Abstract

A process is provided for producing peroxycarboxylic acids from carboxylic acid esters. More specifically, carboxylic acid esters are reacted with an inorganic peroxide, such as hydrogen peroxide, in the presence of an enzyme catalyst having perhydrolysis activity. The present perhydrolase catalysts are classified as members of the carbohydrate esterase family 7 (CE-7) based on the conserved structural features. Further, disinfectant formulations comprising the peracids produced by the processes described herein are provided.

Claims (20)

1. A formulation comprising:

(a) an enzyme catalyst comprising an enzyme, wherein said enzyme has perhydrolase activity and comprises a signature motif that aligns with a reference sequence SEQ ID NO: 2 using CLUSTALW, said signature motif comprising:

(i) an RGQ motif at positions corresponding to positions 118-120 of SEQ ID NO:2;

(ii) a GXSQG motif at positions corresponding to positions 179-183 of SEQ ID NO:2; and

(iii) an HE motif at positions corresponding to positions 298-299 of SEQ ID NO:2;

(b) a substrate selected from the group consisting of an ester, a glyceride, an acetylated saccharide, and a mixture thereof; and

(c) a source of peroxygen.

2. The formulation of claim 1 , wherein the substrate is

(i) an ester having the structure

wherein R 1 ═C1 to C7 straight chain or branched chain alkyl optionally substituted with an hydroxyl or a C1 to C4 alkoxy group and R 2 ═C1 to C10 straight chain or branched chain alkyl, alkenyl, alkynyl, aryl, alkylaryl, alkylheteroaryl, heteroaryl, (CH 2 CH 2 —O) n H or (CH 2 CH(CH 3 )—O) n H and n=1 to 10;

(ii) a glyceride having the structure

wherein R 1 ═C1 to C7 straight chain or branched chain alkyl optionally substituted with an hydroxyl or a C1 to C4 alkoxy group and R 3 and R 4 are individually H or R 1 C(O);

(iii) an acetylated saccharide selected from the group consisting of acetylated monosaccharides, acetylated disaccharides, and acetylated polysaccharides; or

(iv) a mixture thereof.

The formulation of claim 2 , wherein the substrate is selected from the group consisting of methyl lactate, ethyl lactate, methyl glycolate, ethyl glycolate, methyl methoxyacetate, ethyl methoxyacetate, methyl 3-hydroxybutyrate, ethyl 3-hydroxybutyrate, monoacetin, diacetin, triacetin, monopropionin, dipropionin, tripropionin, monobutyrin, dibutyrin, tributyrin, glucose pentaacetate, xylose tetraacetate, acetylated xylan, β-D-ribofuranose-1,2,3,5-tetraacetate, tri-O-acetyl-D-galactal, tri-O-acetyl-glucal, and mixtures thereof.

4. The formulation of claim 1 , wherein the signature motif of the enzyme further comprises an LXD motif at positions corresponding to positions 267-269 of SEQ ID NO:2 using CLUSTALW.

5. The formulation of claim 4 , wherein the signature motif is selected from the group consisting of SEQ ID NO:61 and an amino acid sequence at least 50% identical to SEQ ID NO:61.

6. The formulation of claim 1 , which produces a peroxycarboxylic acid selected from the group consisting of peracetic acid, perpropionic acid, perbutyric acid, perlactic acid, perglycolic acid, permethoxyacetic acid, per-β-hydroxybutyric acid, and mixtures thereof.

7. The formulation of claim 6 , wherein the peroxycarboxylic acid produced is peracetic acid.

8. The formulation of claim 1 further comprising water.
