Abstract

Novel 5-azaindole bisphosphonate compounds are disclosed, as well as methods of preparing the compounds, pharmaceutical compositions including the compounds, and administration of the compounds in methods of treating abnormal calcium and phosphate metabolism, including bone and joint diseases and other disorders.

Claims (28)

1. A compound, or a pharmaceutically acceptable salt thereof, wherein the compound has a structure according to Formula I

wherein:

R 1 is hydrogen, hydroxyl, lower alkyl, methoxy, or F;

R 2 is hydrogen, hydroxyl, lower alkyl, or F; and

R 3 is hydrogen or a halogen.

2. The compound of claim 1 , wherein R 1 and R 2 are each independently hydrogen, hydroxyl, methyl, ethyl, t-butyl, or F.

3. The compound of claim 1 , wherein R 1 is hydrogen or a lower alkyl, and R 2 is hydrogen, hydroxyl, methyl, or F.

4. The compound of claim 1 , wherein R 1 is hydrogen, ethyl, or t-butyl, and R 2 is hydrogen, hydroxyl, methyl, or F.

5. The compound of claim 1 , wherein R 3 is hydrogen or F.

6. The compound of claim 1 , wherein R 1 is hydrogen, R 2 is hydrogen or methyl, and R 3 is hydrogen or F.

7. The compound of claim 1 , wherein R 1 is ethyl or t-butyl, R 2 is hydrogen, and R 3 is hydrogen or F.

8. The compound of claim 1 , wherein the compound has a structure according to Formula Ia:

wherein R 3 is hydrogen and the compound is 5-azaindol-3-yl-ethyl-bisphosphonic acid, or R 3 is F and the compound is 1-fluoro-(5-azaindol)-3-yl-ethyl-bisphosphonic acid.

9. A pharmaceutical composition comprising:

a compound, or a pharmaceutically acceptable salt thereof, wherein the compound has a structure according to Formula I

wherein:

R 1 is hydrogen, hydroxyl, lower alkyl, methoxy, or F;

R 2 is hydrogen, hydroxyl, lower alkyl, or F; and

R 3 is F, Cl, or hydrogen; and

a pharmaceutically acceptable carrier.

10. The composition of claim 9 , wherein R 1 and R 2 are each independently hydrogen, hydroxyl, methyl, ethyl, t-butyl, or F.

11. The composition of claim 9 , wherein R 3 is hydrogen or F.

12. The composition of claim 9 , wherein R 1 is hydrogen; R 2 is hydrogen or methyl; and R 3 is hydrogen or F.

13. The composition of claim 9 , wherein R 1 is ethyl or t-butyl; R 2 is hydrogen; and R 3 is hydrogen or F.

14. The composition of claim 9 , wherein the compound has a structure according to Formula Ia:

wherein R 3 is hydrogen and the compound is 5-azaindol-3-yl-ethyl-bisphosphonic acid, or R 3 is F and the compound is 1-fluoro-(5-azaindol)-3-yl-ethyl-bisphosphonic acid.

15. The composition of claim 9 , wherein the compound further comprises at least one pharmaceutically active ingredient other than a bisphosphonic acid, or pharmaceutically acceptable salt thereof.

16. The composition of claim 15 , wherein the at least one pharmaceutically active ingredient is selected from the group consisting of: an anti-inflammatory, an immunomodulator, a chelator, a musculoskeletal anabolic agent, and a combination thereof.
