Disodium salts, monohydrates, and ethanol solvates for delivering active agents
View Patent ↗The inventors have discovered that the disodium salt of certain delivery agents has surprisingly greater efficacy for delivering active agents than the corresponding monosodium salt. Furthermore, the inventors have discovered that the disodium salts of these delivery agents form solvates with ethanol and hydrates with water. The delivery agents have the formula wherein R 1 , R 2 , R 3 , and R 4 are independently hydrogen, halogen, C 1 -C 4 alkyl, or C 1 -C 4 alkoxy; and R 5 is a substituted or unsubstituted C 2 -C 16 alkylene, substituted or unsubstituted C 2 -C 16 alkenylene, substituted or unsubstituted C 1 -C 12 alkyl(arylene), or substituted or unsubstituted aryl(C 1 -C 12 alkylene). The hydrates and solvates of present invention also have surprisingly greater efficacy for delivering active agents, such as heparin and calcitonin, than their corresponding monosodium salts and free acids. The present invention provides an alcohol solvate, such as ethanol solvate, of a disodium salt of a delivery agent of the formula above. The invention also provides a hydrate of a disodium salt of a delivery agent of the formula above. Preferred delivery agents include, but are not limited to, N-(5-chlorosalicyloyl)-8-aminocaprylic acid (5-CNAC), N-(10-[2-hydroxybenzoyl]amino)decanoic acid (SNAD), and sodium N-(8-[2-hydroxybenzoyl]amino)caprylate (SNAC). The invention also provides methods of preparing the disodium salt, ethanol solvate, and hydrate and compositions containing the disodium salt, ethanol solvate, and/or hydrate.
1. An oral dosage form comprising the disodium salt of N-(5-chlorosalicyloyl)-8-aminocaprylic acid and calcitonin.
2. The oral dosage form of claim 1 , wherein the calcitonin is salmon calcitonin.
3. The oral dosage form of claim 1 , wherein the dosage form is in solid form.
4. The oral dosage form of claim 1 , wherein the dosage form is a tablet.
5. The oral dosage form of claim 1 , wherein the dosage form is a capsule.
6. The oral dosage form of claim 1 , wherein the dosage from comprises from about 50% to about 100% by weight of the disodium salt of N-(5-chlorosalicyloyl) -8-aminocaprylic acid, based upon 100% total weight of N-(5-chlorosalicyloyl)-aminocaprylic acid and salts thereof in the dosage form.
7. The oral dosage form of claim 1 , wherein the dosage from comprises from about 50% to about 90% by weight of the disodium salt of N-(5-chlorosalicyloyl) -8-aminocaprylic acid, based upon 100% total weight of N-(5-chlorosalicyloyl)-aminocaprylic acid and salts thereof in the dosage form.
8. The oral dosage form of claim 1 , wherein the dosage from comprises from about 50% to about 96% by weight of the disodium salt of N-(5-chlorosalicyloyl) -8-aminocaprylic acid, based upon 100% total weight of N-(5-chlorosalicyloyl)-aminocaprylic acid and salts thereof in the dosage form.
9. The oral dosage form of claim 1 , wherein the dosage from comprises about 90% or more by weight of the disodium salt of N-(5-chlorosalicyloyl)-8-aminocaprylic acid, based upon 100% total weight of N-(5-chlorosalicyloyl)-aminocaprylic acid and salts thereof in the dosage form.
10. The oral dosage form of claim 1 , wherein the dosage from comprises about 96% or more by weight of the disodium salt of N-(5-chlorosalicyloyl)-8-aminocaprylic acid, based upon 100% total weight of N-(5-chlorosalicyloyl)-aminocaprylic acid and salts thereof in the dosage form.
11. The oral dosage form of claim 1 , wherein the disodium salt is in a monohydrate form.
12. The oral dosage form of claim 1 , wherein the disodium salt is anhydrous.
13. The oral dosage form of claim 1 , wherein the dosage form further comprises an excipient, a diluent, a disintegrant, a lubricant, a plasticizer, a colorant, a dosing vehicle, or any combination thereof.
14. The oral dosage form of claim 1 , wherein the weight ratio of calcitonin to the disodium salt of N-(5-chlorosalicyloyl)-8-aminocaprylic acid is from about 1:300 to about 1:700.
15. The oral dosage form of claim 1 , wherein the weight ratio of calcitonin to the disodium salt of N-(5-chlorosalicyloyl)-8-aminocaprylic acid is from about 1:100 to about 1:500.
16. The oral dosage form of claim 1 , wherein the dosage form comprises a lyophilized mixture of a disodium salt of N-(5-chlorosalicyloyl)-8-aminocaprylic acid and calcitonin.
17. The oral dosage form of claim 16 , wherein the lyophilized mixture contains less than 4% by weight of water, based upon 100% total weight of the mixture.
