Abstract

Nanoelectronic devices for the detection and quantification of biomolecules are provided. In certain embodiments, the devices are configured to detect and measure blood glucose levels. Also provided are methods of fabricating nanoelectronic devices for the detection of biomolecules.

Claims (28)

1. A method of immobilizing a biomolecule for sensing a target species on a substrate to fabricate a nanosensor device for sensing the target species, said method comprising:

providing a plurality of nanostructures and a quantity of an antibody configured to interact with the target species in an aqueous solution;

dispersing the plurality of nanotubes and at least some of the quantity of the antibody to thereby create a stable suspension of nanotubes and an in the aqueous solution;

providing a substrate surface having an electrode surface thereon; and simultaneously depositing nanotubes and biomolecules from the stable suspension to at least the electrode surface to thereby immobilize the biomolecules on the substrate.

2. The method of claim 1 wherein the immobilized biomolecules retain biological activity and stability.

3. The method of claim 1 wherein the plurality of nanostructures to be dispersed are pristine.

4. The method of claim 1 wherein the plurality of nanostructures are functionalized with a solubilizing chemistry.

5. The method of claim 4 wherein the a solubilizing agent is selected from PABS and carboxylic acid groups.

6. The method of claim 1 wherein the nanostructures are single walled nanotubes.

7. The method of claim 1 wherein the nanostructures are multi-walled nanotubes.

8. The method of claim 1 wherein dispersing the nanostructures and at least some of the quantity of the antibody comprises sonicating the aqueous solution.

9. The method of claim 1 wherein dispersing the nanostructures and at least some of the quantity of the antibody comprises sonicating the nanostructures in the aqueous solution prior to adding the quantity of the antibody to the solution.

10. The method of claim 1 wherein the concentration of nanostructures in the aqueous solution is between about 0.1-1 mg/ml.

11. The method of claim 1 wherein the concentration of nanostructures in the aqueous solution is about 0.1 mg/ml.

12. The method of claim 1 wherein no separation operations are performed between the dispersing and depositing operations.

13. The method of claim 1 further comprising separating unattached or loosely attached antibodies and nanostructures from the suspension prior to deposition.

14. The method of claim 1 wherein immobilizing the antibody to the substrate is done without the use of a binding agent.

15. A nanoelectronic sensor for sensing a target species comprising:

a non-conductive substrate;

a counter electrode and working electrode on said substrate, said working electrode comprising a carbon conductive path and having a surface comprising a carbon nanotube network;

a quantity of a biomolecule configured to interact with the target species in contact with said carbon nanotube network and immobilized on the working electrode;

a first layer overlying the substrate and separated therefrom by an adhesive;

a sample well defined at least in part by a gap in the adhesive and configured for receiving a sample and contacting said sample with the biomolecule.

16. The nanoelectronic sensor of claim 15 , wherein the immobilized biomolecule retains biological activity and stability.

17. The method of claim 15 , wherein the immobilized biomolecule is an enzyme, antibody or nucleic acid.

18. The method of claim 15 , wherein the biomolecule is configured to interact with glucose.

19. The method of claim 15 , wherein the biomolecule is selected from the group consisting of gluocose oxidase, PQQ-GDH and FAD-GDH.

20. The method of claim 15 , wherein the first layer is a polymeric material.
