Antiviral drugs for treatment of arenavirus infection
View Patent ↗Methods, compounds, and pharmaceutical compositions for treating arenavirus infections are disclosed. Methods for preparing the compounds and pharmaceutical compositions are also disclosed. The novel compounds and compositions are used for treatment of arenavirus infections, which include but are not limited to the following viruses: Junin, Machupo, Guanarito, Sabia, Lassa, Tacaribe, Pichinde, and LCMV.
1. A method for the treatment of an Arenavirus infection comprising administering in a therapeutically effective amount to a mammal in need thereof, a compound of Formula I below:
wherein R 1 and R 2 are independently hydrogen, alkyl, alkenyl, alkynyl, cycloalkyl, arylalkyl, aryl, acyl, arylacyl, hydroxy, alkyloxy, alkylthio, amino, alkylamino, acetamido, halogen, cyano or nitro;
R 3 is hydrogen, acyl, arylacyl or sulfonyl; and
Ar 1 and Ar 2 are independently (un)substituted aryl or heteroaryl; and wherein compound of Formula I is selected from the group consisting of:

2. The method of claim 1 , wherein the compound of Formula I is 5-[(4-isopropylphenyl)methylamino]-1-(4-methoxyphenyl)benzimidazole-2-carbonitrile.
3. The method of claim 1 , wherein the mammal is a human.
4. The method of claim 1 , wherein the Arenavirus is selected from the group consisting of Junin, Machupo, Tacaribe, Guanarito, Machupo, Pichinde and Lassa.
5. The method of claim 1 , which further comprises co-administering a combination of cidofovir, cyclic cidofovir, or salts thereof.
