Crude biological derivatives competent for nucleic acid detection
View Patent ↗The invention relates to methods for the detection of a specific sequence of RNA in a cell or tissue sample. The invention also relates to methods to enzymatically manipulate the RNA in a crude cell lysate in a number of applications.
1. A method for measuring a level of RNA in a sample of cells using real time PCR, the method comprising:
preparing an admixture of the sample, a proteinase K and a deoxyribonuclease, wherein the concentration of cells in the admixture is up to 2500 cells per μl;
incubating the admixture under conditions where the proteinase K and the deoxyribonuclease are active to produce a cell lysate;
incubating the cell lysate at a temperature up to about 75° C. wherein the proteinase K and the deoxyribonuclease are inactivated;
incubating a volume of cell lysate with at least one reverse transcriptase under conditions to allow reverse transcription to produce cDNA,
amplifying the cDNA using real-time PCR to produce amplified cDNA,
measuring the amount of amplified cDNA, and
calculating the level of RNA based on the measured amount of amplified cDNA.
2. The method of claim 1 , wherein the cells are eukaryotic cells.
3. The method of claim 2 , wherein the eukaryotic cells are human cells.
4. The method of claim 1 , wherein the cells are obtained from a cell culture.
5. The method of claim 1 , wherein incubating with reverse transcriptase and amplifying the cDNA provide for one-step RT-PCR.
6. The method of claim 1 , wherein the cells are obtained from a subject.
7. The method of claim 1 wherein the concentration of cells in the admixture is from 4 cells/μl to 2500 cells/μl.
8. A method for measuring a level of RNA in a sample of cells using real time PCR, the method comprising:
preparing an admixture of the sample, a proteinase K and a deoxyribonuclease, wherein the concentration of cells in the admixture is up to 2500 cells per μl;
incubating the admixture under conditions where the proteinase K and the deoxyribonuclease are active to produce a cell lysate;
incubating the cell lysate at a temperature up to about 75° C. wherein the proteinase K and the deoxyribonuclease are inactivated;
incubating a volume of cell lysate with at least one reverse transcriptase under conditions to allow reverse transcription to produce cDNA,
amplifying the cDNA using real-time PCR to produce amplified cDNA,
measuring the amount of amplified cDNA, and
calculating the level of RNA based on the measured amount of amplified cDNA wherein the cells are obtained from a sample of body fluid.
