Abstract

The present invention relates to anti-IL13 antibodies that bind specifically and with high affinity to both glycosylated and non-glycosylated human IL13, does not bind mouse IL13, and neutralize human IL13 activity at an approximate molar ratio of 1:2 (MAb:IL13). The invention also relates to the use of these antibodies in the treatment of IL13-mediated diseases, such as allergic disease, including asthma, allergic asthma, non-allergic (intrinsic) asthma, allergic rhinitis, atopic dermatitis, allergic conjunctivitis, eczema, urticaria, food allergies, chronic obstructive pulmonary disease, ulcerative colitis, RSV infection, uveitis, scleroderma, and osteoporosis.

Claims (25)

1. A monoclonal anti-IL-13 antibody that binds specifically to human IL-13, wherein said antibody binds to the same epitope of human IL-13 as an antibody produced by hybridoma 228B/C-1 designated American Type Culture Collection (“ATCC”) accession number PTA-5657.

2. The anti-IL-13 antibody of claim 1 , comprising the complementarity determining regions (CDRs) of the antibody produced by hybridoma 228B/C-1 (PTA-5657).

3. The anti-IL-13 antibody of claim 1 , wherein said anti-IL-13 antibody binds to human IL-13 with a dissociation constant (K D ) of 10 −10 M to 10 −12 M.

4. The anti-IL-13 antibody of claim 1 , wherein said anti-IL-13 antibody competitively inhibits binding of the antibody produced by the hybridoma cell line deposited under ATCC accession number PTA-5657 to human IL-13 by at least 50%.

5. The anti-IL-13 antibody of claim 1 , wherein said anti-IL-13 antibody competitively inhibits binding of the antibody produced by the hybridoma cell line deposited under ATCC accession number PTA-5657 to human IL-13 by at least 60%.

6. The anti-IL-13 antibody of claim 1 , wherein said anti-IL-13 antibody competitively inhibits binding of the antibody produced by the hybridoma cell line deposited under ATCC accession number PTA-5657 to human IL-13 by at least 70%.

7. The anti-IL-13 antibody of claim 1 , wherein said anti-IL-13 antibody is a humanized antibody.

8. The anti-IL-13 antibody of claim 1 , wherein said anti-IL-13 antibody is a chimeric antibody or human antibody.

9. The anti-IL-13 antibody of claim 1 , wherein said anti-IL-13 antibody is a bispecific antibody.

10. The anti-IL-13 antibody of claim 1 , wherein said anti-IL-13 antibody is of the IgG1 isotype.

11. The anti-IL-13 antibody of claim 1 , wherein said anti-IL-13 antibody is an antigen-binding fragment, a Fab, Fab′, F(ab′) 2 , Fd, single-chain Fv (scFv), single-chain antibody, disulfide-linked Fv (sdFv), or single-domain antibody.

12. The anti-IL-13 antibody of claim 1 , wherein said anti-IL-13 antibody neutralizes human IL-13 activity.

13. The anti-IL-13 antibody of claim 1 , wherein said antibody specifically binds to a peptide consisting essentially of the amino acid sequence ESLINVSG (SEQ ID NO: 18) or YCAALESLINVS (SEQ ID NO: 19).

14. A composition comprising the anti-IL-13 antibody of claim 2 .

15. A composition comprising the anti-IL-13 antibody of claim 7 .

16. A composition comprising the anti-IL-13 antibody of claim 8 .

17. A composition comprising the anti-IL-13 antibody of claim 9 .

18. A composition comprising the anti-IL-13 antibody of claim 12 .

19. The anti-IL-13 antibody of claim 1 which comprises a heavy chain variable region comprising complementarity determining regions CDRH1, CDRH2 and CDRH3 and a light chain variable region comprising complementarity determining regions CDRL1, CDRL2 and CDRL3, wherein:

(a) CDRH1 has the amino acid sequence of SEQ ID NO: 117, 118, 119, 120, 121 or 122;

(b) CDRH2 has the amino acid sequence of SEQ ID NO: 123, 124, 125, 126, 127, 128, 129, 130, 131, 132, 133 or 134;

(c) CDRH3 has the amino acid sequence of SEQ ID NO: 135, 136, 137, 138, 139, 140 or 141;

(d) CDRL1 has the amino acid sequence of SEQ ID NO: 99, 100, 101, 102, or 103;

(e) CDRL2 has the amino acid sequence of SEQ ID NO: 104, 105, 106, 107, 108, 109, 110, 111, 112, 113, or 114; and

(f) CDRL3 has the amino acid sequence of SEQ ID NO: 115 or 116.
