Abstract

This invention relates to stable non-aqueous single phase viscous vehicles and to formulations utilizing such vehicles. The formulations comprise at least one beneficial agent uniformly suspended in the vehicle. The formulation is capable of being stored at temperatures ranging from cold to body temperature for long periods of time. The formulations are capable of being uniformly delivered from drug delivery systems at an exit shear rate of between about 1 to 1×10 −7 reciprocal second.

Claims (26)

1. A method of making an implantable drug delivery device, comprising:

a) providing a reservoir having a first end and a second end;

b) inserting a piston into the reservoir, wherein the piston separates the reservoir into a first chamber adjacent the first end and a second chamber adjacent the second end;

c) inserting an osmotic engine into the second chamber;

d) inserting a rate controlling membrane at the second end; and

e) filling the first chamber with a beneficial agent formulation that comprises:

stable non-aqueous single phase biocompatible viscous vehicle, comprising a solvent component and a polymer component, wherein (i) the solvent component is a carboxylic acid ester or lauryl alcohol and the polymer component is polyvinylpyrrolidone, and (ii) the viscous vehicle has a viscosity of between about 10,000 to about 250,000 poise, and

a beneficial agent uniformly suspended in the vehicle, wherein the beneficial agent is a peptide or protein.

2. The method of claim 1 , wherein the vehicle further comprises a surfactant component, and wherein the surfactant component is glycerol monolaurate or a polysorbate.

3. The method of claim 2 , wherein the vehicle comprises about 30% (w/w) to about 50% (w/w) of the solvent component, and about 5% (w/w) to about 60% (w/w) of the polymer component.

4. The method of claim 3 , wherein the vehicle further comprises about 5% (w/w) to about 20% (w/w) of a surfactant component.

5. The method of claim 1 , wherein the beneficial agent formulation further comprises an antioxidant.

6. The method of claim 5 , wherein the antioxidant is selected from the group consisting of tocopherol, ascorbic acid, ascorbyl palmitate, butylated hydroxyanisole, butylated hydroxytoluene, and propyl gallate.

7. The method of claim 1 , wherein the beneficial agent formulation comprises at least about 0.1% (w/w) beneficial agent.

8. The method of claim 1 , wherein the beneficial agent formulation comprises at least about 10% (w/w) beneficial agent.

9. The method of claim 1 , wherein the beneficial agent is a protein.

10. The method of claim 9 , wherein the protein is human growth hormone.

11. The method of claim 1 , wherein the beneficial agent has been dried to a low moisture content prior to incorporation in the beneficial agent formulation.

12. The method of claim 11 , wherein the beneficial agent is prepared by spray-drying.

13. The method of claim 1 , further comprising preparing the stable non-aqueous single phase biocompatible viscous vehicle by blending the solvent component and the polymer component under dry conditions at an elevated temperature to form a liquid, and cooling the liquid to room temperature.

The method of claim 13 , further comprising preparing the beneficial agent formulation by combining the stable non-aqueous single phase biocompatible viscous vehicle and the beneficial agent under dry conditions, blending the vehicle and the beneficial agent under vacuum at an elevated temperature to uniformly disperse the beneficial agent in the vehicle, and allowing the beneficial agent formulation to cool to room temperature.

15. The method of claim 1 , further comprising inserting a back-diffusion regulating outlet at the first end.

16. The method of claim 1 , wherein the carboxylic acid ester is lauryllactate.

17. The method of claim 1 , wherein the solvent component is lauryl alcohol.

18. The method of claim 1 , wherein the beneficial agent is prepared by spray-drying.

19. The method of claim 1 , wherein the solvent component and the polymer component are in a weight-percent range of 40:60 to 60:40.
