Abstract

A particle analyzer in which tagged particles to be analyzed are drawn through a suspended capillary tube where a predetermined volume in the capillary tube is illuminated. The illumination scattered by said particles is detected by a detector to count all particles. The fluorescent illumination emitted by tagged particles is detected and the output signals from the fluorescent detectors and scatter detector are processed to provide an analysis of the particles.

Claims (21)

1. A method of detecting particles in a sample fluid, comprising the steps of:

providing a capillary tube having a first end portion, a second opposite end portion, and a predetermined internal cross-section area, said first end portion being adapted to be suspended for immersion into a sample liquid containing particles to be analyzed;

directing a substantially rectangular beam of light onto the capillary tube to form an analyzing region defined by the internal wall of the capillary tube and a thickness of the substantially rectangular beam;

drawing a volume of the sample fluid through the first end portion of the capillary tube and through the analyzing region using a fluidic system comprising a pump, wherein the pump is configured to draw a known volume of the sample fluid into the capillary tube; and

detecting fluorescent light emitted by the particles in response to the illumination of the light beam;

wherein in drawing the volume of the sample fluid, a holder containing the sample fluid moves relative to the capillary to allow the first end portion of the capillary to immerse into the sample fluid.

2. The method of claim 1 wherein the sample fluid is drawn through the first end portion of the capillary tube at a substantially constant rate.

3. The method of claim 1 further comprising the step of detecting light scattered by the particles as they travel through said analyzing region.

4. The method of claim 1 further comprising the step of identifying the particles and their concentration in the sample fluid.

5. A method of detecting particles in a sample fluid, comprising the steps of:

providing a capillary tube having a first end portion, a second opposite end portion, and a predetermined internal cross-section area, said first end portion being adapted to be suspended for immersion into a sample liquid containing particles to be analyzed;

directing a substantially rectangular beam of light onto the capillary tube to form an analyzing region defined by the internal wall of the capillary tube and a thickness of the substantially rectangular beam;

drawing a volume of the sample fluid through the first end portion of the capillary tube and through the analyzing region using a fluidic system comprising a pump, wherein the pump is configured to draw a known volume of the sample fluid into the capillary tube; and

detecting fluorescent light emitted by the particles in response to the illumination of the light beam;

wherein said capillary tube comprises a substantially straight section, said substantially straight section including the first end portion adapted to be suspended for immersion into a sample fluid, and the analyzing region is defined in the substantially straight section.

6. The method of claim 5 wherein said substantially straight section of the capillary tube is disposed substantially vertical with respect to the ground.

7. The method of claim 6 wherein the volume of the sample fluid is drawn by moving a holder containing the sample fluid substantially vertically relative to the capillary to immerse the first end portion into the sample fluid.

8. The method of claim 1 wherein the directing step comprises directing a sheet-like rectangular beam through the capillary.

9. The method of claim 8 wherein said rectangular beam has a thickness of about 20 μm and a width of about 400 μm.

10. The method of claim 1 wherein the step of drawing a volume of the sample fluid comprising using a syringe pump to draw a known volume of the sample fluid into the capillary tube.

11. The method of claim 5 wherein the step of drawing a volume of the sample fluid comprising using a syringe pump to draw a known volume of the sample fluid into the capillary tube.
