Abstract

The present invention is directed to modulators of indoleamine 2,3-dioxygenase (IDO), as well as compositions and pharmaceutical methods thereof.

Claims (274)

1. A method of inhibiting immunosuppression or treating cancer, in a cancer patient in need thereof comprising administering to said cancer patient a therapeutically effective amount of a compound of formula:

or a pharmaceutically acceptable salt thereof, wherein:

W, X 1 , and X 2 are independently selected from (CR a R b ) t , (CR a R b ) u O(CR a R b ) v , (CR a R b ) u C(O)(CR a R b ) v , (CR a R b ) u C(O)NR c (CR a R b ), (CR a R b ) u C(O)O(CR a R b ) v , (CR a R b ) u C(S)(CR a R b ) u , (CR a R b ) u C(S)NR c (CR a R b ) v , (CR a R b ) u S(O)(CR a R b ) v , (CR a R b ) u S(O)NR c (CR a R b ) v , (CR a R b ) u S(O) 2 (CR a R b ) v , (CR a R b ) u S(O) 2 NR c (CR a R b ) v , (CR a R b ) u NR c (CR a R b ) v , and (CR a R b ) u C(═NR d )NR c (CR a R b ) v ;

R 2 is H, C 1-6 alkyl, or C 3-7 cycloalkyl;

R 3a and R 5a are independently selected from C 1-8 alkyl, C 2-8 alkenyl, C 2-8 alkynyl, aryl, cycloalkyl, heteroaryl, and heterocycloalkyl, each optionally substituted by 1, 2, 3, 4 or 5 substituents independently selected from halo, C 1-6 alkyl, C 2-6 alkenyl, C 2-6 alkynyl, C 1-4 haloalkyl, Cy 1 , CN, NO 2 , OR e1 , SR e1 , C(O)R f1 , C(O)NR g1 R h1 , C(O)OR e1 , OC(O)R f1 , OC(O)NR g1 R h1 , NR g1 C(O)NR g1 R h1 , NR g1 R h1 , NR g1 C(O)R f1 , NR g1 C(O)OR e1 , P(R f1 ) 2 , P(OR e1 ) 2 , P(O)R e1 R f1 , P(O)OR e1 OR f1 , S(O)R f1 , S(O)NR g1 R h1 , S(O) 2 R f1 , and S(O) 2 NR g1 R b1 ;

R 3b is aryl or heteroaryl, each optionally substituted by 1, 2, 3, 4 or 5 substituents independently selected from halo, C 1-6 alkyl, C 2-6 alkenyl, C 2-6 alkynyl, C 1-4 haloalkyl, Cy 2 , CN, NO 2 , OR e1 , SR e1 , C(O)R f1 , C(O)NR g1 R h1 , C(O)OR e1 , OC(O)R f1 , OC(O)NR g1 R h1 , NR g1 C(O)NR g1 R h1 , NR g1 R h1 , NR g1 C(O)R f1 , NR g1 C(O)OR e1 , P(R f1 ) 2 , P(OR e1 ) 2 , P(O)R e1 R f1 , P(O)OR e1 OR f1 , S(O)R f1 , S(O)NR g1 R h1 , S(O) 2 R f1 , and S(O) 2 NR g1 R h1 ;

R 5b is selected from H, C 1-8 alkyl, C 2-8 alkenyl, C 2-8 alkynyl, aryl, cycloalkyl, heteroaryl, and heterocycloalkyl, wherein said C 1-8 alkyl, C 2-8 alkenyl, C 2-8 alkynyl, aryl, cycloalkyl, heteroaryl, and heterocycloalkyl are each optionally substituted by 1, 2, 3, 4 or 5 substituents independently selected from halo, C 1-6 alkyl, C 2-6 alkenyl, C 2-6 alkynyl, C 1-4 haloalkyl, Cy 2 , CN, NO 2 , OR e1 , SR e1 , C(O)R f1 , C(O)NR g1 R h1 , C(O)OR e1 , OC(O)R f1 , OC(O)NR g1 R h1 , NR g1 C(O)NR g1 R h1 , NR g1 R h1 , NR g1 C(O)R f1 , NR g1 C(O)OR e1 , P(R f1 ) 2 , P(OR e1 ) 2 , P(O)R e1 R f1 , P(O)OR e1 OR f1 , S(O)R f1 , S(O)NR g1 R h1 , S(O) 2 R f1 , and S(O) 2 NR g1 R h1 ;

Cy 1 and Cy 2 are independently selected from aryl, heteroaryl, cycloalkyl, and heterocycloalkyl, each optionally substituted by 1, 2, 3, 4 or 5 substituents independently selected from halo, C 1-4 alkyl, C 2-4 alkenyl, C 2-4 alkynyl, C 1-4 haloalkyl, CN, NO 2 , OR e3 , SR e3 , C(O)R f3 , C(O)NR g3 R h3 , C(O)OR e3 , OC(O)R f3 , OC(O)NR g3 R h3 , NR g3 R h3 , NR g3 C(O)R h3 , NR g3 C(O)OR e3 , P(R f3 ) 2 , P(OR e3 ) 2 , P(O)R e3 R f3 , P(O)OR e3 OR f3 , S(O)R f3 , S(O)NR g3 R h3 , S(O) 2 R f3 , and S(O) 2 NR g3 R h3 ;

R a and R b are independently selected from H, halo, C 1-6 alkyl, C 2-6 alkenyl, C 2-6 alkynyl, C 1-4 haloalkyl, aryl, cycloalkyl, heteroaryl, heterocycloalkyl, arylalkyl, cycloalkylalkyl, heteroarylalkyl, heterocycloalkylalkyl, CN, NO 2 , OR e4 , SR e4 , C(O)R f4 , C(O)NR g4 R h4 , C(O)OR e4 , OC(O)R f4 , OC(O)NR g4 R h4 , NR g4 R h4 , NR g4 C(O)R h4 , NR g4 C(O)OR e4 , P(R f4 ) 2 , P(OR e4 ) 2 , P(O)R e4 R f4 , P(O)OR e4 OR f4 , S(O)R f4 , S(O)NR g4 R h4 , S(O) 2 R f4 , and S(O) 2 NR g4 R h4 ;

R d1 is H, C 1-6 alkyl, C 1-6 haloalkyl, C 2-6 alkenyl, C 2-6 alkynyl, aryl, cycloalkyl, arylalkyl, or cycloalkylalkyl;

R e1 , R e3 , and R e4 are independently selected from H, C 1-6 alkyl, C 1-6 haloalkyl, C 2-6 alkenyl, (C 1-6 alkoxy)-C 1-6 alkyl, C 2-6 alkynyl, aryl, cycloalkyl, heteroaryl, heterocycloalkyl, arylalkyl, cycloalkylalkyl, heteroarylalkyl and heterocycloalkylalkyl;

R f1 , R f3 , and R f4 are independently selected from H, C 1-6 alkyl, C 1-6 haloalkyl, C 2-6 alkenyl, C 2-6 alkynyl, aryl, cycloalkyl, heteroaryl and heterocycloalkyl;

R g1 , R g3 , and R g4 are independently selected from H, C 1-6 alkyl, C 1-6 haloalkyl, C 2-6 alkenyl, C 2-6 alkynyl, aryl, cycloalkyl, arylalkyl, and cycloalkylalkyl;

R h1 , R h3 , and R h4 are independently selected from H, C 1-6 alkyl, C 1-6 haloalkyl, C 2-6 alkenyl, C 2-6 alkynyl, aryl, cycloalkyl, arylalkyl, and cycloalkylalkyl;

or R g1 and R h1 together with the N atom to which they are attached form a 4-, 5-, 6- or 7-membered heterocycloalkyl group;

or R g3 and R h3 together with the N atom to which they are attached form a 4-, 5-, 6- or 7-membered heterocycloalkyl group;

or R g4 and R h4 together with the N atom to which they are attached form a 4-, 5-, 6- or 7-membered heterocycloalkyl group;

a is 0 or 1;

b is 0 or 1;

m is 1;

n is 0 or 1;

p is 0 or 1;

t is, independently, 1, 2, 3, 4, 5 or 6;

u is, independently, 0, 1, 2, 3, 4, 5 or 6; and

v is, independently, 0, 1, 2, 3, 4, 5 or 6.

2. A method of inhibiting activity of indoleamine 2,3-dioxygenase comprising contacting said indoleamine 2,3-dioxygenase with a compound of formula:

or a pharmaceutically acceptable salt thereof, wherein:

W, X 1 , and X 2 are independently selected from (CR a R b ) t , (CR a R b ) u O(CR a R b ) v , (CR a R b ) u C(O)(CR a R b ) v , (CR a R b ) u C(O)NR c (CR a R b ) v , (CR a R b ) u C(O)O(CR a R b ) v , (CR a R b ) u C(S)(CR a R b ) u , (CR a R b ) u C(S)NR c (CR a R b ) v , (CR a R b ) u S(O)(CR a R b ) v , (CR a R b ) u S(O)NR c (CR a R b ) v , (CR a R b ) u S(O) 2 (CR a R b ) v , (CR a R b ) u S(O) 2 NR c (CR a R b ) v , (CR a R b ) u NR c (CR a R b ) v , and (CR a R b ) u C(═NR d )NR c (CR a R b ) v ;

R 2 is H, C 1-6 alkyl, or C 3-7 cycloalkyl;

R 3a and R 5a are independently selected from C 1-8 alkyl, C 2-8 alkenyl, C 2-8 alkynyl, aryl, cycloalkyl, heteroaryl, and heterocycloalkyl, each optionally substituted by 1, 2, 3, 4 or 5 substituents independently selected from halo, C 1-6 alkyl, C 2-6 alkenyl, C 2-6 alkynyl, C 1-4 haloalkyl, Cy 1 , CN, NO 2 , OR e1 , SR e1 , C(O)R f1 , C(O)NR g1 R h1 , C(O)OR e1 , OC(O)R f1 , OC(O)NR g1 R h1 , NR g1 C(O)NR g1 R h1 , NR g1 R h1 , NR g1 C(O)R f1 , NR g1 C(O)OR e1 , P(R f1 ) 2 , P(OR e1 ) 2 , P(O)R e1 R f1 , P(O)OR e1 OR f1 , S(O)R f1 , S(O)NR g1 R h1 , S(O) 2 R f1 , and S(O) 2 NR g1 R h1 ;

R e1 , R e3 , and R e4 , are independently selected from H, C 1-6 alkyl, C 1-6 haloalkyl, C 2-6 alkenyl, C 1-6 alkoxy)-C 1-6 alkyl, C 2-6 alkynyl, aryl, cycloalkyl, heteroaryl, heterocycloalkyl, arylalkyl, cycloalkylalkyl, heteroarylalkyl and heterocycloalkylalkyl;

R f1 , R f3 , and R f4 are independently selected from H, C 1-6 alkyl, C 1-6 haloalkyl, C 2-6 alkenyl, C 2-6 alkynyl, aryl, cycloalkyl, heteroaryl and heterocycloalkyl;

R g1 , R g3 , and R g4 are independently selected from H, C 1-6 alkyl, C 1-6 haloalkyl, C 2-6 alkenyl, C 2-6 alkynyl, aryl, cycloalkyl, arylalkyl, and cycloalkylalkyl;

R h1 , R h3 , and R h4 are independently selected from H, C 1-6 alkyl, C 1-6 haloalkyl, C 2-6 alkenyl, C 2-6 alkynyl, aryl, cycloalkyl, arylalkyl, and cycloalkylalkyl;

or R g1 and R h1 together with the N atom to which they are attached form a 4-, 5-, 6- or 7-membered heterocycloalkyl group;

or R g3 and R h3 together with the N atom to which they are attached form a 4-, 5-, 6- or 7-membered heterocycloalkyl group;

or R g4 and R h4 together with the N atom to which they are attached form a 4-, 5-, 6- or 7-membered heterocycloalkyl group;

a is 0 or 1;

b is 0 or 1;

m is 1;

n is 0 or 1;

p is 0 or 1;

t is, independently, 1, 2, 3, 4, 5 or 6;

u is, independently, 0, 1, 2, 3, 4, 5 or 6; and

v is, independently, 0, 1, 2, 3, 4, 5 or 6.

3. The method of claim 1 , further comprising administration of an anti-cancer vaccine, an anti-viral agent, a chemotherapeutic, an immunosuppressant, radiation, an anti-tumor vaccine, an anti-viral vaccine, cytokine therapy, or a tyrosine kinase inhibitor.

4. The method of claim 3 , wherein said cytokine therapy is IL-2.

5. The method of claim 3 , wherein said chemotherapeutic is a cytotoxic agent.

6. The method of claim 1 , further comprising administration of an anti-CTLA-4 antibody.

7. The method of claim 1 , wherein said method is a method of treating cancer.

8. The method of claim 7 , wherein said cancer is cancer of the colon.

9. The method of claim 7 , wherein said cancer is pancreatic cancer.

10. The method of claim 7 , wherein said cancer is breast cancer.

11. The method of claim 7 , wherein said cancer is prostate cancer.

12. The method of claim 7 , wherein said cancer is lung cancer.

13. The method of claim 7 , wherein said cancer is brain cancer.

14. The method of claim 7 , wherein said cancer is ovarian cancer.

15. The method of claim 7 , wherein said cancer is cervical cancer.

16. The method of claim 7 , wherein said cancer is testicular cancer.

17. The method of claim 7 , wherein said cancer is renal cancer.

18. The method of claim 7 , wherein said cancer is cancer of the head and neck.

19. The method of claim 7 , wherein said cancer is lymphoma.

20. The method of claim 7 , wherein said cancer is leukemia.

21. The method of claim 7 , wherein said cancer is melanoma.

22. The method of claim 1 , wherein R 2 is H.

23. The method of claim 22 wherein n is 0.

24. The method of claim 23 wherein p is 1.

25. The method of claim 24 wherein R 5b is H.

26. The method of claim 25 wherein a is 0.

27. The method of claim 26 wherein b is 0.

28. The method of claim 27 wherein R 3b is phenyl optionally substituted by 1, 2, 3, 4 or 5 substituents independently selected from halo, C 1-6 alkyl, C 2-6 alkenyl, C 2-6 alkynyl, C 1-4 haloalkyl, Cy 2 , CN, NO 2 , OR e1 , SR e1 , C(O)R f1 , C(O)NR g1 R h1 , C(O)OR e1 , OC(O)R f1 , OC(O)NR g1 R h1 , NR g1 C(O)NR g1 R h1 , NR g1 R h1 , NR g1 C(O)R f1 , NR g1 C(O)OR e1 , P(R f1 ) 2 , P(OR e1 ) 2 , P(O)R e1 R f1 , P(O)OR e1 OR f1 , S(O)R f1 , S(O)NR g1 R h1 , S(O) 2 R f1 , and S(O) 2 NR g1 R h1 .

29. The method of claim 26 wherein b is 1.

30. The method of claim 29 wherein W is (CR a R b ) t or (CR a R b ) u O(CR a R b ) v .

The method of claim 30 wherein R 3b is heteroaryl, optionally substituted by 1, 2, 3, 4 or 5 substituents independently selected from halo, C 1-6 alkyl, C 2-6 alkenyl, C 2-6 alkynyl, C 1-4 haloalkyl, Cy 2 , CN, NO 2 , OR e1 , SR e1 , C(O)R f1 , C(O)NR g1 R h1 , C(O)OR e1 , OC(O)R f1 , OC(O)NR g1 R h1 , NR g1 C(O)NR g1 R h1 , NR g1 R h1 , NR g1 C(O)R f1 , NR g1 C(O)OR e1 , P(R f1 ) 2 , P(OR e1 ) 2 , P(O)R e1 R f1 , P(O)OR e1 OR f1 , S(O)R f1 , S(O)NR g1 R h1 , S(O) 2 NR g1 R h1 , S(O) 2 R f1 , and S(O) 2 NR g1 R h1 .

32. The method of claim 1 , wherein said compound is selected from:

4-[(Anilinocarbonyl)amino]-N-(3-chlorophenyl)-N-hydroxy-1,2,5-oxadiazole-3-carboximidamide;

tert-Butyl {4-[({4-[(E,Z)-[(3-chlorophenyl)amino](hydroxyimino)methyl]-1,2,5-oxadiazol-3-yl}amino)carbonyl]benzyl}carbamate;

4-(Aminomethyl)-N-{4-[(E,Z)-[(3-chlorophenyl)amino](hydroxyimino)methyl]-1,2,5-oxadiazol-3-yl}benzamide;

N-(3-Bromo-4-fluorophenyl)-4-[(3-cyanobenzyl)amino]-N′-hydroxy-1,2,5-oxadiazole-3-carboximidamide, or a pharmaceutically acceptable salt of any of the aforementioned.

33. The method of claim 1 , wherein said compound is selected from:

N-{4-[[(3-Chlorophenyl)amino](hydroxyimino)methyl]-1,2,5-oxadiazol-3-yl}-2-phenylacetamide;

N-{-4-[[(3-Chlorophenyl)amino](hydroxyimino)methyl]-1,2,5-oxadiazol-3-yl}benzamide; and

N-{4-[(Benzylamino)(hydroxyimino)methyl]-1,2,5-oxadiazol-3-yl}benzamide;

or a pharmaceutically acceptable salt thereof.
