Abstract

The present invention relates to the use of a quinazolinone compound of the formula (I) wherein R 1 , R 2 , R 3 , R 4 , R 5 and m are as defined in the specification and in the claims, in free form or in salt form, and, where possible, in acid addition salt form, as a vanilloid.

Claims (33)

1. A method for modulating vanilloid receptor 1 activity, comprising administering to a mammal in need thereof a therapeutically effective amount of a quinazolinone compound of the formula (Ia)

wherein

R 1 is C 2 -C 6 alkyl, (C 1 -C 6 alkyl)C 1 -C 6 alkyl, di-(C 1 -C 6 alkyl)C 1 -C 6 alkyl, or C 3 -C 6 cycloalkyl;

each R 2 , independently, is halogen, C 1 -C 6 alkyl, halogen-substituted C 1 -C 6 alkyl, hydroxyC 1 -C 6 alkyl, cyano or a group —C(═O)—R 2a , where R 2a is C 1 -C 6 alkyl;

R 3 is hydrogen, halogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, hydroxy, hydroxy-substituted C 1 -C 6 alkyl, C 1 -C 6 alkoxy, C 3 -C 6 cycloalkyl, cyano, —C(C═O)H, phenyl, (C 3 -C 6 cycloalkyl)C 1 -C 6 alkoxy, (C 1 -C 6 alkoxycarbonylamino)C 1 -C 6 alkoxy or (C 1 -C 6 alkylcarbonylamino)C 1 -C 6 alkoxy;

R 4 is hydroxy, esterified hydroxy, etherified hydroxy, a group

or a group

where R 4a is halogen-substituted C 1 -C 6 alkyl,

or a group

where R 4b is benzyl or phenylethyl;

and

m is 1 or 2,

in free base form or in pharmaceutically acceptable salt form.

2. The method according to claim 1 , wherein

R 1 is C 2 -C 6 alkyl, (C 1 -C 6 alkyl)C 1 -C 6 alkyl, di-(C 1 -C 6 alkyl)C 1 -C 6 alkyl, or C 3 -C 6 cycloalkyl;

each R 2 , independently, is cyano or a group —C(═O)—R 2a , where R 2a is C 1 -C 6 alkyl;

R 3 is hydrogen, halogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, hydroxy, hydroxy-substituted C 1 -C 6 alkyl, C 1 -C 6 alkoxy, C 3 -C 6 cycloalkyl, cyano, —C(C═O)H, phenyl, (C 3 -C 6 cycloalkyl)C 1 -C 6 alkoxy, (C 1 -C 6 alkoxycarbonylamino)C 1 -C 6 alkoxy or (C 1 -C 6 alkylcarbonylamino)C 1 -C 6 alkoxy;

R 4 is hydroxy, etherified hydroxy, a group

or a group

where R 4a is halogen-substituted C 1 -C 6 alkyl,

or a group

where R 4b is benzyl or phenylethyl; and

m is 1 or 2,

in free base form or in pharmaceutically acceptable salt form.

3. The method according to claim 1 , wherein the compound of formula (Ia) is selected from

3-(4-chlorophenyl)-7-hydroxy-2-isopropyl-3H-quinazolin-4-one,

in free base form or in pharmaceutically acceptable salt form.

4. The method according to claim 1 , wherein the compound of formula (Ia) is

in free base form or in pharmaceutically acceptable salt form.

5. The method according to claim 1 , wherein the compound of formula (Ia) is

in free base form or in pharmaceutically acceptable salt form.

6. The method according to claim 1 , wherein the compound of formula (Ia) is

in free base form or in pharmaceutically acceptable salt form.
