Abstract

The present invention provides a method of extracting and recovering embryonic-like stem cells, including, but not limited to pluripotent or multipotent stem cells, from an exsanguinated human placenta. A placenta is treated to remove residual umbilical cord blood by perfusing an exsanguinated placenta, preferably with an anticoagulant solution, to flush out residual cells. The residual cells and perfusion liquid from the exsanguinated placenta are collected, and the embryonic-like stem cells are separated from the residual cells and perfusion liquid. The invention also provides a method of utilizing the isolated and perfused placenta as a bioreactor in which to propagate endogenous cells, including, but not limited to, embryonic-like stem cells. The invention also provides methods for propagation of exogenous cells in a placental bioreactor and collecting the propagated exogenous cells and bioactive molecules therefrom.

Claims (14)

1. A method of repairing heart tissue in an individual that has suffered from myocardial infarction, comprising administering to the individual a therapeutically effective amount of isolated human placental stem cells, wherein said placental stem cells are:

OCT-4 + , CD34 − , CD10 + , CD29 + , CD44 + , CD45 − , CD54 + , CD90 + , SH3 + , SH4 + , SSEA3 − , and/or SSEA4 − , wherein OCT-4 is octamer binding protein 4;

OCT-4 + , CD34 − , SSEA3 − and SSEA4 − ;

OCT-4 + and CD34 − , and additionally SH3 + or SH4 + ; or

CD34 − and one or more of CD29 + , CD45 − , CD90 + , SH2 + , SH3 + , SH4 + , or MHC Class II − .

2. The method of claim 1 , wherein said placental stem cells are OCT-4 + , CD34 − , CD10 + , CD29 + , CD44 + , CD45 − , CD54 + , CD90 + , SH3 + , SH4 + , SSEA3 − , and SSEA4 − .

3. The method of claim 1 , wherein said placental stem cells are OCT-4 + , CD34 − , SSEA3 − and SSEA4 − .

4. The method of claim 3 , wherein said placental stem cells are additionally SH3 + or SH4 + .

5. The method of claim 3 , wherein said placental stem cells are additionally SH3 + and SH4 + .

6. The method of claim 3 , wherein said placental stem cells are additionally CD29 + , CD45 − , CD90 + , SH2 + , SH3 + , SH4 + , or MHC Class II − .

7. The method of claim 3 , wherein said placental stem cells have at least one of the following additional characteristics: CD10 + , CD29 + , CD44 + , CD45 − , CD54 + , CD90 + , SSEA3 − , or SSEA4 − .

8. The method of claim 3 , wherein said placental stem cells have at least the following additional characteristics: CD10 + , CD29 + , CD44 + , CD45 − , CD54 + , CD90 + , SSEA3 − , and SSEA4 − .

9. The method of claim 3 , wherein said placental stem cells have at least one of the following additional characteristics: CD10 + , CD29 + , CD44 + , CD45 − , CD54 + , or CD90 + .

10. The method of claim 3 , wherein said placental stem cells have at least the following additional characteristics: CD10 + , CD29 + , CD44 + , CD45 − , CD54 + , and CD90 + .
