Methods and compositions for tissue regeneration
View Patent ↗Disclosed is a method of treating a wound comprising applying a composition to a wound site on a patient in need of wound treatment, wherein the composition comprises mitotically inactive allogeneic cells that secrete one or more biologically active molecules selected from the group consisting of GM-CSF, VEGF, KGF, bFGF, TGFβ, angiopoietin, EGF, IL-Iβ, IL-6 IL-8, TGFα, and TNFα, and wherein the cells are keratinocytes or fibroblasts, or mixtures thereof.
1. A method of treating a wound comprising applying a composition to a wound site on a patient in need of wound treatment, wherein the composition comprises mitotically inactive allogeneic cells that secrete one or more biologically active molecules selected from the group consisting of GM-CSF, VEGF, KGF, bFGF, TGFβ, angiopoietin, EGF, IL-Iβ, IL-6, IL-8, TGFα, and TNFα, and wherein the cells are keratinocytes or fibroblasts, or mixtures thereof.
2. The method of claim 1 , wherein the cells are mitotically inactivated by irradiation.
3. The method of claim 1 , wherein the cells are differentiated.
4. The method of claim 1 , wherein the concentration of cells are in the range of about 1×10 3 cells/μl to about 50×10 3 cells/μl.
5. The method of claim 1 , wherein the cells are a mixture of keratinocytes and fibroblasts and the ratio of keratinocytes to fibroblasts is 1:9.
6. The method of claim 1 , wherein the composition further comprises a cryoprotectant or a buffered solution, or mixtures thereof.
7. The method of claim 6 , wherein the composition comprises a cryoprotectant and the cryoprotectant is glycerol.
8. The method of claim 6 , wherein the cryoprotectant is glycerol and human serum albumin solution.
9. The method of claim 6 , wherein the composition comprises a buffered solution and the buffered solution is HBSS.
10. The method of claim 9 , wherein the HBSS comprises Ca ++ and Mg ++ .
11. The method of claim 1 , wherein the composition is topically applied to the wound site.
12. The method of claim 11 , wherein the composition is topically applied to the wound site by spraying.
13. The method of claim 1 , wherein the wound is a cutaneous wound.
14. The method of claim 13 , wherein the cutaneous wound is a chronic wound or a burn wound.
15. The method of claim 14 , wherein the chronic wound is a neuropathic ulcer, a pressure sore, an arterial ulcer, a venous ulcer, a mixed arterio-venous ulcer, or a diabetic ulcer.
16. The method of claim 15 , wherein the chronic wound is a venous ulcer.
17. The method of claim 16 , wherein the venous ulcer is a venous leg ulcer.
