Abstract

The present invention is directed to compounds, compositions, and methods useful for delivering polynucleotides or other cell-impermeable molecules to mammalian cells. Described are polyconjugates systems that incorporate targeting, anti-opsonization, anti-aggregation, and transfection activities into small biocompatible in vivo delivery vehicles. The use of multiple reversible or labile linkages connecting component parts provides for physiologically responsive activity modulation.

Claims (42)

1. An RNA interference polynucleotide delivery polyconjugate for delivering the RNA interference polynucleotide to a mammalian cell in vivo comprising: a membrane inactive reversibly modified amphipathic membrane active random copolymer

poly(A a -co-(B b -graft-(C c ;D d ;E e )))

wherein

A is a hydrophobic group-containing monomer,

B is a primary amine-containing monomer,

C comprises an RNA interference polynucleotide linked to a primary amine-containing monomer via a physiologically labile linkage orthogonal to maleamate,

D comprises a targeting ligand linked to a primary amine-containing monomer via a maleamate linkage having the structure —NH—CO—C(R)═C(L)—COO − or —NH—CO—C(L)═C(R)—COO − , wherein the nitrogen is derived from the primary amine, L is charge neutral and comprises the targeting ligand and R is an alkyl group,

E comprises a steric stabilizer linked to a primary amine-containing monomer via a maleamate linkage having the structure —NH—CO—C(R′)═C(SS)—COO − or —NH—CO—C(SS)═C(R′)—COO − , wherein the nitrogen is derived from the primary amine, SS is charge neutral and comprises the steric stabilizer and R′ is an alkyl group,

a is an integer greater than zero,

b is an integer greater than or equal to two,

c is an integer greater than or equal to one,

d is an integer greater than or equal to zero,

e is an integer greater than or equal to zero,

the value of d+e is greater than 50% of the value of b, and

poly(A a -co-B b ) is membrane active.

2. The polyconjugate of claim 1 wherein the RNA interference polynucleotide is selected from the group consisting of: siRNA, miRNA, and dicer substrate.

3. The polyconjugate of claim 2 wherein R and R′ are independently selected from the group consisting of: CH 2 and CH 2 CH 3 .

4. The polyconjugate of claim 2 wherein L is uncharged.

5. The polyconjugate of claim 2 wherein SS is uncharged.

6. The polyconjugate of claim 2 wherein L is selected from the group consisting of: compound with affinity to cell surface protein, cell receptor ligand, and antibody with affinity to a cell surface molecule.

7. The polyconjugate of claim 6 wherein the cell receptor ligand comprises an asialoglycoprotein receptor ligand.

8. The polyconjugate of claim 7 wherein the asialoglycoprotein receptor ligand is selected from the group consisting of: saccharide, galactose, galactose derivative, and N-acetylgalactosamine.

9. The polyconjugate of claim 6 wherein the cell receptor ligand is selected from the group consisting of mannose, mannose derivative, vitamin, folate, biotin, peptide, RGD-containing peptide, insulin, aptamer, and EGF.

10. The polyconjugate of claim 5 wherein SS is a polyethylene glycol.

11. The polyconjugate of claim 10 wherein the polyethylene glycol contains 2 to 500 ethylene glycol units.

12. The polyconjugate of claim 1 wherein the value of d+e is greater than 70% of the value of b.

13. The polyconjugate of claim 12 wherein the value of d+e is greater than 75% of the value of b.

14. The polyconjugate of claim 13 wherein the value of d+e is greater than 80% of the value of b.

15. The polyconjugate of claim 14 wherein the value of d+e is greater than 90% of the value of b.

16. The polyconjugate of claim 1 wherein the ratio of d to e is 1:2 to 2:1.

17. The polyconjugate of claim 1 wherein d is zero.

18. The polyconjugate of claim 1 wherein e is zero.

19. The polyconjugate of claim 1 wherein the cell is a liver cell.

20. A method for delivering an RNA interference polynucleotide to a cell comprising: injecting the polyconjugate of claim 1 into a mammal.

21. The method of claim 20 wherein the polyconjugate is administered to the mammal in a pharmaceutically acceptable carrier or diluent.

22. The method of claim 21 wherein the polyconjugate is administered to the mammal via intravascular, intravenous, intraarterial, intramuscular, intraparenchymal, intradermal, subdermal, subcutaneous, intratumoral, intraperitoneal, intrathecal, subdural, epidural, or intralymphatic injection.

23. A method for forming a mammalian in vivo RNA interference polynucleotide delivery polyconjugate comprising:

a) forming a membrane active polyamine;

b) forming a first neutral masking agent comprising a disubstituted maleic anhydride containing a targeting ligand;

c) forming a second neutral masking agent comprising a disubstituted maleic anhydride containing a steric stabilizer;

d) linking the RNA interference polynucleotide to the polyamine via a physiologically labile covalent bond that is orthogonal to a maleamate linkage;

e) reversibly inhibiting membrane activity of the membrane active polyamine wherein the inhibiting consists of modifying 50% or more of the amines on the polyamine by reacting the polyamine with the first and second neutral masking agents thereby linking a plurality of targeting ligands and a plurality of steric stabilizers to the membrane active polymer via physiologically pH-labile disubstituted maleamate linkages.
