Imidazo[1,2-a] pyridinyl bisphosphonates
View Patent ↗Novel imidazo[1,2-α]pyridinyl bisphosphonate compounds are disclosed, as well as methods of preparing the compounds, pharmaceutical compositions including the compounds, and administration of the compounds in methods of treating abnormal calcium and phosphate metabolism, including bone and joint diseases and other disorders.
1. A compound, or a pharmaceutically acceptable salt thereof, wherein the compound has a structure according to Formula I
wherein:
R 1 is hydrogen, hydroxyl, lower alkyl, methoxy, or F;
R 2 is hydrogen, hydroxyl, lower alkyl, or F; and
R 3 is F, CI, or hydrogen.
2. The compound of claim 1 , wherein R 1 and R 2 are each independently hydrogen, hydroxyl, methyl, ethyl, t-butyl, or F.
3. The compound of claim 1 , wherein R 1 is hydrogen or a lower alkyl, and R 2 is hydrogen, hydroxyl, methyl, or F.
4. The compound of claim 1 , wherein R 1 is hydrogen, ethyl, or t-butyl, and R 2 is hydrogen, hydroxyl, methyl, or F.
5. The compound of claim 1 , wherein R 3 is hydrogen or F.
6. The compound of claim 1 , wherein R 1 is hydrogen, R 2 is hydrogen or methyl, and R 3 is hydrogen or F.
7. The compound of claim 1 , wherein R 1 is ethyl or t-butyl, R 2 is hydrogen, and R 3 is hydrogen or F.
8. The compound of claim 1 , wherein the compound is selected from the group consisting of: 1-fluoro-2-(imidazo[1,2-α]pyridin-3-yl-ethyl-bisphosphonic acid; 2-(imidazo[1,2-α]pyridin-3-yl)-ethyl-bisphosphonic acid; 2-(6-methylimidazo[1,2-α]pyridin-3-yl) -ethyl-bisphosphonic acid; [(2-ethyl-imidazo[1,2-α]pyridin-3-yl)-methyl]-bisphosphonic acid; 2-(2-t-butylimidazo[1,2-α]pyridin-3-yl)ethane-1,1- bisphosphonic acid; 2-(7-methylimidazo[1,2-α]pyridin-3-yl)ethyl-bisphosphonic acid; [2-(8-hydroxy-imidazo [1,2-α]pyridin-3-yl)-1-ethane]-bisphosphonic acid; 2-(6-fluoroimidazo[1,2-α]pyridin-3-yl)-ethyl-bisphosphonic acid; and1-fluoro-2-(7-methylimidazo[1,2-α]pyridin-3-yl)-ethyl-bisphosphonic acid, or a pharmaceutically acceptable salt thereof.
9. The compound of claim 1 , wherein the compound is 1-fluoro-2-(imidazo[1,2-α]pyridin-3-yl)-ethyl-bisphosphonic acid.
10. A pharmaceutical composition comprising:
a compound, or a pharmaceutically acceptable salt thereof, wherein the compound has a structure according to Formula I
wherein:
R 1 is hydrogen, hydroxyl, lower alkyl, methoxy, or F;
R 2 is hydrogen, hydroxyl, lower alkyl, or F; and
R 3 is F, CI, or hydrogen; and
a pharmaceutically acceptable carrier.
11. The composition of claim 10 , wherein R 1 and R 2 are each independently hydrogen, hydroxyl, methyl, ethyl, t-butyl, or F.
12. The composition of claim 10 , wherein R 3 is hydrogen or F.
13. The composition of claim 10 , wherein R 1 is hydrogen; R 2 is hydrogen or methyl; and R 3 is hydrogen or F.
14. The composition of claim 10 , wherein R 1 is ethyl or t-butyl; R 2 is hydrogen; and R 3 is hydrogen or F.

The composition of claim 10 , wherein the compound is selected from the group consisting of: 1-fluoro-2-(imidazo[1,2-α]pyridin-3-yl-ethyl-bisphosphonic acid; 2-(imidazo[1,2-α]pyridin-3-yl)-ethyl-bisphosphonic acid; 2-(6-methylimidazo[1,2-α]pyridin-3-yl) -ethyl-bisphosphonic acid; [(2-ethyl-imidazo[1,2-α]pyridin-3-yl)-methyl]-bisphosphonic acid; 2-(2-t-butylimidazo[1,2-α]pyridin-3-yl)ethane-1,1- bisphosphonic acid; 2-(7-methylimidazo[1,2-α]pyridin-3-yl)ethyl-bisphosphonic acid; [2-(8-hydroxy-imidazo [1,2-α]pyridin-3-yl)-1-ethane]-bisphosphonic acid; 2-(6-fluoroimidazo[1,2-α]pyridin-3-yl)-ethyl-bisphosphonic acid; and1-fluoro-2-(7-methylimidazo[1,2-α]pyridin-3-yl)-ethyl-bisphosphonic acid, or a pharmaceutically acceptable salt thereof.
16. The composition of claim 10 , wherein the compound comprises 1-fluoro-2-(imidazo[1,2-α]pyridin-3-yl)ethane-1,1-diyldiphosphonic acid, or a pharmaceutically acceptable salt thereof, and a pharmaceutically acceptable carrier.
17. The composition of claim 10 , wherein the compound further comprises at least one pharmaceutically active ingredient other than a bisphosphonic acid, or pharmaceutically acceptable salt thereof.
18. The composition of claim 17 , wherein the at least one pharmaceutically active ingredient is selected from the group consisting of: an anti-inflammatory, an immunomodulator, a chelator, a musculoskeletal anabolic agent, and a combination thereof.
