Abstract

The invention is directed to compositions and methods for selectively reducing the expression of a gene product from a desired target gene in a cell, as well as for treating diseases caused by the expression of the gene. More particularly, the invention is directed to compositions that contain double stranded RNA (“dsRNA”), and methods for preparing them, that are capable of reducing the expression of target genes in eukaryotic cells. The dsRNA has a first oligonucleotide sequence that is between 25 and about 30 nucleotides in length and a second oligonucleotide sequence that anneals to the first sequence under biological conditions. In addition, a region of one of the sequences of the dsRNA having a sequence length of at least 19 nucleotides is sufficiently complementary to a nucleotide sequence of the RNA produced from the target gene to trigger the destruction of the target RNA by the RNAi machinery.

Claims (120)

1. A nucleic acid molecule capable of reducing expression of a target gene following cleavage by human Dicer enzyme comprising:

a first oligonucleotide strand 25-30 nucleotides in length comprising RNA, a 3′ terminus and a 5′ terminus, and

a second oligonucleotide strand at least 26 nucleotides in length comprising RNA, a 3′ terminus, a 5′ terminus and a nucleotide sequence that is complementary to a sequence of a target mRNA of said target gene,

wherein said second strand is annealed to said first strand under biological conditions to form a duplex region of at least 25 nucleotides in length,

wherein the 5′ terminus of said second strand forms a blunt end with the 3′ terminus of said first strand,

wherein said second strand comprises a 1-4 nucleotide 3′-overhang with respect to the 5′ terminus of said first strand, and

wherein said nucleic acid molecule is cleaved by human Dicer enzyme to facilitate incorporation of a cleavage product of said second oligonucleotide strand into RISC.

2. The nucleic acid molecule of claim 1 , wherein said second oligonucleotide strand is complementary to said target mRNA along at least 19 nucleotides of said second oligonucleotide strand length.

The nucleic acid molecule of claim 1 , wherein said second oligonucleotide strand is sufficiently complementary to a target mRNA of said target gene along at least 19 nucleotides of said second oligonucleotide strand length to reduce target gene expression when said nucleic acid molecule is introduced into a mammalian cell.

4. The nucleic acid molecule of claim 1 , wherein said first strand has a length which is at least 26 nucleotides.

5. The nucleic acid molecule of claim 1 , wherein said nucleic acid molecule comprises a duplex region of at least 26 nucleotides in length.

6. The nucleic acid molecule of claim 1 , wherein said second strand is two nucleotides longer at its 3′ terminus than said first strand.

7. The nucleic acid molecule of claim 1 , wherein said first strand of said nucleic acid molecule is 25 nucleotides in length and said second strand is 27 nucleotides in length.

8. The nucleic acid molecule of claim 1 , wherein said blunt end formed by said 3′ terminus of said first strand and said 5′ terminus of said second strand is a base-paired blunt end.

9. The nucleic acid molecule of claim 1 , wherein said second strand is fully complementary to the target mRNA.

10. The nucleic acid molecule of claim 1 , wherein said 5′ terminus of each of said first and said second strands comprises a 5′ phosphate.

The nucleic acid molecule of claim 1 , wherein the relative length in nucleotide residues of said second and first strands is selected from the group consisting of: second strand 26-29 nucleotide residues in length and said first strand 25 nucleotide residues in length, second strand 27-30 nucleotide residues in length and said first strand 26 nucleotide residues in length, second strand 28-30 nucleotide residues in length and first strand 27 nucleotide residues in length, second strand 29-30 nucleotide residues in length and first strand 28 nucleotide residues in length, and second strand 30 nucleotide residues in length and first strand 29 nucleotide residues in length.

The nucleic acid molecule of claim 1 comprising a modified nucleotide selected from the group consisting of a deoxyribonucleotide, a 4-thiouracil, a 5-bromouracil, a 5-iodouracil, a 5-(3-aminoallyl)-uracil, a 2′-O-alkyl ribonucleotide, a 2′-O-methyl ribonucleotide, a 2′-amino ribonucleotide, a 2′-fluoro ribonucleotide, and a locked nucleic acid.

13. The nucleic acid molecule of claim 1 comprising a phosphate backbone modification selected from the group consisting of a phosphonate, a phosphorothioate and a phosphotriester.

14. The nucleic acid molecule of claim 1 , wherein said nucleic acid molecule reduces target gene expression when introduced into a mammalian cell in vitro by an amount (expressed by %) selected from the group consisting of at least 10%, at least 50% and at least 80%.

The nucleic acid molecule of claim 1 , wherein said nucleic acid molecule possesses greater potency than an isolated 21mer siRNA directed to the identical nucleotide sequence of said target mRNA of said target gene in reducing target gene expression when suitably formulated at a concentration of 1 nanomolar or less.

The nucleic acid molecule of claim 1 , wherein said nucleic acid molecule does not induce in a mammalian cell at least one response selected from the group consisting of interferon-alpha induction, interferon-beta induction and PKR activation, when suitably formulated at a concentration of 20 nM or less.

17. The nucleic acid molecule of claim 1 , wherein said mammalian cell is a HEK 293 cell.

18. The nucleic acid molecule of claim 1 , wherein said nucleic acid molecule reduces target gene expression by at least 50% in said mammalian cell at least 6, 8 or 10 days post-administration, when suitably formulated at a concentration of 5 nM.

19. A nucleic acid molecule capable of reducing expression of a target gene following cleavage by human Dicer enzyme comprising:

a first oligonucleotide strand 25-29 nucleotides in length comprising RNA, a 3′ terminus and a 5′ terminus, and

a second oligonucleotide strand at most 30 nucleotides in length comprising RNA, a 3′ terminus, a 5′ terminus and a nucleotide sequence that is complementary to a sequence of a target mRNA of said target gene,

wherein said second strand is annealed to said first strand under biological conditions to form a duplex region of at least 25 nucleotides in length,

wherein the 5′ terminus of said second strand forms a blunt end with the 3′ terminus of said first strand,

wherein said second strand comprises a 1-4 nucleotide 3′-overhang with respect to the 5′ terminus of said first strand, and

wherein said nucleic acid molecule is cleaved by human Dicer enzyme to facilitate incorporation of a cleavage product of said second oligonucleotide strand into RISC.

20. The nucleic acid molecule of claim 19 , wherein said second oligonucleotide strand is complementary to said target mRNA along at least 19 nucleotides of said second oligonucleotide strand length.

21. The nucleic acid molecule of claim 19 , wherein said second oligonucleotide strand is sufficiently complementary to a target mRNA of said target gene along at least 19 nucleotides of said second oligonucleotide strand length to reduce target gene expression when said nucleic acid molecule is introduced into a mammalian cell.

22. The nucleic acid molecule of claim 19 , wherein said first strand has a length which is at least 26 nucleotides.

23. The nucleic acid molecule of claim 19 , wherein said nucleic acid molecule comprises a duplex region of at least 26 nucleotides in length.

24. The nucleic acid molecule of claim 19 , wherein said second strand is two nucleotides longer at its 3′ terminus than said first strand.

25. The nucleic acid molecule of claim 19 , wherein said first strand of said nucleic acid molecule is 25 nucleotides in length and said second strand is 27 nucleotides in length.

26. The nucleic acid molecule of claim 19 , wherein said blunt end formed by said 3′ terminus of said first strand and said 5′ terminus of said second strand is a base-paired blunt end.

27. The nucleic acid molecule of claim 19 , wherein said second strand is fully complementary to the target mRNA.

28. The nucleic acid molecule of claim 19 , wherein said 5′ terminus of each of said first and said second strands comprises a 5′ phosphate.

29. The nucleic acid molecule of claim 19 , wherein the relative length in nucleotide residues of said second and first strands is selected from the group consisting of: second strand 26-29 nucleotide residues in length and said first strand 25 nucleotide residues in length, second strand 27-30 nucleotide residues in length and said first strand 26 nucleotide residues in length, second strand 28-30 nucleotide residues in length and first strand 27 nucleotide residues in length, second strand 29-30 nucleotide residues in length and first strand 28 nucleotide residues in length, and second strand 30 nucleotide residues in length and first strand 29 nucleotide residues in length.

30. The nucleic acid molecule of claim 19 comprising a modified nucleotide selected from the group consisting of a deoxyribonucleotide, a 4-thiouracil, a 5-bromouracil, a 5-iodouracil, a 5-(3-aminoallyl)-uracil, a 2′-O-alkyl ribonucleotide, a 2′-O-methyl ribonucleotide, a 2′-amino ribonucleotide, a 2′-fluoro ribonucleotide, and a locked nucleic acid.

31. The nucleic acid molecule of claim 19 comprising a phosphate backbone modification selected from the group consisting of a phosphonate, a phosphorothioate and a phosphotriester.

32. The nucleic acid molecule of claim 19 , wherein said nucleic acid molecule reduces target gene expression when introduced into a mammalian cell in vitro by an amount (expressed by %) selected from the group consisting of at least 10%, at least 50% and at least 80%.

33. The nucleic acid molecule of claim 19 , wherein said nucleic acid molecule possesses greater potency than an isolated 21mer siRNA directed to the identical nucleotide sequence of said target mRNA of said target gene in reducing target gene expression when suitably formulated at a concentration of 1 nanomolar or less.

34. The nucleic acid molecule of claim 19 , wherein said nucleic acid molecule does not induce in a mammalian cell at least one response selected from the group consisting of interferon-alpha induction, interferon-beta induction and PKR activation, when suitably formulated at a concentration of 20 nM or less.

35. The nucleic acid molecule of claim 19 , wherein said mammalian cell is a HEK 293 cell.

36. The nucleic acid molecule of claim 19 , wherein said nucleic acid molecule reduces target gene expression by at least 50% in said mammalian cell at least 6, 8 or 10 days post-administration, when suitably formulated at a concentration of 5 nM.

37. A nucleic acid molecule capable of reducing expression of a target gene following cleavage by human Dicer enzyme comprising:

a first oligonucleotide strand 25 nucleotides in length comprising RNA, a 3′ terminus and a 5′ terminus, and

a second oligonucleotide strand 27 nucleotides in length comprising RNA, a 3′ terminus, a 5′ terminus and a nucleotide sequence that is complementary to a sequence of a target mRNA of said target gene,

wherein said second strand is annealed to said first strand under biological conditions to form a duplex region of 25 nucleotides in length,

wherein the 5′ terminus of said second strand forms a blunt end with the 3′ terminus of said first strand,

wherein said second strand comprises a two nucleotide 3′-overhang with respect to the 5′ terminus of said first strand, and

wherein said nucleic acid molecule is cleaved by human Dicer enzyme to facilitate incorporation of a cleavage product of said second oligonucleotide strand into RISC.

38. The nucleic acid molecule of claim 37 , wherein said second oligonucleotide strand is complementary to said target mRNA along at least 19 nucleotides of said second oligonucleotide strand length.

39. The nucleic acid molecule of claim 37 , wherein said second oligonucleotide strand is sufficiently complementary to a target mRNA of said target gene along at least 19 nucleotides of said second oligonucleotide strand length to reduce target gene expression when said nucleic acid molecule is introduced into a mammalian cell.

40. The nucleic acid molecule of claim 37 , wherein said blunt end formed by said 3′ terminus of said first strand and said 5′ terminus of said second strand is a base-paired blunt end.

41. The nucleic acid molecule of claim 37 , wherein said second strand is fully complementary to the target mRNA.

42. The nucleic acid molecule of claim 37 , wherein said 5′ terminus of each of said first and said second strands comprises a 5′ phosphate.

43. The nucleic acid molecule of claim 37 comprising a modified nucleotide selected from the group consisting of a deoxyribonucleotide, a 4-thiouracil, a 5-bromouracil, a 5-iodouracil, a 5-(3-aminoallyl)-uracil, a 2′-O-alkyl ribonucleotide, a 2′-O-methyl ribonucleotide, a 2′-amino ribonucleotide, a 2′-fluoro ribonucleotide, and a locked nucleic acid.

44. The nucleic acid molecule of claim 37 comprising a phosphate backbone modification selected from the group consisting of a phosphonate, a phosphorothioate and a phosphotriester.

45. The nucleic acid molecule of claim 37 , wherein said nucleic acid molecule reduces target gene expression when introduced into a mammalian cell in vitro by an amount (expressed by %) selected from the group consisting of at least 10%, at least 50% and at least 80%.

46. The nucleic acid molecule of claim 37 , wherein said nucleic acid molecule possesses greater potency than an isolated 21mer siRNA directed to the identical nucleotide sequence of said target mRNA of said target gene in reducing target gene expression when suitably formulated at a concentration of 1 nanomolar or less.

47. The nucleic acid molecule of claim 37 , wherein said nucleic acid molecule does not induce in a mammalian cell at least one response selected from the group consisting of interferon-alpha induction, interferon-beta induction and PKR activation, when suitably formulated at a concentration of 20 nM or less.

48. The nucleic acid molecule of claim 37 , wherein said mammalian cell is a HEK 293 cell.

49. The nucleic acid molecule of claim 37 , wherein said nucleic acid molecule reduces target gene expression by at least 50% in said mammalian cell at least 6, 8 or 10 days post-administration, when suitably formulated at a concentration of 5 nM.

50. A double stranded nucleic acid molecule comprising first and second oligonucleotide strands, each strand comprising RNA and having a 5′ terminus and a 3′ terminus, wherein starting from the first nucleotide (position 1) at the 3′ terminus of the first oligonucleotide strand, position 1, 2 and/or 3 is substituted with a modified nucleotide, wherein said first strand has a length which is at least 25 and at most 30 nucleotides and wherein said second strand is at least one and at most four nucleotides longer at its 3′ terminus than said first strand and forms a blunt end at its 5′ terminus with said 3′ terminus of said first strand, wherein said double-stranded nucleic acid comprises a duplex region of at least 25 nucleotides in length, and wherein said second oligonucleotide strand comprises a sequence complementary to a target mRNA of a target gene and said double stranded nucleic acid molecule reduces target gene expression when introduced into a mammalian cell.

51. The double stranded nucleic acid molecule of claim 50 , wherein said double stranded nucleic acid molecule is cleaved endogenously in a mammalian cell by human Dicer.

52. The double stranded nucleic acid molecule of claim 50 , wherein said double stranded nucleic acid molecule is cleaved by human Dicer in a mammalian cell so as to facilitate incorporation of a cleaved second oligonucleotide strand into RISC.

53. The double stranded nucleic acid molecule of claim 50 , wherein said second oligonucleotide strand is complementary to said target mRNA along at least 19 nucleotides of said second oligonucleotide strand length.

54. The double stranded nucleic acid molecule of claim 50 , wherein said first strand has a length which is at least 26 nucleotides.

55. The double stranded nucleic acid molecule of claim 50 , wherein said double-stranded nucleic acid comprises a duplex region of at least 26 nucleotides in length.

56. The double stranded nucleic acid molecule of claim 50 , wherein said second strand is two nucleotides longer at its 3′ terminus than said first strand.

57. The double stranded nucleic acid molecule of claim 50 , wherein said first strand of said isolated double stranded nucleic acid molecule is 25 nucleotides in length and said second strand is 27 nucleotides in length.

58. The double stranded nucleic acid molecule of claim 50 , wherein said blunt end formed by said 3′ terminus of said first strand and said 5′ terminus of said second strand is a base-paired blunt end.

59. The double stranded nucleic acid molecule of claim 50 , wherein said modified nucleotide residue of said 3′ terminus of said first strand is selected from the group consisting of a deoxyribonucleotide, an acyclonucleotide and a fluorescent molecule.

60. The double stranded nucleic acid molecule of claim 50 , wherein position 1 of said 3′ terminus of the first oligonucleotide strand is a deoxyribonucleotide.

61. The double stranded nucleic acid molecule of claim 50 , wherein positions 1 and 2 of said 3′ terminus of the first oligonucleotide strand are deoxyribonucleotides.

62. The double stranded nucleic acid molecule of claim 50 , wherein said double stranded nucleic acid molecule is cleaved endogenously in a mammalian cell to produce a double-stranded nucleic acid molecule of a length in the range of 19-23 nucleotides that reduces target gene expression.

63. The double stranded nucleic acid molecule of claim 50 , wherein said second strand is fully complementary to the target mRNA.

64. The double stranded nucleic acid molecule of claim 50 , wherein said 5′ terminus of each of said first and said second strands comprises a 5′ phosphate.

65. The double stranded nucleic acid molecule of claim 50 , wherein the relative length in nucleotide residues of said second and first strands is selected from the group consisting of: second strand 26-29 nucleotide residues in length and said first strand 25 nucleotide residues in length, second strand 27-30 nucleotide residues in length and said first strand 26 nucleotide residues in length, second strand 28-30 nucleotide residues in length and first strand 27 nucleotide residues in length, second strand 29-30 nucleotide residues in length and first strand 28 nucleotide residues in length, and second strand 30 nucleotide residues in length and first strand 29 nucleotide residues in length.

66. The double stranded nucleic acid molecule of claim 50 comprising a modified nucleotide selected from the group consisting of a deoxyribonucleotide, a dideoxyribonucleotide, an acyclonucleotide, a 3′-deoxyadenosine (cordycepin), a 3′-azido-3′-deoxythymidine (AZT), a 2′,3′-dideoxyinosine (ddI), a 2′,3′-dideoxy-3′-thiacytidine (3TC), a 2′,3′-didehydro-2′,3′-dideoxythymidine (d4T), a monophosphate nucleotide of 3′-azido-3′-deoxythymidine (AZT), a 2′,3′-dideoxy-3′-thiacytidine (3TC) and a monophosphate nucleotide of 2′,3′-didehydro-2′,3′-dideoxythymidine (d4T), a 4-thiouracil, a 5-bromouracil, a 5-iodouracil, a 5-(3-aminoallyl)-uracil, a 2′-O-alkyl ribonucleotide, a 2′-O-methyl ribonucleotide, a 2′-amino ribonucleotide, a 2′-fluoro ribonucleotide, and a locked nucleic acid.

67. The double stranded nucleic acid molecule of claim 50 comprising a phosphate backbone modification selected from the group consisting of a phosphonate, a phosphorothioate and a phosphotriester.

The double stranded nucleic acid molecule of claim 50 , wherein said double stranded nucleic acid molecule reduces target gene expression when introduced into a mammalian cell in vitro by an amount (expressed by %) selected from the group consisting of at least 10%, at least 50% and at least 80%.

69. The double stranded nucleic acid molecule of claim 50 , wherein said double stranded nucleic acid molecule possesses greater potency than an isolated 21mer siRNA directed to the identical nucleotide sequence of said target mRNA of said target gene in reducing target gene expression when suitably formulated at a concentration of 1 nanomolar or less.

70. The double stranded nucleic acid molecule of claim 50 , wherein said double stranded nucleic acid molecule does not induce in a mammalian cell at least one response selected from the group consisting of interferon-alpha induction, interferon-beta induction and PKR activation, when suitably formulated at a concentration of 20 nM or less.

71. The double stranded nucleic acid molecule of claim 50 , wherein said mammalian cell is a HEK 293 cell.

72. The double stranded nucleic acid molecule of claim 50 , wherein said double stranded nucleic acid reduces target gene expression by at least 50% in said mammalian cell at least 6, 8 or 10 days post-administration, when suitably formulated at a concentration of 5 nM.

73. A double stranded nucleic acid molecule comprising first and second oligonucleotide strands, each strand comprising RNA and having a 5′ terminus and a 3′ terminus, wherein starting from the first nucleotide (position 1) at the 3′ terminus of the first oligonucleotide strand, position 1, 2 and/or 3 is substituted with a modified nucleotide, wherein said first strand has a length which is at least 25 and at most 29 nucleotides and said second strand has a length which is at most 30 nucleotides, wherein said 3′ terminus of said first strand and said 5′ terminus of said second strand form a blunt end and said second strand is at least one and at most four nucleotides longer at its 3′ terminus than said first strand, wherein said double-stranded nucleic acid comprises a duplex region of at least 25 nucleotides in length, and wherein said second oligonucleotide strand comprises a sequence complementary to a target mRNA of a target gene and said double stranded nucleic acid molecule reduces target gene expression when introduced into a mammalian cell.

74. The double stranded nucleic acid molecule of claim 73 , wherein said double stranded nucleic acid molecule is cleaved endogenously in a mammalian cell by human Dicer.

75. The double stranded nucleic acid molecule of claim 73 , wherein said double stranded nucleic acid molecule is cleaved by human Dicer in a mammalian cell so as to facilitate incorporation of a cleaved second oligonucleotide strand into RISC.

76. The double stranded nucleic acid molecule of claim 73 , wherein said modified nucleotide residue of said 3′ terminus of said first strand is selected from the group consisting of a deoxyribonucleotide, an acyclonucleotide and a fluorescent molecule.

77. The double stranded nucleic acid molecule of claim 73 , wherein said 5′ terminus of each of said first and said second strands comprises a 5′ phosphate.

78. The double stranded nucleic acid molecule of claim 73 comprising a phosphate backbone modification selected from the group consisting of a phosphonate, a phosphorothioate and a phosphotriester.

79. The double stranded nucleic acid molecule of claim 73 , wherein said double stranded nucleic acid molecule reduces target gene expression when introduced into a mammalian cell in vitro by an amount (expressed by %) selected from the group consisting of at least 10%, at least 50% and at least 80%.

The double stranded nucleic acid molecule of claim 73 , wherein said double stranded nucleic acid molecule possesses greater potency than an isolated 21mer siRNA directed to the identical nucleotide sequence of said target mRNA of said target gene in reducing target gene expression when suitably formulated at a concentration of 1 nanomolar or less.

81. The double stranded nucleic acid molecule of claim 73 , wherein said double stranded nucleic acid molecule does not induce in a mammalian cell at least one response selected from the group consisting of interferon-alpha induction, interferon-beta induction and PKR activation, when suitably formulated at a concentration of 20 nM or less.

82. The double stranded nucleic acid molecule of claim 73 , wherein said mammalian cell is a HEK 293 cell.

83. The double stranded nucleic acid molecule of claim 73 , wherein said double stranded nucleic acid molecule reduces target gene expression by at least 50% in said mammalian cell at least 6, 8 or 10 days post-administration, when suitably formulated at a concentration of 5 nM.

84. A double stranded nucleic acid molecule comprising first and second oligonucleotide strands, each strand comprising RNA and having a 5′ terminus and a 3′ terminus, wherein said first strand is 25 nucleotide residues in length, and wherein starting from the first nucleotide (position 1) at the 3′ terminus of the first oligonucleotide strand, position 1, 2 and/or 3 is substituted with a modified nucleotide, and said second oligonucleotide strand is 27 nucleotide residues in length and is two nucleotides longer at its 3′ terminus than said first strand, wherein said double-stranded nucleic acid molecule comprises a duplex region of at least 25 nucleotides in length, wherein said second strand comprises a sequence complementary to a target mRNA of a target gene and forms a base-paired blunt end at its 5′ terminus with said 3′ terminus of said first strand, wherein said double stranded nucleic acid molecule reduces target gene expression when introduced into a mammalian cell.

85. The double stranded nucleic acid molecule of claim 84 , wherein said double stranded nucleic acid molecule is cleaved endogenously in a mammalian cell by human Dicer.

86. The double stranded nucleic acid molecule of claim 84 , wherein said double stranded nucleic acid molecule is cleaved by human Dicer in a mammalian cell so as to facilitate incorporation of a cleaved second oligonucleotide strand into RISC.

87. The double stranded nucleic acid molecule of claim 84 , wherein said modified nucleotide residue of said 3′ terminus of said first strand is selected from the group consisting of a deoxyribonucleotide, an acyclonucleotide and a fluorescent molecule.

88. The double stranded nucleic acid molecule of claim 84 , wherein said 5′ terminus of each of said first and said second strands comprises a 5′ phosphate.

89. The double stranded nucleic acid molecule of claim 84 comprising a phosphate backbone modification selected from the group consisting of a phosphonate, a phosphorothioate and a phosphotriester.

90. The double stranded nucleic acid molecule of claim 84 , wherein said double stranded nucleic acid molecule reduces target gene expression when introduced into a mammalian cell in vitro by an amount (expressed by %) selected from the group consisting of at least 10%, at least 50% and at least 80%.

91. The double stranded nucleic acid molecule of claim 84 , wherein said double stranded nucleic acid molecule is cleaved by human Dicer in a mammalian cell so as to facilitate incorporation of a cleaved second oligonucleotide strand into RISC.

92. The double stranded nucleic acid molecule of claim 84 , wherein said second oligonucleotide strand is complementary to said target mRNA along at least 19 nucleotides of said second oligonucleotide strand length.

93. The double stranded nucleic acid molecule of claim 84 , wherein said double stranded nucleic acid molecule possesses greater potency than an isolated 21mer siRNA directed to the identical nucleotide sequence of said target mRNA of said target gene in reducing target gene expression when suitably formulated at a concentration of 1 nanomolar or less.

94. The double stranded nucleic acid molecule of claim 84 , wherein said double stranded nucleic acid molecule does not induce in a mammalian cell at least one response selected from the group consisting of interferon-alpha induction, interferon-beta induction and PKR activation, when suitably formulated at a concentration of 20 nM or less.

95. The double stranded nucleic acid molecule of claim 84 , wherein said mammalian cell is a HEK 293 cell.

96. The double stranded nucleic acid molecule of claim 84 , wherein said double stranded nucleic acid molecule reduces target gene expression by at least 50% in said mammalian cell at least 6, 8 or 10 days post-administration, when suitably formulated at a concentration of 5 nM.

97. The nucleic acid molecule of claim 1 , wherein said nucleic acid molecule is an isolated, synthetic or an isolated, synthetic nucleic acid molecule.

98. The nucleic acid molecule of claim 19 , wherein said nucleic acid molecule is an isolated, synthetic or an isolated, synthetic nucleic acid molecule.

99. The nucleic acid molecule of claim 37 , wherein said nucleic acid molecule is an isolated, synthetic or an isolated, synthetic nucleic acid molecule.

100. The double stranded nucleic acid molecule of claim 50 , wherein said nucleic acid molecule is an isolated, synthetic or an isolated, synthetic nucleic acid molecule.

101. The double stranded nucleic acid molecule of claim 73 , wherein said nucleic acid molecule is an isolated, synthetic or an isolated, synthetic nucleic acid molecule.

102. The double stranded nucleic acid molecule of claim 84 , wherein said nucleic acid molecule is an isolated, synthetic or an isolated, synthetic nucleic acid molecule.
