Fusion molecules and IL-15 variants
View Patent ↗The instant invention provides soluble fusion protein complexes and IL-15 variants that have therapeutic and diagnostic use, and methods for making such proteins. The instant invention additionally provides methods of stimulating or suppressing immune responses in a mammal using the fusion protein complexes and IL-15 variants of the invention.
1. A method of stimulating immune responses in a mammal comprising administering to the mammal an effective amount of an isolated human interleukin 15 (hIL-15) variant comprising an amino acid substitution at position 72 of a mature hIL-15 sequence, wherein the amino acid substitution results in an hIL-15 variant that has IL-15 agonist activity and increased binding activity for the IL-15 receptor beta/gamma chain (IL-15RβγC) receptors compared to the native hIL-15 polypeptide.
2. The method of claim 1 , wherein the isolated hIL-15 variant comprises an amino acid substitution of N to D at position 72 of the mature human IL-15 sequence.
3. The method of claim 2 , wherein said binding activity of said hIL-15 variant for the IL-15RβγC receptors is increased by at least 3-fold compared to the native hIL-15 polypeptide.
4. The method of claim 2 , wherein said hIL-15 variant increases proliferation of an IL15R-bearing cell compared to the native hIL-15 polypeptide.
5. The method of claim 4 , wherein said IL15R-bearing cell comprises a T cell, and wherein said IL-15 variant increases proliferation of said T cell by at least 3-fold compared to the native hIL-15 polypeptide.
6. The method of claim 1 , wherein said mammal is a human.
7. The method of claim 1 , wherein said mature hIL-15 sequence consists of the amino acid sequence set forth in SEQ ID NO: 1.
8. The method of claim 1 , wherein the isolated hIL-15 variant comprises an amino acid substitution of N to A at position 72 of the mature human IL-15 sequence.
