Abstract

The present invention discloses humanised anti-IL-18 antibodies, methods of manufacture, and methods of treatment with said antibodies. Further disclosed are screening methods using for example surface plasmon resonance to identify antibodies with therapeutic potential.

Claims (14)

1. A pharmaceutical composition comprising a pharmaceutical carrier and a humanized anti-interleukin-18 antibody; said humanized anti-interleukin-18 antibody comprising a heavy chain and light chain comprising the following complementarity determining regions:

a CDRH1 with the amino acid sequence shown in SEQ ID NO:1;

a CDRH2 with the amino acid sequence shown in SEQ ID NO:2;

a CDRH3 with the amino acid sequence shown in SEQ ID NO:3;

a CDRL1 with the amino acid sequence shown in SEQ ID NO:4;

a CDRL2 with the amino acid sequence shown in SEQ ID NO:5; and

a CDRL3 with the amino acid sequence shown in SEQ ID NO:6,

wherein said antibody comprises a heavy chain selected from the group consisting of: the amino acid sequence shown in SEQ ID NO: 9, the amino acid sequence shown in SEQ ID NO: 17, and the amino acid sequence shown in SEQ ID NO: 21; and a light chain selected from the group consisting of: the amino acid sequence shown in SEQ ID NO: 13 and the amino acid sequence shown in SEQ ID NO: 29.

The pharmaceutical composition of claim 1 , wherein said humanized anti-interleukin-18 antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 9 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 13, or a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 9 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 29.

The pharmaceutical composition of claim 1 , wherein said humanized anti-interleukin-18 antibody comprises a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 17 and a light chain comprising the amino acid sequence shown in SEQ ID NO:13, or a heavy chain comprising the amino acid sequence shown in SEQ ID NO: 17 and a light chain comprising the amino acid sequence shown in SEQ ID NO: 29.

A pharmaceutical composition comprising a pharmaceutical carrier and a humanized anti-interleukin-18 antibody; said humanized anti-interleukin-18 antibody comprising a heavy chain comprising the amino acid sequence shown in SEQ ID NO:21 and a light chain comprising the amino acid sequence shown in SEQ ID NO:13, or a heavy chain comprising the amino acid sequence shown in SEQ ID NO:21 and a light chain comprising the amino acid sequence shown in SEQ ID NO:29.

A pharmaceutical composition comprising a pharmaceutical carrier and a humanized anti-interleukin-18 antibody; said humanized anti-interleukin-18 antibody comprising a heavy chain comprising the amino acid sequence shown in SEQ ID NO:9 and a light chain comprising the amino acid sequence shown in SEQ ID NO:13.

A pharmaceutical composition comprising a pharmaceutical carrier and a humanized anti-interleukin-18 antibody; said humanized anti-interleukin-18 antibody comprising a heavy chain selected from the group consisting of: the amino acid sequence shown in SEQ ID NO:9, the amino acid sequence shown in SEQ ID NO:17, and the amino acid sequence shown in SEQ ID NO:21.

A pharmaceutical composition comprising a pharmaceutical carrier and a humanized anti-interleukin-18 antibody; said humanized anti-interleukin-18 antibody comprising a light chain selected from the group consisting of: the amino acid sequence shown in SEQ ID NO:13 and the amino acid sequence shown in SEQ ID NO:29.
