Abstract

Newly identified mammalian taste-cell-specific G protein-coupled receptors, and the genes and cDNA encoding said receptors are described. Specifically, T1R G protein-coupled receptors active in taste signaling, and the genes and cDNA encoding the same, are described, along with methods for isolating such genes and for isolating and expressing such receptors. Methods for representing taste perception of a particular taste stimulus in a mammal are also described, as are methods for generating novel molecules or combinations of molecules that elicit a predetermined taste perception in a mammal, and methods for simulating one or more tastes. Further, methods for stimulating or blocking taste perception in a mammal are also disclosed.

Claims (17)

1. An isolated or recombinant polypeptide, wherein said polypeptide possesses at least 90% sequence identity to the entire extracellular or transmembrane regions of the human T1R2 polypeptide contained in SEQ ID NO:21.

2. An isolated or recombinant polypeptide, wherein said polypeptide possesses at least 95% sequence identity to the entire extracellular or transmembrane regions of the human T1R2 polypeptide contained in SEQ ID NO:21.

3. The polypeptide of claim 1 that possesses at least 90% sequence identity to the extracellular region of the human T1R2 polypeptide contained in SEQ ID NO:21.

4. The polypeptide of claim 1 that possesses at least 90% sequence identity to the transmembrane region of the human T1R2 polypeptide contained in SEQ ID NO:21.

5. The polypeptide of claim 1 that possesses at least 95% sequence identity to the extracellular region of the human T1R2 polypeptide contained in SEQ ID NO:21.

6. The polypeptide of claim 1 that possesses at least 95% sequence identity to the transmembrane region of the human T1R2 polypeptide contained in SEQ ID NO:21.

7. The polypeptide of claim 1 , which is fused to part or all of another GPCR polypeptide.

8. The polypeptide of claim 2 , which is fused to part or all of another GPCR polypeptide.

9. The polypeptide of claim 3 , which is fused to part or all of another GPCR polypeptide.

10. The polypeptide of claim 4 , which is fused to part or all of another GPCR polypeptide.

11. The polypeptide of claim 5 , which is fused to part or all of another GPCR polypeptide.

12. The polypeptide of claim 6 , which is fused to part or all of another GPCR polypeptide.

13. The polypeptide of claim 1 , wherein said polypeptide comprises a polypeptide that which possesses at least 90% sequence identity to the entire extracellular region of the human T1R2 polypeptide contained in SEQ ID NO:21, which is fused to a polypeptide comprising, the transmembrane region of another GPCR polypeptide.

14. The polypeptide of claim 1 , which is fused to part or all of another GPCR polypeptide which is not a T1R polypeptide.

15. The polypeptide of claim 1 , which is fused to a selectable marker.

16. The polypeptide of claim 15 , wherein the selectable marker is a Green fluorescent polypeptide.

17. The polypeptide of claim 1 , wherein said polypeptide comprises a polypeptide that which possesses at least 90% sequence identity to the entire transmembrane region of the human T1R2 polypeptide contained in SEQ ID NO:21, which is fused to a polypeptide comprising the extracellular region of another GPCR polypeptide.
