Abstract

The invention provides soluble fusion protein complexes having at least two soluble fusion proteins. The first fusion protein is a biologically active polypeptide covalently linked to an interleukin-15 (IL-15) polypeptide or a functional fragment thereof. The second fusion protein is a second biologically active polypeptide covalently linked to a soluble interleukin-15 receptor alpha (IL-15Rα) polypeptide or a functional fragment thereof. In the complexes of the invention, one or both of the first and second fusion proteins further includes an immunoglobulin Fc domain or a functional fragment thereof; and the first fusion protein binds to the soluble IL-15Rα domain of the second fusion protein to form a soluble fusion protein complex. The invention further provides methods for making and using the complexes of the invention.

Claims (14)

1. A nucleic acid sequence encoding an IL-15 receptor fusion polypeptide comprising an IL-15 sushi-binding domain fused to an Fc domain (IL-15RαSu/Fc), and a variant interleukin-15 polypeptide comprising an N72D mutation (IL-15N72D).

2. A DNA vector comprising the nucleic acid of claim 1 .

3. A method of making an IL-15N72D:IL-15RαSu/Fc fusion protein complex, the method comprising:

a) introducing into a host cell a first DNA encoding a variant interleukin-15 (IL-15) polypeptide comprising an N72D mutation (IL-15N72D) and a second DNA encoding an IL-15Rα/Fc fusion protein comprising an IL-15 sushi-binding domain fused to an Fc domain (IL-15RαSu/Fc);

b) culturing the host cell in media under conditions sufficient to express the IL-15N72D and the IL-15Rα/Fc fusion protein; and

c) purifying the IL-15:IL-15Rα/Fc complex from the host cell or media.

4. A method of making an IL-15N72D:IL-15RαSu/Fc fusion protein complex, the method comprising:

a) co-expressing a variant interleukin-15 (IL-15) polypeptide comprising an N72D mutation (IL-15N72D) and an IL-15RαSu/Fc fusion protein comprising an IL-15 sushi-binding domain fused to an Fc domain (IL-15RαSu/Fc) in a host cell;

b) culturing the host cell in media under conditions sufficient to express the IL-15N72D and the IL-15RαSu/Fc fusion protein; and

c) purifying the IL-15N72D:IL-15RαSu/Fc fusion protein complex from the host cell or media wherein both IL-15 binding sites of the IL-15N72D:IL- 15 RαSu/Fc complex are fully occupied.

5. A cell comprising a first polynucleotide encoding a variant interleukin-15 (IL-15) comprising an N72D mutation (IL-15N72D) and a second polynucleotide encoding an IL-15 receptor fusion protein comprising an IL-15 sushi-binding domain fused to an Fc domain (IL-15RαSu/Fc).

6. An isolated fully occupied IL-15N72D:IL-15RαSu/Fc complex comprising:

(1) a dimer comprising two interleukin-15 (IL-15) receptor fusion polypeptides, wherein each IL-15 receptor fusion polypeptide comprises an IL-15 sushi-binding domain fused to an Fc domain (IL-15RαSu/Fc), and

(2) two variant interleukin-15 polypeptides comprising an N72D mutation (IL-15N72D polypeptides), wherein each interleukin-15 sushi-binding domain is bound to an IL-15N72D polypeptide.
