Abstract

Methods for treating a subject suffering from a compromised endogenous hematopoietic system are described that comprise administering to the subject a therapeutically effective amount of adherent stromal cells. Methods of preparing adherent stromal cells and pharmaceutical compositions comprising the cells are also described.

Claims (51)

1 . A method for treating a subject suffering from a compromised endogenous hematopoietic system, comprising administering to the subject a therapeutically effective amount of adherent stromal cells for inducing repopulation of endogenous hematopoietic cells in the endogenous hematopoietic system.

2 . The method of claim 1 , wherein the endogenous hematopoietic cells were produced by the subject's hematopoietic system.

3 . The method of claim 1 , wherein repopulation of endogenous hematopoietic cells comprises increasing the number of hematopoietic cells in the endogenous hematopoietic system of the subject.

4 . The method of claim 3 , wherein repopulation of endogenous hematopoietic cells comprises increasing the number of hematopoietic cells expressing the CD45 marker.

5 . The method of claim 1 , wherein the subject has been exposed to radiation.

6 . The method of claim 1 , wherein the subject is immune deficient due to chemotherapy.

7 . The method of claim 1 , wherein the origin of the adherent stromal cells is placenta or adipose tissue.

8 . The method of claim 1 wherein the adherent stromal cells were cultured under three dimensional culturing conditions supporting cell expansion.

9 . The method of claim 8 , wherein the cultured adherent stromal cells secrete Flt-3 ligand, IL-6, and SCF into the culture medium.

10 . The method of claim 1 , wherein the origin of the adherent stromal cells is placenta or adipose tissue, and the adherent stromal cells were cultured under three dimensional culturing conditions supporting cell expansion.

11 . The method of claim 1 wherein the subject has been treated with chemotherapy.

12 . A pharmaceutical composition comprising a therapeutically effective amount of adherent stromal cells for inducing repopulation of endogenous hematopoietic cells in the endogenous hematopoietic system in a subject suffering from a compromised hematopoietic system.

13 . The pharmaceutical composition of claim 12 , wherein the endogenous hematopoietic cells were produced by the subject's hematopoietic system.

14 . The pharmaceutical composition of claim 12 , wherein repopulation of endogenous hematopoietic cells in the endogenous hematopoietic system comprises increasing the number of endogenous hematopoietic cells in the hematopoietic system of the subject.

15 . The pharmaceutical composition of claim 12 , wherein repopulation of hematopoietic cells in the endogenous hematopoietic system comprises increasing the number of hematopoietic cells expressing the CD45 marker.

16 . The pharmaceutical composition of claim 12 , wherein the subject has been exposed to radiation.

17 . The pharmaceutical composition of claim 12 , wherein the subject is immune deficient due to chemotherapy.

18 . The pharmaceutical composition of claim 12 , wherein the origin of the adherent stromal cells is placenta or adipose tissue.

19 . The pharmaceutical composition of claim 12 wherein the adherent stromal cells were cultured under three dimensional culturing conditions supporting cell expansion.

20 . The pharmaceutical composition of claim 19 , wherein the cultured adherent stromal cells secrete Flt-3 ligand, IL-6, and SCF into the culture medium.

21 . The pharmaceutical composition of claim 12 , wherein the origin of the adherent stromal cells is placenta or adipose tissue, and the adherent stromal cells were cultured under three dimensional culturing conditions supporting cell expansion.

22 . The pharmaceutical composition of claim 12 wherein the subject has been treated with chemotherapy.

23 . A method of treating a subject suffering from a compromised endogenous hematopoietic system due to exposure to radiation or chemotherapy, comprising:

administering to the subject a first therapeutically effective amount of adherent stromal cells within a specified period after the exposure to radiation or chemotherapy, for inducing repopulation of endogenous hematopoietic cells in the endogenous hematopoietic system, and

administering to the subject a second therapeutically effective amount of adherent stromal cells together with exogenous hematopoietic stem cells after a matching period following the exposure, for further enhancing the repopulation of endogenous hematopoietic cells in the endogenous hematopoietic system.

24 . The method of claim 23 , further comprising matching the exogenous hematopoietic stem cells to the subject.

25 . The method of claim 23 , wherein the specified period is within 1-10 days.

26 . The method of claim 25 , wherein the specified period is within 7-10 days.

27 . The method of claim 25 , wherein the specified period is 1-2 days.

28 . The method of claim 23 , wherein the matching period is between 4 and 21 days.

29 . The method of claim 23 , wherein the administration of the first therapeutically effective amount is carried out by at least one of: intravascular, intramuscular, intraperitoneal, and inhalation.

30 . The method of claim 23 , wherein the administration of the second therapeutically effective amount is carried out by at least one of: intravascular, intramuscular, intraperitoneal, and inhalation.

31 . The method of claim 23 , wherein the endogenous hematopoietic cells were produced by the subject's hematopoietic system.

32 . The method of claim 23 , wherein repopulation of endogenous hematopoietic cells comprises increasing the number of hematopoietic cells in the endogenous hematopoietic system of the subject.

33 . The method of claim 32 , wherein repopulation of endogenous hematopoietic cells comprises increasing the number of hematopoietic cells expressing the CD45 marker.

34 . The method of claim 23 , wherein the origin of the adherent stromal cells is placenta or adipose tissue.

35 . The method of claim 23 , wherein the adherent stromal cells were cultured under three dimensional culturing conditions supporting cell expansion.

36 . The method of claim 35 , wherein the cultured adherent stromal cells secrete Flt-3 ligand, IL-6, and SCF into the culture medium.

37 . The method of claim 23 , wherein the origin of the adherent stromal cells is placenta or adipose tissue, and the adherent stromal cells were cultured under three dimensional culturing conditions supporting cell expansion.

38 . The method of claim 23 , wherein the exogenous hematopoietic stem cells are matched allogeneic cord blood or bone marrow cells.

39 . A kit for treating a subject suffering from a compromised endogenous hematopoietic system due to exposure to radiation or chemotherapy, comprising:

a first therapeutically effective amount of adherent stromal cells in a first sterile package, for administration within a specified period after the exposure to radiation or chemotherapy, for inducing repopulation of endogenous hematopoietic cells in the endogenous hematopoietic system,

a second therapeutically effective amount of adherent stromal cells in a second sterile package, for administration after a matching period following the exposure, for further enhancement of repopulation of endogenous hematopoietic cells in the endogenous hematopoietic system, and

instructions for administration of the first and second therapeutically effective amounts.

40 . The kit of claim 39 , wherein the second therapeutically effective amount of adherent stromal cells is packaged together with exogenous hematopoietic stem cells in the second sterile package.

41 . The kit of claim 39 , wherein the first and the second sterile packages are configured to enable administration by at least one of: intravascular, intramuscular, intraperitoneal, and inhalation.

42 . The kit of claim 40 , wherein the exogenous hematopoietic stem cells are matched allogeneic cord blood or bone marrow cells.

43 . A method of treating a subject suffering from a compromised endogenous hematopoietic system due to exposure to radiation or chemotherapy, comprising:

administering to the subject a first therapeutically effective amount of adherent stromal cells within a specified period after the exposure to radiation or chemotherapy, for inducing repopulation of endogenous hematopoietic cells in the endogenous hematopoietic system, and

administering to the subject at least one additional therapeutically effective amount of adherent stromal cells to further induce repopulation of endogenous hematopoietic cells in the endogenous hematopoietic system.

44 . The method of claim 43 , wherein the origin of the each of the therapeutically effective amounts of adherent stromal cells is placenta or adipose tissue, and the adherent stromal cells were cultured under three dimensional culturing conditions supporting cell expansion.
