Abstract

An adjuvant composition which comprises an anionic macromolecule component particularly an ionic polysaccharide such as DEAE-dextran, and a saponin component, particularly an immunostimulating complex component. Immunogenic compositions comprising an immunogen and this adjuvant composition are also disclosed together with methods of use thereof.

Claims (10)

1. A method of eliciting an effective immune response in an animal against LHRH, comprising administering to an animal an effective amount of a liquid vaccine composition that comprises:

(1) an LHRH-diphtheria toxoid conjugate;

(2) an adjuvant that comprises (a) an ionic polysaccharide, and (b) immunostimulating complexes that comprise a saponin, a sterol, and a phospholipid; and

(3) optionally, a pharmaceutically acceptable carrier.

2. A method of eliciting an effective immune response in an animal, comprising administering to an animal an effective amount of a single dose of the vaccine composition of claim 1 .

3. The method of claim 1 wherein the adjuvant provides a mass ratio of ionic polysaccharide to immunostimulating complex in the range of about 100 to about 140.

4. The method of claim 1 , wherein the immunostimulating complex is a protein-free immunostimulating complex.

5. The method of claim 1 , wherein the ionic polysaccharide is DEAE-dextran or QAE-dextran.

6. The method of claim 1 , wherein the vaccine composition provides from about 5 to about 500 μg of LHRH-diphtheria toxoid conjugate, from about 5 to about 500 mg of the ionic polysaccharide, and from about 40 to about 4000 μg of the immunostimulating complex.

7. A method of eliciting an effective immune response in an animal, comprising administering to an animal an effective amount of two or more doses of the vaccine composition of claim 1 .
