Abstract

This invention relates to compounds, pharmaceutical compositions and methods for use in the prevention and treatment of cerebral insufficiency, including enhancement of receptor functioning in synapses in brain networks responsible for basic and higher order behaviors. These brain networks, which are involved in cognitive abilities related to memory impairment, such as is observed in a variety of dementias, in imbalances in neuronal activity between different brain regions, as is suggested in disorders such as Parkinson's disease, schizophrenia, sleep apneas, attention deficit hyperactivity disorder and affective or mood disorders, and in disorders wherein a deficiency in neurotrophic factors is implicated, as well as conditions such as stroke-induced central sleep apnea, obstructive sleep apnea, congenital hypoventilation syndrome, among others. In a particular aspect, the invention relates to bicyclic amide compounds useful for treatment of such conditions, and methods of using these compounds for such treatment.

Claims (20)

1. A method of treating a disease state or condition in a patient in need, said disease state or condition being selected from the group consisting of schizophrenia, Parkinson's disease, Alzheimer's disease, ADHD, Rett syndrome, sleep apnea, depression and bipolar disorder, said method comprising administering to said patient an effective amount of a compound according to formula A:

X═O, or (CH 2 ) n

n=0 or 1, or a pharmaceutically acceptable salt thereof.

2. The method according to claim 1 wherein X is O.

3. The method according to claim 1 wherein X is (CH 2 ) n .

4. The method according to claim 1 wherein n is 1.

5. The method according to claim 3 wherein n is 0.

6. The method according to claim 1 wherein said disease state or condition is schizophrenia.

7. The method according to claim 1 wherein said disease state or condition is Parkinson's disease.

8. The method according to claim 1 wherein said disease state or condition is Alzheimer's disease.

9. The method according to claim 1 wherein said disease state or condition is ADHD.

10. The method according to claim 1 wherein said disease state or condition is Rett syndrome.

11. The method according to claim 1 wherein said disease state or condition is depression.

12. The method according to claim 1 wherein said disease state or condition is bipolar disorder.

13. The method according to claim 1 wherein said disease state or condition is sleep apnea.

14. The method according to claim 2 wherein said disease state or condition is sleep apnea.

15. The method according to claim 3 wherein said disease state or condition is sleep apnea.

16. The method according to claim 4 wherein said disease state or condition is sleep apnea.

17. The method according to claim 5 wherein said disease state or condition is sleep apnea.
