage defects. The devices and methods promote accelerated formation of repair tissue with enhanced stability using less osteogenic protein than devices in the art. Defects susceptible to repair with the instant invention include, but are not limited to: critical size defects, non-critical size defects, non-union fractures, fractures, osteochondral defects, subchondral defects, and detects resulting from degenerative diseases such as osteochondritis dessicans.

Claims (34)

1. A device for inducing local bone or cartilage formation, comprising:

an osteogenic protein selected from the group consisting of OP-1, OP-2, OP-3, BMP3, BMP4, BMP5, BMP6, BMP9, BMP10, BMP11, BMP12, BMP15, BMP16, DPP, Vgl, Vgr, 60A protein, GDF1, GDF3, GDF5, GDF6, GDF7, GDF8, GDF9, GDF10, and GDF11;

a matrix selected from the group consisting of collagen, demineralized bone, apatites, hydroxyapatites, tricalcium phosphates, and admixtures thereof; and

a low viscosity grade of an alkylcellulose; wherein when the alkylcellulose is carboxymethylcellulose, said carboxymethylcellulose has a viscosity of 10-200 cP at a concentration of 4% (w/v); and

wherein the ratio of the alkylcellulose to the matrix in said device is 1 part by weight alkylcellulose to 2-10 parts by weight matrix.

2. The device of claim 1 , wherein said osteogenic protein is selected from the group consisting of: OP-1, OP-2, BMP2, BMP4, BMP5, and BMP6.

3. The device of claim 1 , wherein said osteogenic protein is OP-1.

4. The device of claim 1 , wherein said device comprises at least two different osteogenic proteins.

5. The device of claim 1 , wherein said matrix is collagen.

6. The device of claim 1 , wherein said device comprises at least two different matrix materials.

7. The device of claim 1 , wherein said alkylcellulose is selected from the group consisting of methylcellulose, methylhydroxyethylcellulose, hydroxyethylcellulose, hydroxypropylmethylcellulose, carboxymethylcellulose, sodium carboxymethylcellulose, hydroxyalkylcelluloses, and admixtures thereof.

8. The device of claim 1 , wherein said alkylcellulose is carboxymethylcellulose or a sodium salt thereof.

9. The device of claim 1 , wherein said device comprises at least two different alkylcelluloses.

10. The device of claim 1 , further comprising a wetting agent.

11. The device of claim 10 , wherein said wetting agent is saline.

12. The device of claim 1 , wherein the ratio of the alkylcellulose to the matrix in said device is 1 part by weight alkylcellulose to 5parts by weight matrix.

13. A method for inducing local bone or cartilage formation, comprising the step of:

providing the device of claim 1 to a defect site.

14. The method of claim 13 , wherein said bone formation is endochondral bone formation.

15. The method of claim 13 , wherein said cartilage formation is articular cartilage formation.

16. The method of claim 13 , wherein said defect site is selected from the group consisting of: critical size defect, non-critical size defect, segmental non-union defect, non-union fracture, fracture, osteochondral defect, and subchondral defect.

17. The method of claim 13 , wherein the volume of said device provided to the defect site is sufficient to fill the defect site.

18. A kit for inducing local bone or cartilage formation using the device of claim 1 , the kit comprising:

(a) a first receptacle adapted to house said osteogenic protein and said matrix; and

(b) a second receptacle adapted to house said alkylcellulose,

wherein the osteogenic protein and matrix are provided in the first receptacle of part (a), and the alkylcellulose is provided in the second receptacle of part (b).

19. The kit of claim 18 , further comprising a third receptacle adapted to house a wetting agent.

20. A kit for inducing local bone or cartilage formation using the device of claim 1 , the kit comprising a receptacle adapted to house said osteogenic protein, said matrix material, and said alkylcellulose.

21. The kit of claim 20 , further comprising a second receptacle adapted to house a wetting agent.

22. The device of claim 8 , wherein said carboxymethylcellulose or said sodium salt thereof has a degree of substitution range from 0.65to 0.90.

23. The device of claim 5 , wherein the amount of the OP-1 ranges from approximately 0.125 mg to 10.0 mg.

24. The device of claim 5 , wherein the amount of the OP-1 is approximately 3.5 mg.

25. The method of claim 13 , wherein the defect site is endochondral bone, osteochondral bone, or intramembraneous bone.

26. The method of claim 13 , wherein the defect site is articular cartilage, fibrocartilage or elastic cartilage.
