Aβ conformer selective anti-Aβ globulomer monoclonal antibodies
View Patent ↗The subject invention relates to monoclonal antibodies that may be used in the treatment and diagnosis of Alzheimer's Disease. In particular, the present invention relates to monoclonal antibodies referred to as 10F4 and 3C5 and to other monoclonal antibodies (e.g., murine, human or humanized) having similar properties thereto.
1. A method for treating amyloidosis in a patient in need of said treatment comprising administering to said patient an isolated antibody having a higher affinity to Aβ(1-42) globulomer than to at least one amyloid beta protein selected from the group consisting of a) Aβ(1-42) peptide present in cerebrospinal fluid (CSF) and b) Aβ(1-40) peptide present in CSF in an amount sufficient to effect said treatment, wherein said antibody comprises:
a variable heavy chain region (VH) comprising the sequences of SEQ ID NOs: 9-11 and a variable light chain region (VL) comprising the sequences of SEQ ID NOs: 12-14:
a VH comprising the sequences of SEQ ID NOs: 15-17 and a VL comprising the sequences of SEQ ID NOs: 18-20;
a VH comprising the sequence of SEQ ID NO: 5;
a VL comprising the sequence of SEQ ID NO: 6;
a VH comprising the sequence of SEQ ID NO: 7; or
a VL comprising the sequence of SEQ ID NO: 8.
2. The method of claim 1 wherein said amyloidosis is Alzheimer's disease or Down's syndrome.
3. The method of claim 1 , wherein the antibody is a recombinant antibody.
4. The method of claim 1 , wherein the antibody is human or humanized.
5. The method of claim 1 , wherein said antibody binds to at least one epitope as a monoclonal antibody selected from the group consisting of the monoclonal antibody 10F4 obtainable from a hybridoma designated by American Type Culture Collection deposit number PTA-7808 and the monoclonal antibody 3C5 obtainable from a hybridoma designated by American Type Culture Collection deposit number PTA-7406.
