Abstract

The present invention relates to substituted imidazoquinoxaline compounds of general formula (I) as inhibitors of Mps-1 Kinase or TTK, and being active against inflammation and cancer.

Claims (414)

1. A compound of general formula (I):

in which:

R 1 represents a C 2 -C 10 -alkyl-, halo-C 1 -C 10 -alkyl-, HO—C 1 -C 10 -alkyl-, C 1 -C 10 -alkoxy-C 1 -C 10 -alkyl-, aryloxy-C 1 -C 10 -alkyl-, halo-C 1 -C 10 -alkoxy-C 1 -C 10 -alkyl-, C 2 -C 10 -alkenyl-, C 2 -C 10 -alkynyl-, C 3 -C 10 -cycloalkyl-, 3- to 10-membered heterocycloalkyl-, aryl-, heteroaryl-, C 1 -C 6 -alkylene-aryl-, C 1 -C 6 -alkylene-heteroaryl-, —C 1 -C 6 -alkylene-C 3 -C 10 -cycloalkyl, —C 1 -C 6 -alkylene-(3- to 10-membered hetero cycloalkyl), —C 1 -C 6 -alkylene-aryl, —C 1 -C 6 -alkylene-heteroaryl group;

wherein said group is optionally substituted one or more times, in the same way or differently, with a substituent selected from: halo-, hydroxyl-, cyano-, nitro-, C 1 -C 6 -alkyl-, halo-C 1 -C 6 -alkyl-, C 1 -C 6 -alkoxy-, halo-C 1 -C 6 -alkoxy-, C 1 -C 6 -alkoxy-C 1 -C 6 -alkyl-, halo-C 1 -C 6 -alkoxy-C 1 -C 6 -alkyl-, C 3 -C 10 -cycloalkyl-, 3- to 10-membered heterocycloalkyl-, —C(═O)OH, —C(═O)O—C 1 -C 6 -alkyl, —C(═O)O—C 3 -C 10 -cycloalkyl, —OC(═O)—C 1 -C 6 -alkyl, —OC(═O)—C 3 -C 10 -cycloalkyl, —N(H)C(═O)R 8 , —N(C 1 -C 6 -alkyl)C(═O)R 8 , —N(H)S(═O) 2 R 8 , —N(C 1 -C 6 -alkyl)S(═O) 2 R 8 , —C(═O)NR 9 R 10 , —OC(═O)NR 9 R 10 , —N(H)C(═O)OR 8 , —N(C 1 -C 6 -alkyl)C(═O)OR 8 , —N(H)C(═O)NR 9 R 10 , —N(C 1 -C 6 -alkyl)C(═O)NR 9 R 10 , —SR 8 , —S(═O)R 8 , —S(═O) 2 R 8 , —S(═O) 2 NR 9 R 10 , —NR 9 R 10 ;

R 9 , R 10 , together with the nitrogen atom to which they are attached, represent a 3- to 10-membered heterocycloalkyl group;

under the proviso that:

R 1 is not —CH 2 CH 2 CH 2 OH,

—CH 2 CH 2 CH 2 O(C═O)—R a ,

—CH 2 CH 2 CH 2 O(C═O)—OR a ,

—CH 2 CH 2 CH 2 O(C═O)—CHR a NR b R c ,

in which R a , R b , R c , identical or different, are selected from the group consisting of H, C 1 -C 10 -alkyl, C 2 -C 10 -alkenyl, C 2 -C 10 -alkynyl, phenyl and heteroaryl;

or a stereoisomer, a tautomer, an N-oxide, or a salt thereof, or a mixture of same.

2. The compound according to claim 1 , wherein:

R 1 represents a C 2 -C 10 -alkyl-, halo-C 1 -C 10 -alkyl-, HO—C 1 -C 10 -alkyl-, C 1 -C 10 -alkoxy-C 1 -C 10 -alkyl-, aryloxy-C 1 -C 10 -alkyl-, halo-C 1 -C 10 -alkoxy-C 1 -C 10 -alkyl-, C 2 -C 10 -alkenyl-, C 2 -C 10 -alkynyl-, C 3 -C 10 -cycloalkyl-, 3- to 10-membered heterocycloalkyl-, aryl-, C 1 -C 6 -alkylene-aryl-, C 1 -C 6 -alkylene-heteroaryl-, —C 1 -C 6 -alkylene-C 3 -C 10 -cycloalkyl, —C 1 -C 6 -alkylene-(3- to 10-membered heterocycloalkyl), —C 1 -C 6 -alkylene-aryl, —C 1 -C 6 -alkylene-heteroaryl group;

5. A method of preparing a compound according to claim 1 , in which method an intermediate compound of general formula (4):

wherein R 1 , R 3 , R 4 , R 5 , R 6 , and R 7 are as defined in claim 1 , and Y represents a halogen atom, is allowed to react with an intermediate compound of general formula:

R 2 —Z

in which R 2 is as defined in claim 1 , and Z is a suitable functional group via which the R 2 of the R 2 —Z compound can be coupled onto the Y-bearing carbon atom of a compound (4), thereby replacing said Y with said R 2 moiety,

thus providing a compound of general formula (I):

in which R 1 , R 2 , R 3 , R 4 , R 5 , R 6 and R 7 are as defined in claim 1 .

6. A pharmaceutical composition comprising a compound according to claim 1 , and a pharmaceutically acceptable diluent or carrier.

7. A pharmaceutical combination comprising:

one or more compounds according to claim 1 ;

and

one or more agents selected from: a taxane; an epothilone; Mitoxantrone; Prednisolone; Dexamethasone; Estramustine; Vinblastine; Vincristine; Doxorubicin; Idarubicin; Daunorubicin; Bleomycin; Etoposide; Cyclophosphamide; Ifosfamide; Procarbazine; Melphalan; 5-Fluorouracil; Capecitabine; Fludarabine; Cytarabine; 2-Chloro-2′-deoxyadenosine; Thioguanine; an anti-androgen; Bortezomib; a platinum derivative; Chlorambucil; Methotrexate; and Rituximab.

8. The pharmaceutical combination according to claim 7 , wherein the taxane is selected from Docetaxel and Paclitaxel.

9. The pharmaceutical combination according to claim 7 , wherein the epothilone is selected from Ixabepilone, Patupilone, and Sagopilone.

10. The pharmaceutical combination according to claim 7 , wherein the anti-androgen is selected from Flutamide, Cyproterone acetate, and Bicalutamide.

11. The pharmaceutical combination according to claim 7 , wherein the platinum derivative is selected from Cisplatin and Carboplatin.

12. The compound according to claim 1 , wherein:

R 2 represents an aryl- or heteroaryl-group; wherein said group is substituted one time with —C(═O)NR 9 R 10 and is optionally substituted with a substituent selected from: halo-, hydroxy-, cyano-, C 1 -C 6 -alkyl-, halo-C 1 -C 6 -alkyl-.

13. The compound according to claim 1 , wherein:

R 2 represents an aryl- or heteroaryl-group; wherein said group is substituted one time with —C(═O)NH-cyclopropyl and is optionally substituted with a substituent selected from: halo-, hydroxy-, cyano-, C 1 -C 6 -alkyl-, halo-C 1 -C 6 -alkyl-.
