Abstract

Influenza vaccines include hemagglutinin from at least one influenza A virus strain and at least one influenza B virus strain. They also include an oil-in-water emulsion adjuvant with submicron oil droplets, comprising squalene. In some embodiments the hemagglutinin concentration is >12 μg/ml per strain. In some embodiments the squalene concentration is <19 mg/ml. In some embodiments the vaccine is mercury-free. In some embodiments the vaccine has a unit dose volume between 0.2-0.3 mL. In some embodiments the squalene concentration is 9.75 mg/mL or 4.88 mg/mL. In some embodiments the vaccine includes antigens from two influenza A virus strains and two influenza B virus strains.

Claims (21)

1. A method of raising an immune response in a human subject, the method comprising administering a vaccine to the human subject who is ≦5 years old and characterized in that the administration of the vaccine induces a protective immune response in the subject against at least one influenza B strain, wherein the vaccine comprises: (i) a concentration of hemagglutinin in the vaccine of at least 12 μg/mL per strain from at least two influenza A virus strains, a B/Victoria/2/87-like influenza B virus strain, and a B/Yamagata/16/88-like influenza B virus strain; and (ii) an oil-in-water emulsion adjuvant with submicron oil droplets, wherein the oil-in-water emulsion adjuvant comprises squalene and the squalene concentration in the vaccine is <19 mg/ml.

2. The method of claim 1 , wherein the hemagglutinins are in the form of split virions, or purified surface antigens.

3. The method of claim 1 , wherein the at least two influenza A virus strains include (i) a H1N1 influenza A virus strain; and (ii) a H3N2 influenza A virus strain.

4. The method of claim 1 , wherein the concentration of hemagglutinin per strain in the vaccine is at least 25 μg/ml.

5. The method of claim 1 , wherein the concentration of squalene in the vaccine is <10 mg/ml.

6. The method of claim 1 , wherein the weight ratio of squalene to hemagglutinin in the vaccine is in the range from 20 to 180.

7. The method of claim 1 , wherein the vaccine has a unit dose volume of 0.5 mL.

The method of claim 1 , wherein the vaccine is a tetravalent inactivated influenza vaccine with a A/H1N1 strain, a A/H3N2 strain, the B/Yamagata/16/88-like influenza B virus strain, and the B/Victoria/2/87-like influenza B virus strain wherein the concentration of hemagglutinin in the vaccine is about 30 μg/ml per strain, the concentration of squalene in the vaccine is about 9.75 mg/ml, and the vaccine has a unit dosage volume of about 0.5 ml.

The method of claim 1 , wherein the vaccine is a tetravalent inactivated influenza vaccine with a A/H1N1 strain, a A/H3N2 strain, the B/Yamagata/16/88-like influenza B virus strain, and the B/Victoria/2/87-like influenza B virus strain, wherein the concentration of hemagglutinin in the vaccine is about 30 μg/ml per strain, the concentration of squalene in the vaccine is about 9.75 mg/ml, and the vaccine a unit dosage volume of about 0.25 ml.

The method of claim 1 , wherein the vaccine is a tetravalent inactivated influenza vaccine with a A/H1N1 strain, a A/H3N2 strain, the B/Yamagata/16/88-like influenza B virus strain, and the B/Victoria/2/87-like influenza B virus strain, wherein the concentration of hemagglutinin is about 30 μg/ml per strain, the concentration of squalene in the vaccine is about 4.88 mg/ml, and the vaccine has a unit dosage volume of about 0.5 ml.

The method of claim 1 , wherein the vaccine is a tetravalent inactivated influenza vaccine with a A/H1N1 strain, a A/H3N2 strain, the B/Yamagata/16/88-like influenza B virus strain, and the B/Victoria/2/87-like influenza B virus strain, wherein the concentration of hemagglutinin in the vaccine is about 30 μg/ml per strain, the concentration of squalene in the vaccine is about 4.88 mg/ml, and the vaccine has a unit dosage volume of about 0.25 ml.

12. The method of claim 1 , wherein the vaccine has a unit dose volume of between 0.2 and 0.3 mL.

13. The method of claim 12 , wherein the vaccine has a unit dose volume of about 0.25 mL.

14. The method of claim 12 or 13 , wherein the concentration of squalene in the vaccine is in the range of 2.5 mg/ml to 19 mg/ml.

15. The method of claim 12 or 13 , wherein the concentration of squalene is in the range of 2.5 mg/m1 to 10 mg/ml.

16. The method of claim 13 , wherein the concentration of squalene in the vaccine is 9.75 mg/mL or 4.88 mg/mL.

17. The method of claim 13 , wherein the amount of squalene per unit dose of the vaccine is 2.7 mg, 2.4 mg, or 1.2 mg.

18. The method of claim 13 , wherein the concentration of hemagglutinin per strain in the vaccine is about 30 μg/ml.

19. The method of claim 13 wherein the amount of hemagglutinin per strain is about 7.5 μg.

20. The method of claim 1 wherein the influenza virus strains have not been passaged through eggs.

21. The method of claim 1 wherein the submicron oil in water emulsion adjuvant includes polysorbate 80 and/or αtocopherol.
