Abstract

The invention relates to the isolation and characterization of a maize gene, RAMOSA3 (RA3), responsible for meristem development and inflorescence development including branching. The gene, gene product, and regulatory regions may be used to manipulate branching, meristem growth, inflorescence development and arrangement, and ultimately to improve yield of plants. The invention includes the gene and protein product as well as the use of the same for temporal and spatial expression in transgenic plants to alter plant morphology and affect yield in plants. The invention also includes the gene and protein product for SISTER OF RAMOSA3 (SRA).

Claims (30)

1. An isolated polynucleotide comprising:

(a) a nucleotide sequence encoding a polypeptide associated with branching of the tassel, ear, or both, of a maize plant, wherein said polypeptide has an amino acid sequence of at least 90% sequence identity, based on the Clustal V method of alignment, when compared to SEQ ID NO:19; or

(b) a complement of the nucleotide sequence, wherein the complement and the nucleotide sequence consist of the same number of nucleotides and are 100% complementary.

2. An isolated polynucleotide comprising:

(a) a nucleotide sequence encoding a polypeptide associated with pollen shed of a plant, wherein said polypeptide has an amino acid sequence of at least 90% sequence identity, based on the Clustal V method of alignment, when compared to SEQ ID NO:19; or

(b) a complement of the nucleotide sequence, wherein the complement and the nucleotide sequence consist of the same number of nucleotides and are 100% complementary.

3. A recombinant DNA construct comprising the polynucleotide of claim 1 operably linked to a promoter that is functional in a plant.

4. A recombinant DNA construct comprising the polynucleotide of claim 2 operably linked to a promoter that is functional in a plant.

5. A method for altering branching of the tassel, ear, or both, of a maize plant, comprising:

(a) introducing into a regenerable maize plant cell the recombinant DNA construct of claim 3 to produce a transformed maize plant cell; and

(b) regenerating a transgenic maize plant from said transformed maize plant cell, wherein said transgenic maize plant comprises in its genome said recombinant DNA construct and wherein said transgenic maize plant exhibits an alteration in branching of the tassel, ear, or both, when compared to a control maize plant not comprising said recombinant DNA construct.

6. The method of claim 5 , further comprising (c) obtaining a progeny maize plant derived from said transgenic maize plant, wherein said progeny maize plant comprises in its genome the recombinant DNA construct.

7. The method of claim 5 , wherein the transgenic maize plant exhibits a decrease in branching of the tassel, ear, or both.

8. A method for altering pollen shed of a plant, comprising:

(a) introducing into a regenerable plant cell the recombinant DNA construct of claim 4 to produce a transformed plant cell; and

(b) regenerating a transgenic plant from said transformed plant cell, wherein said transgenic plant comprises in its genome said recombinant DNA construct and wherein said transgenic plant exhibits an alteration in pollen shed, when compared to a control plant not comprising said recombinant DNA construct.

9. The method of claim 8 , further comprising (c) obtaining a progeny plant derived from said transgenic plant, wherein said progeny plant comprises in its genome the recombinant DNA construct.

10. The method of claim 8 , wherein the transgenic plant exhibits a decrease in pollen shed.

11. The polynucleotide of claim 1 , wherein the amino acid sequence of the polypeptide has at least 95% sequence identity, based on the Clustal V method of alignment, when compared to SEQ ID NO:19.

12. The polynucleotide of claim 1 wherein the amino acid sequence of the polypeptide comprises SEQ ID NO:19.

13. The polynucleotide of claim 1 , wherein the nucleotide sequence comprises SEQ ID NO:18.

14. A maize plant comprising the recombinant DNA construct of claim 3 .

15. A maize seed comprising the recombinant DNA construct of claim 3 .

16. The method of claim 7 , wherein the transgenic maize plant further exhibits a decrease in pollen shed.

17. The polynucleotide of claim 2 , wherein the amino acid sequence of the polypeptide has at least 95% sequence identity, based on the Clustal V method of alignment, when compared to SEQ ID NO:19.

18. The polynucleotide of claim 2 wherein the amino acid sequence of the polypeptide comprises SEQ ID NO:19.

19. The polynucleotide of claim 2 , wherein the nucleotide sequence comprises SEQ ID NO:18.

20. A plant comprising the recombinant DNA construct of claim 4 .

21. A seed comprising the recombinant DNA construct of claim 4 .

22. The method of claim 8 , wherein the plant is maize.
