Abstract

The present invention concerns methods and compositions for introducing miRNA activity or function into cells using synthetic nucleic acid molecules. Moreover, the present invention concerns methods and compositions for identifying miRNAs with specific cellular functions that are relevant to therapeutic, diagnostic, and prognostic applications wherein synthetic miRNAs and/or miRNA inhibitors are used in library screening assays.

Claims (16)

1. A method for inhibiting cell proliferation of a cancer cell comprising administering to the cancer cell an effective amount of a single-stranded synthetic miRNA inhibitor between 17 and 25 nucleotides in length having a sequence that is at least 90% complementary to the sequence of a mature human miR-21 wherein the cancer cells is a breast cancer cell, a colon cancer cell, a thyroid cancer cell, or a lung cancer cell.

2. The method of claim 1 , wherein the synthetic miRNA inhibitor is 22 or 23 nucleotides in length.

3. The method of claim 1 , wherein the miRNA inhibitor further comprises one or more sugar modifications.

4. The method of claim 3 , wherein the sugar modification is 2′OMe.

5. The method of claim 1 , wherein the miRNA inhibitor comprises RNA.

A method for treating cancer in a human subject comprising administering to the subject an effective amount of a composition comprising a single-stranded synthetic miRNA inhibitor between 17 and 25 nucleotides in length having a sequence that is at least 90% complementary to the sequence of a mature human miR-21 wherein the cancer is a breast cancer, a colon cancer, a thyroid cancer, or a lung cancer.

7. The method of claim 6 , wherein the miRNA inhibitor is 22 or 23 nucleotides in length.

8. The method of claim 6 , wherein the miRNA inhibitor further comprises one or more sugar modifications.

9. The method of claim 8 , wherein the sugar modification is 2′OMe.

10. The method of claim 6 , wherein the miRNA inhibitor comprises RNA.

A method for inhibiting cell proliferation of a cancer cell comprising administering to the cancer cell an miRNA inhibitor between 17 and 25 nucleotides in length having a sequence that is at least 90% complementary to the sequence of a mature human miR-21, wherein the inhibitor is a single-stranded oligonucleotide comprising at least a sugar modification or a terminal modification wherein the cancer cells is a breast cancer cell, a colon cancer cell, a thyroid cancer cell, or a lung cancer cell.

12. The method of claim 11 , wherein the oligonucleotide is a ribo-oligonucleotide.

13. The method of claim 11 , wherein the modification is 2′OMe.

14. A method for treating cancer in a human subject comprising administering to the subject a composition comprising a miRNA inhibitor between 17 and 25 nucleotides in length having a sequence that is at least 90% complementary to the sequence of a mature human miR-21, wherein the inhibitor is a single-stranded oligonucleotide comprising at least a sugar modification or a terminal modification wherein the cancer is a breast cancer, a colon cancer, a thyroid cancer, or a lung cancer.

15. The method of claim 14 , wherein the modification is 2′OMe.

16. The method of claim 14 , wherein the oligonucleotide is a ribo-oligonucleotide.
