Reagents for directed biomarker signal amplification
Described herein are methods, compositions and articles of manufacture involving neutral conjugated polymers including methods for synthesis of neutral conjugated water-soluble polymers with linkers along the polymer main chain structure and terminal end capping units. Such polymers may serve in the fabrication of novel optoelectronic devices and in the development of highly efficient biosensors. The invention further relates to the application of these polymers in assay methods.
1. A water soluble conjugated polymer having the structure of Formula:

G 1 and G 2 are each independently selected from the group consisting of 1-9 having the structures:
linker L 1 is
and is evenly or randomly distributed along the polymer main chain, wherein each R′ is independently a halogen, C 1 -C 12 alkyl, or (C 1 -C 12 alkyl)NH 2 ;
and n is an integer from 1 to about 10,000; and
a and c define the mol % of each unit within the structure which each can be evenly or randomly repeated and where a is a mol % from about 75 to about 99% and c is mol % from about 1 to about 25%.
2. The water soluble conjugated polymer of claim 1 wherein the linker L 1 is
and is about 10% of the entire polymer.
3. The water soluble conjugated polymer of claim 1 having the structure:

The water soluble conjugated polymer of claim 1 , further comprising a signaling chromophore selected from the group consisting of rhodamine, coumarin, xanthenes, cyanine, polymethine, pyrene, dipyrromethene borondifluoride, napthalimide, phycobiliprotein, peridinum chlorophyll protein, conjugates thereof, and combinations thereof.
5. The water soluble conjugated polymer of claim 1 , further comprising a signaling chromophore that is a cyanine dye.
6. The water soluble conjugated polymer of claim 1 , further comprising a signaling chromophore that is rhodamine or xanthene dye.
7. The water soluble conjugated polymer of claim 1 , wherein the polymer has a purity of at least 80%.
8. The water soluble conjugated polymer of claim 1 , wherein the polymer has a purity of at least 95%.
9. The water soluble conjugated polymer of either claim 7 or claim 8 , wherein G 1 and G 2 are each independently selected from the group consisting of 5-9.
10. The water soluble conjugated polymer of claim 1 , wherein the polymer is conjugated to an antibody.
11. The water soluble conjugated polymer of claim 1 , wherein the polymer is conjugated to an antibody and wherein the polymer-antibody conjugate has a purity of at least 95%.
12. An assay method for detecting a target biomolecule in a sample comprising cells comprising:
providing a sample that is suspected of containing a target biomolecule;
providing a sensor protein conjugated to a water soluble conjugated polymer of claim 1 that is further conjugated to at least one signaling chromophore and wherein the sensor protein is capable of interacting with the target biomolecule or a target-associated biomolecule;
contacting the sample with the sensor protein and the conjugated polymer in a solution under conditions in which the sensor protein can bind to the target biomolecule or a target-associated biomolecule if present;
applying a light source to the sample that can excite the conjugated polymer; and
detecting whether light is emitted from the signaling chromophore.
13. A flow cytometry system comprising:
a flow cytometer;
a sample that is suspected of containing a target biomolecule;
a sensor protein conjugated to a water soluble conjugated polymer of claim 1 that is further conjugated at least one signaling chromophore and wherein the sensor protein is capable of interacting with the target biomolecule or a target-associated biomolecule.
