Abstract

The present invention relates to inhibitors of 11-β hydroxyl steroid dehydrogenase type 1, antagonists of the mineralocorticoid receptor (MR), and pharmaceutical compositions thereof. The compounds of the invention can be useful in the treatment of various diseases associated with expression or activity of 11-β hydroxyl steroid dehydrogenase type 1 and/or diseases associated with aldosterone excess.

Claims (650)

1. A compound of Formula I:

or a pharmaceutically acceptable salt thereof, wherein:

Cy is aryl, heteroaryl, cycloalkyl, or heterocycloalkyl, each optionally substituted with 1, 2, 3, 4 or 5 —W—X—Y—Z;

L is (CR 12 R 13 ) q1 , (CR 12 R 13 ) q1 O(CR 12 R 13 ) q2 , (CR 12 R 13 ) q1 S(CR 12 R 13 ) q2 , (CR 12 R 13 ) q1 SO(CR 12 R 13 ) q2 , (CR 12 R 13 ) q1 SO(CR 12 R 13 ) q2 , or (CR 12 R 13 ) q1 CO(CR 12 R 13 ) q2 ;

Q is —(CR 1 R 2 ) m -A;

A is cycloalkyl or heterocycloalkyl, each optionally substituted with 1, 2, 3, 4 or 5 —W′—X′—Y′—Z′;

E is —(CR 3a R 3b ) n1 —, —(CR 3a R 3b ) n2 CO—, —(CR 3a R 3b ) n2 OCO—, —(CR 3a R 3b ) n2 SO—, —(CR 3a R 3b ) n2 SO 2 —, —(CR 3a R 3b ) n2 NR 3c —, or a group of formula:

D 1 , D 2 , D 3 and D 4 are each N or CR 15 ;

R 1 and R 2 are each, independently, H or C 1-8 alkyl;

R 3a and R 3b are each, independently, H, OC(O)R a′ , OC(O)OR b′ , C(O)OR b′ , OC(O)NR c′ R d′ , NR c′ R d′ , NR c′ (O)R a′ , NR c′ (O)OR b′ , S(O)R a′ , S(O)NR c′ R d′ , S(O) 2 R a′ , S(O) 2 NR c′ R d′ , OR b′ , SR b′ , halo, C 1-10 alkyl, C 1-10 haloalkyl, C 2-10 alkenyl, C 2-10 alkynyl, aryl, cycloalkyl, heteroaryl, heterocycloalkyl, arylalkyl, heteroarylalkyl, cycloalkylalkyl or heterocycloalkylalkyl, wherein said C 1-10 alkyl, C 1-10 haloalkyl, C 2-10 alkenyl, C 2-10 alkynyl, aryl, cycloalkyl, heteroaryl, heterocycloalkyl, arylalkyl, heteroarylalkyl, cycloalkylalkyl or heterocycloalkylalkyl is optionally substituted by R 16 ;

R 3c is H, C 1-4 alkyl, C 1-4 haloalkyl, C 2-4 alkenyl, C 2-4 alkynyl, or CO—(C 1-4 alkyl);

R 4 , R 5 , R 6 , R 7 , R 8 , R 9 , R 10 and R 11 are each, independently, H, OC(O)R a′ , OC(O)OR b′ , C(O)OR b′ , OC(O)NR c′ R d′ , NR c′ R d′ , NR c′ C(O)R a′ , NR c′ C(O)OR b′ , S(O)R a′ , S(O)NR c′ R d′ , S(O) 2 R a′ , S(O) 2 NR c′ R d′ , OR b′ , SR b′ , halo, C 1-10 alkyl, C 1-10 haloalkyl, C 2-10 alkenyl, C 2-10 alkynyl, aryl, cycloalkyl, heteroaryl, heterocycloalkyl, arylalkyl, heteroarylalkyl, cycloalkylalkyl or heterocycloalkylalkyl, wherein said C 1-10 alkyl, C 1-10 haloalkyl, C 2-10 alkenyl, C 2-10 alkynyl, aryl, cycloalkyl, heteroaryl, heterocycloalkyl, arylalkyl, heteroarylalkyl, cycloalkylalkyl or heterocycloalkylalkyl is optionally substituted by R 14 ;

or R 4 and R 5 together with the carbon atom to which they are attached form a 3-14 membered cycloalkyl or heterocycloalkyl group which is optionally substituted by R 14 ;

or R 6 and R 7 together with the carbon atom to which they are attached form a 3-14 membered cycloalkyl or heterocycloalkyl group which is optionally substituted by R 14 ;

or R 8 and R 9 together with the carbon atom to which they are attached form a 3-14 membered cycloalkyl or heterocycloalkyl group which is optionally substituted by R 14 ;

or R 10 and R 11 together with the carbon atom to which they are attached form a 3-14 membered cycloalkyl or heterocycloalkyl group which is optionally substituted by R 14 ;

or R 4 and R 6 together with the carbon atom to which they are attached form a 3-7 membered fused cycloalkyl group or 3-7 membered fused heterocycloalkyl group which is optionally substituted by R 14 ;

or R 6 and R 8 together with the carbon atom to which they are attached form a 3-7 membered fused cycloalkyl group or 3-7 membered fused heterocycloalkyl group which is optionally substituted by R 14 ;

or R 4 and R 9 together form a C 1-3 alkylene bridge which is optionally substituted by R 14 ;

or R 4 and R 19 together form a C 1-3 alkylene bridge which is optionally substituted by R 14 ;

or R 6 and R 10 together form a C 1-3 alkylene bridge which is optionally substituted by R 14 ;

or R 9 and R 10 together form a C 1-3 alkylene bridge which is optionally substituted by R 14 ;

R 12 and R 13 are each, independently, H, halo, C 1-4 alkyl, C 1-4 haloalkyl, aryl, cycloalkyl, heteroaryl, heterocycloalkyl, CN, NO 2 , OR a′ , SR a′ , C(O)R b′ , C(O)NR c′ R d′ , C(O)OR a′ , OC(O)R b′ , OC(O)NR c′ R d′ , NR c′ R d′ , NR c′ C(O)R d′ , NR c′ (O)OR a′ , S(O)R b′ , S(O)NR c′ R d′ , S(O) 2 R b′ , or S(O) 2 NR c′ R d′ ;

R 14 is halo, C 1-4 alkyl, C 1-4 haloalkyl, aryl, cycloalkyl, heteroaryl, heterocycloalkyl, CN, NO 2 , OR a′ , SR a′ , C(O)R b′ , C(O)NR c′ R d′ , C(O)OR a′ , OC(O)R b′ , OC(O)NR c′ R d′ , NR c′ R d′ , NR c′ C(O)R d′ , NR c′ C(O)OR a′ , S(O)R b′ , S(O)NR c′ R d′ , S(O) 2 R b′ , or S(O) 2 NR c′ R d′ ;

R 15 is H, halo, C 1-4 alkyl, C 1-4 haloalkyl, aryl, cycloalkyl, heteroaryl, heterocycloalkyl, CN, NO 2 , OR a″ , SR a″ , C(O)R b″ , C(O)NR c″ R d″ , C(O)OR a″ , OC(O)R b″ , OC(O)NR c″ R d″ , NR c″ R d″ , NR c″ C(O)R d″ , NR c″ C(O)OR a″ , S(O)R b″ , S(O)NR c″ R d″ , S(O) 2 R b″ , or S(O) 2 NR c″ R d″ ;

R 16 is halo, C 1-4 alkyl, C 1-4 haloalkyl, aryl, cycloalkyl, heteroaryl, heterocycloalkyl, CN, NO 2 , OR a′ , SR a′ , C(O)R b′ , C(O)NR c′ R d′ , C(O)OR a′ , OC(O)R b′ , OC(O)NR c′ R d′ , NR c′ R d′ , NR c′ (O)R d′ , NR c′ (O)OR a′ , S(O)R b′ , S(O)NR c′ R d′ , S(O) 2 R b′ , or S(O) 2 NR c′ R d′ ;

W, W′ and W″ are each, independently, absent, C 1-6 alkylenyl, C 2-6 alkenylenyl, C 2-6 alkynylenyl, O, S, NR e , CO, COO, CONR e , SO, SO 2 , SONR e , or NR e CONR f , wherein said C 1-6 alkylenyl, C 2-6 alkenylenyl, C 2-6 alkynylenyl are each optionally substituted by 1, 2 or 3 halo, OH, C 1-4 alkoxy, C 1-4 haloalkoxy, amino, C 1-4 alkylamino or C 2-8 dialkylamino;

X, X′ and X″ are each, independently, absent, C 1-6 alkylenyl, C 2-6 alkenylenyl, C 2-6 alkynylenyl, aryl, cycloalkyl, heteroaryl or heterocycloalkyl, wherein said C 1-6 alkylenyl, C 2-6 alkenylenyl, C 2-6 alkynylenyl, aryl, cycloalkyl, heteroaryl or heterocycloalkyl is optionally substituted by one or more halo, oxo, CN, NO 2 , OH, C 1-4 alkoxy, C 1-4 haloalkoxy, amino, C 1-4 alkylamino or C 2-8 dialkylamino;

Y, Y′ and Y″ are each, independently, absent, C 1-6 alkylenyl, C 2-6 alkenylenyl, C 2-6 alkynylenyl, O, S, NR e , CO, COO, CONR e , SO, SO 2 , SONR e , or NR e CONR f , wherein said C 1-6 alkylenyl, C 2-6 alkenylenyl, C 2-6 alkynylenyl are each optionally substituted by 1, 2 or 3 halo, OH, C 1-4 alkoxy, C 1-4 haloalkoxy, amino, C 1-4 alkylamino or C 2-8 dialkylamino;

Z, Z′ and Z″ are each, independently, H, halo, CN, NO 2 , OH, C 1-4 alkoxy, C 1-4 haloalkoxy, amino, C 1-4 alkylamino, C 2-8 dialkylamino, C 1-6 alkyl, C 2-6 alkenyl, C 2-6 alkynyl, aryl, cycloalkyl, heteroaryl or heterocycloalkyl, wherein said C 1-6 alkyl, C 2-6 alkenyl, C 2-6 alkynyl, aryl, cycloalkyl, heteroaryl or heterocycloalkyl is optionally substituted by 1, 2 or 3 halo, oxo, C 1-6 alkyl, C 2-6 alkenyl, C 2-6 alkynyl, C 1-4 haloalkyl, aryl, cycloalkyl, heteroaryl, heterocycloalkyl, CN, NO 2 , OR a , SR a , C(O)R b , C(O)NR c R d , C(O)OR a , OC(O)R b , OC(O)NR c R d , NR c R d , NR c C(O)R d , NR c C(O)OR a , NR c S(O) 2 R b , S(O)R b , S(O)NR c R d , S(O) 2 R b , or S(O) 2 NR c R d ;

wherein two —W—X—Y—Z attached to the same atom optionally form a 3-20 membered cycloalkyl or heterocyloalkyl group, each optionally substituted by 1, 2 or 3 —W″—X″—Y″—Z″;

wherein two —W′—X′—Y′—Z′ attached to the same atom optionally form a 3-20 membered cycloalkyl or heterocyloalkyl group, each optionally substituted by 1, 2 or 3 —W″—X″—Y″—Z″;

wherein —W—X—Y—Z is other than H;

wherein —W′—X′—Y′—Z′ is other than H;

wherein —W″—X″—Y″—Z″ is other than H;

R a , R a′ and R a″ are each, independently, H, C 1-6 alkyl, C 1-6 haloalkyl, C 2-6 alkenyl, C 2-6 alkynyl, aryl, cycloalkyl, heteroaryl or heterocycloalkyl, wherein said C 1-6 alkyl, C 1-6 haloalkyl, C 2-6 alkenyl, C 2-6 alkynyl, aryl, cycloalkyl, heteroaryl or heterocycloalkyl; heterocycloalkyl, heterocycloalkylalkyl is optionally substituted with H, OH, amino, halo, C 1-6 alkyl, C 1-6 haloalkyl, aryl, arylalkyl, heteroaryl, heteroarylalkyl, cycloalkyl or heterocycloalkyl;

R b , R b′ and R b″ are each, independently, H, C 1-6 alkyl, C 1-6 haloalkyl, C 2-6 alkenyl, C 2-6 alkynyl, aryl, cycloalkyl, heteroaryl, heterocycloalkyl, arylalkyl, heteroarylalkyl, cycloalkylalkyl or heterocycloalkylalkyl, wherein said C 1-6 alkyl, C 1-6 haloalkyl, C 2-6 alkenyl, C 2-6 alkynyl, aryl, cycloalkyl, heteroaryl, heterocycloalkyl, arylalkyl, heteroarylalkyl, cycloalkylalkyl or heterocycloalkylalkyl is optionally substituted with H, OH, amino, halo, C 1-6 alkyl, C 1-6 haloalkyl, C 1-6 haloalkyl, aryl, arylalkyl, heteroaryl, heteroarylalkyl, cycloalkyl or heterocycloalkyl;

m is 0, 1, 2 or 3;

n1 is 1, 2, 3 or 4;

n2 is 0, 1, 2, 3 or 4;

n3 is 0, 1, 2, 3 or 4;

p is 0, 1 or 2;

q1 is 0, 1 or 2;

q2 is 0, 1 or 2; and

r is 0, 1 or 2;

with the proviso:

when Cy is aryl optionally substituted with 1, 2, 3, 4 or 5 —W—X—Y—Z or heteroaryl optionally substituted with 1, 2, 3, 4 or 5 —W—X—Y—Z; L is SO 2 , SO 2 CH 2 or CH 2 ; and m is 0, then A is other than tetrahydropyran-4-yl, 2,3-dihydroinden-2-yl or 2,2-difluoro-1,3-benzodioxol-5-yl.

2. The compound of claim 1 , or a pharmaceutically acceptable salt thereof, wherein Cy is aryl or heteroaryl, each optionally substituted with 1, 2, 3, 4 or 5 —W—X—Y—Z.

3. The compound of claim 1 , or a pharmaceutically acceptable salt thereof, wherein Cy is cycloalkyl or heterocycloalkyl, each optionally substituted with 1, 2, 3, 4 or 5 —W—X—Y—Z.

4. The compound of claim 1 , or a pharmaceutically acceptable salt thereof, wherein L is SO 2 .

5. The compound of claim 1 , or a pharmaceutically acceptable salt thereof, wherein L is CO.

6. The compound of claim 1 , or a pharmaceutically acceptable salt thereof, wherein L is (CR 12 R 13 ) q1 .

7. The compound of claim 1 , or a pharmaceutically acceptable salt thereof, wherein Q is cycloalkyl or heterocycloalkyl, each optionally substituted with 1, 2, 3, 4 or 5 —W′—X′—Y′—Z′.

8. The compound of claim 1 , or a pharmaceutically acceptable salt thereof, wherein Q is cyclohexyl substituted at the 4-position with at least one —W′—X′—Y′—Z′.

9. The compound of claim 1 , or a pharmaceutically acceptable salt thereof, wherein Q is cyclohexyl substituted at the 4-position with at least one OH, CN, or —O—X′—Y′—Z′.

10. The compound of claim 1 , or a pharmaceutically acceptable salt thereof, wherein Q is —(CR 1 R 2 ) m -A and m is 1, 2 or 3.

11. The compound of claim 1 , or a pharmaceutically acceptable salt thereof, wherein E is methylene, ethylene, or propylene.

12. The compound of claim 1 , or a pharmaceutically acceptable salt thereof, wherein R 4 , R 5 , R 6 , R 7 , R 8 , R 9 , R 10 and R 11 are each, independently, H, C 1-10 alkyl or C 1-10 haloalkyl.

13. The compound of claim 1 , or a pharmaceutically acceptable salt thereof, wherein R 4 , R 5 , R 6 , R 7 , R 8 , R 9 , R 10 and R 11 are each H.

14. The compound of claim 1 , or a pharmaceutically acceptable salt thereof, wherein R 12 and R 13 are each H.

15. The compound of claim 1 having Formula II:

or a pharmaceutically acceptable salt thereof.

17. A composition comprising a compound of claim 1 , or a pharmaceutically acceptable salt thereof, and a pharmaceutically acceptable carrier.

A method of treating a disease in a patient comprising administering to the patient a therapeutically effective amount of a compound according to claim 1 , or a pharmaceutically acceptable salt thereof, wherein said disease is obesity, glucose intolerance, insulin resistance, hyperglycemia, hypertension, hyperlipidemia, depression, dementia, glaucoma, osteoporosis, inflammation, heart failure, atherosclerosis, arteriosclerosis, coronary artery disease, thrombosis, angina, peripheral vascular disease, vascular wall damage, stroke, dyslipidemia, hyperlipoproteinaemia, diabetic dyslipidemia, mixed dyslipidemia, hypercholesterolemia, hypertriglyceridemia, metabolic syndrome or general aldosterone-related target organ damage.

29. A method of treating a type 2 diabetes in a patient comprising administering to the patient a therapeutically effective amount of a compound according to claim 20 , or a pharmaceutically acceptable salt thereof.

30. A method of treating a type 2 diabetes in a patient comprising administering to the patient a therapeutically effective amount of a compound according to claim 21 , or a pharmaceutically acceptable salt thereof.

31. A method of treating a type 2 diabetes in a patient comprising administering to the patient a therapeutically effective amount of a compound according to claim 23 , or a pharmaceutically acceptable salt thereof.
