Abstract

Methods for the treatment of CD30-expressing cancers are provided. The methods comprise administering to a subject in need thereof a weekly dose of from about 0.8 mg/kg to about 1.8 mg/kg of an antibody-drug conjugate compound having formula (I); or a pharmaceutically acceptable salt thereof; wherein: mAb is an anti-CD30 antibody unit, S is a sulfur atom of the antibody, A- is a Stretcher unit, and p is from about 3 to about 5.

Claims (19)

1. A method for treating a CD30-expressing hematologic cancer in a human subject, the method comprising administering to a subject in need thereof a weekly dose of a pharmaceutical composition comprising cAC10-MC-vc-PAB-MMAE

wherein the administered dose of cAC10-MC-vc-PAB-MMAE is from 0.8 mg/kg of the subject's body weight to 1.2 mg/kg of the subject's body weight and the pharmaceutical composition is administered for at least three weeks and as a monotherapy.

2. The method of claim 1 wherein the CD30-expressing hematologic cancer is Hodgkin Lymphoma.

3. The method of claim 1 wherein the CD30-expressing hematologic cancer is anaplastic large cell lymphoma.

4. The method of claim 1 wherein the weekly dose is from 0.8 mg/kg to 1.0 mg/kg.

5. The method of claim 1 wherein the weekly dose is 0.8 mg/kg.

6. The method of claim 1 wherein the weekly dose is 1 mg/kg.

7. The method of claim I wherein the weekly dose is 1.2 mg/kg.

8. The method of any one of claims 1 to 7 wherein the weekly dose is a single weekly dose.

9. The method of claim 1 wherein the pharmaceutical composition is administered for at least two 21 day treatment cycles with a one week resting period between each treatment cycle.

10. The method of claim 1 wherein the pharmaceutical composition is administered consecutively, 3 out of 4 weeks, for at least one 28 day treatment cycle.

11. The method of claim 1 wherein the pharmaceutical composition is administered consecutively, 3 out of 4 weeks, for at least two 28 day treatment cycles.

12. The method of claim 11 further comprising administering the pharmaceutical composition of claim 1 as a maintenance therapy following the at least two 28 day treatment cycles.

13. The method of claim 12 wherein the maintenance therapy commences following an evaluation indicating that the subject has no detectable cancer.

14. The method of claim 12 wherein the maintenance therapy comprises administering the pharmaceutical composition of claim 1 once every three to six weeks.

15. The method of claim 12 wherein the administered dose of cAC10-MC-vc-PAB-MMAE for the maintenance therapy is from 1 mg/kg body weight of 1.8 mg/kg body weight.

16. The method of claim 1 wherein said subject has a relapsed or refractory CD30-expressing hematologic cancer.

17. A method for treating a CD30-expressing hematologic cancer in a subject, the method consisting essentially of administering to a human subject in need thereof a weekly dose of a pharmaceutical composition comprising cAC10-MC-vc-PAB-MMAE or a pharmaceutically acceptable salt thereof; and a pharmaceutically acceptable carrier;

wherein the administered dose of cAC10-MC-vc-PAB-MMAE is from 0.8 mg/kg of the subject's body weight to 1.2 mg/kg of the subject's body weight and the pharmaceutical composition is administered for at least three weeks and as a monotherapy.
