Abstract

The present invention relates to methods of inhibiting the activity of Hsp90 in a subject in need thereof and methods for treating non-Hodgkin's lymphoma, in a subject in need thereof.

Claims (24)

1. A method of treating a B-cell non-Hodgkin's lymphoma in a subject, comprising administering to the subject an effective amount of a compound represented by formula (XIV):

or a tautomer, or a pharmaceutically acceptable salt, wherein

R 5 is an optionally substituted 8 to 14 membered aryl, wherein the optional substitution is selected from an alkyl, an alkenyl, an alkynyl, a cycloalkyl, haloalkyl, a heterocyclyl, an aryl, a heteroaryl, an aralkyl, a heteroaralkyl, cyano, halo, nitro, —OR 30 , or —NR 28 R 29 ;

R 7 and R 8 , for each occurrence, are, independently, —H, an alkyl, an alkenyl, an alkynyl, a cycloalkyl, a cycloalkenyl, a heterocyclyl, an aryl, a heteroaryl, an aralkyl, or a heteroaralkyl;

R 10 and R 11 , for each occurrence, are independently —H, an alkyl, an alkenyl, an alkynyl, a cycloalkyl, a cycloalkenyl, a heterocyclyl, an aryl, a heteroaryl, an aralkyl, or a heteroaralkyl; or R 10 and R 11 , taken together with the nitrogen to which they are attached, form a heterocyclyl or a heteroaryl;

R 26 is a lower alkyl;

R 28 and R 29 , for each occurrence are, independently, H, an alkyl, an alkenyl, an alkynyl, a cycloalkyl, a cycloalkenyl, a heterocyclyl, an aryl, a heteroaryl, an aralkyl, or a heteraralky; or R 28 and R 29 taken together with the nitrogen to which they are attached is heterocyclyl or heteroaryl;

R 30 for each occurrence is, independently, H, an alkyl, an alkenyl, an alkynyl, a cycloalkyl, a cycloalkenyl, a heterocyclyl, an aryl, an heteroaryl, an aralkyl, or a heteroaralkyl

p, for each occurrence, is, independently, 0, 1 or 2;

k is 1, 2, 3, or 4;

r is zero or an integer from 1 to 3; and

m, for each occurrence, is independently, 1, 2, 3, or 4.

2. The method of claim 1 , wherein R 5 is represented by the following formula:

R 9 , for each occurrence, is independently a substituent selected from the group consisting of an alkyl, an alkenyl, an alkynyl, a cycloalkyl, a heterocyclyl, an aryl, a heteroaryl, an aralkyl, a heteroaralkyl, halo, cyano, nitro, a haloalkyl, —NR 10 R 11 , —OR 7 , and

m′ is zero or an integer from 1 to 7.

The method of claim 2 , wherein the B-cell non-Hodgkin's lymphoma is selected from the group consisting of Burkitt's lymphoma, follicular lymphoma, diffuse large B-cell lymphoma, nodal marginal zone B-cell lymphoma, plasma cell neoplasms, small lymphocytic lymphoma/chronic lymphocytic leukemia, mantle cell lymphoma, and lymphoplamacytic lymphoma/Waldenstrom macroglobulinemia.

4. The method of claim 2 , wherein the compound is administered with an additional anticancer agent.

The method of claim 1 , wherein the B-cell non-Hodgkin's lymphoma is selected from the group consisting of Burkitt's lymphoma, follicular lymphoma, diffuse large B-cell lymphoma, nodal marginal zone B-cell lymphoma, plasma cell neoplasms, small lymphocytic lymphoma/chronic lymphocytic leukemia, mantle cell lymphoma, and lymphoplamacytic lymphoma/Waldenstrom macroglobulinemia.

6. The method of claim 1 , wherein the compound is administered with an additional anticancer agent.
