Method and molecular diagnostic device for detection, analysis and identification of genomic DNA
View Patent ↗At least one exemplary embodiment of the invention is directed to a molecular diagnostic device that comprises a cartridge configured to eject samples comprising genomic material into a microfluidic chip that comprises an amplification area, a detection area, and a matrix analysis area.
1. A microfluidic system comprising:
a cartridge configured to isolate genomic material comprising:
a reaction chamber;
an ejector head;
a waste outlet;
wherein the reaction chamber is connected to the ejector head and with the waste outlet by a first channel and second channel, respectively;
at least one binding and release substrate, wherein the at least one binding and release substrate lies within the reaction chamber and is configured to bind and release at least a portion of a sample comprising genomic material and undesirable material;
two electrodes, wherein a first electrode is positioned in the reaction chamber and a second electrode is positioned in the first channel connecting the reaction chamber and the ejector head; and
a power source to effect changes in a state of the binding and release substrate;
the genomic material releasably bound to the binding and release substrate, wherein the genomic material is released from the at least one binding and release substrate based upon a change in the state of the binding and release substrate;
wherein the two electrodes are constructed and arranged to direct charged genomic material into the ejector head while waste material is directed into the waste outlet;
and
a microfluidic chip, separate from the cartridge, comprising at least one microfluidic inline reaction channel, wherein the genomic material is ejected from the ejector head into an inlet port of the microfluidic chip across an airspace between the cartridge and the microfluidic chip.
2. The microfluidic system according to claim 1 , wherein the at least one binding and release substrate comprises charge switch material.
3. The microfluidic system according to claim 1 , wherein the power source provides an electric voltage to change the state of the at least one binding and release substrate to bind and release at least a portion of the sample is in response to the electric voltage.

The microfluidic system according to claim 1 , wherein a fluid received within the reaction chamber has a changeable ionic concentration, wherein the binding and releasing of at least a portion of the sample is in response to a difference in ionic composition of the fluid in contact with the at least one binding and release substrate, and wherein the sample is contained within the fluid.

The microfluidic system according to claim 1 , wherein a fluid received within the reaction chamber has a changeable pH, wherein the binding and releasing of at least a portion of the sample is in response to a difference in pH of the fluid in contact with the at least one binding and release substrate, and wherein the sample is contained within the fluid.
6. The microfluidic system according to claim 1 , wherein the at least one binding and release substrate is a particle or bead.
7. The microfluidic system according to claim 1 , wherein the at least one binding and release substrate is magnetic or paramagnetic.
8. The microfluidic system according to claim 1 , wherein the at least one binding and release substrate is bound to the inner surface of the reaction chamber.
9. The microfluidic system according to claim 1 , wherein the cartridge is configured to concentrate the genomic material prior to ejecting the genomic material into the microfluidic chip.
10. The microfluidic system according to claim 1 , wherein the microfluidic chip is configured for performing an amplification reaction.
