Abstract

Recombinant vectors comprise simian adenovirus A1321 (SAdV-A1321), SAdV-A1325, SAdV-A1295, SAdV-A1309, SAdV-A1316, and/or SAdV-A1322 sequences and a heterologous gene under the control of regulatory sequences. A cell line which expresses simian adenovirus SAdV-A1321, SAdV-A1325, SAdV-A1295, SAdV-A1309, SAdV-A1316, and/or SAdV-A1322 gene(s) is also disclosed. Methods of using the vectors and cell lines are provided.

Claims (38)

1. An adenovirus having a capsid comprising a hexon protein, a penton protein, and a fiber protein, wherein said hexon protein is the hexon protein of SAdV-A1309 with the amino acids 1 to 939 of SEQ ID NO: 96, and said fiber protein is the fiber protein of SAdV-A1309 with the amino acids 1 to 425 of SEQ ID NO: 106;

said capsid encapsidating a heterologous nucleic acid comprising a gene operably linked to expression control sequences which direct transcription, translation, and/or expression thereof in a host cell.

2. The adenovirus according to claim 1 , further comprising a 5′ and a 3′ adenovirus cis-element necessary for replication and encapsidation.

3. The adenovirus according to claim 1 , wherein said adenovirus lacks all or a part of the E1 gene.

4. The adenovirus according to claim 3 , wherein said adenovirus is replication—defective.

5. The adenovirus according to claim 1 , wherein said capsid is a hybrid capsid.

6. The adenovirus according to claim 5 , wherein said hybrid capsid comprises at least one capsid protein from an adenovirus selected from SAdV-A1321, SAdV-A1325, SAdV-A1316, and SAdV-1322.

7. A composition comprising the adenovirus according to claim 1 in a pharmaceutically acceptable carrier.

8. A composition comprising the adenovirus according to claim 1 , said adenovirus further comprising at least one simian adenovirus protein selected from the group consisting of:

E1a, SEQ ID NO: 113;

E1b, small T/19K, SEQ ID NO: 87;

E1b, large T/55K, SEQ ID NO: 108;

IX, SEQ ID NO: 88;

52/55D, SEQ ID NO: 89;

IIIa, SEQ ID NO: 90;

Penton, SEQ ID NO: 91;

VII, SEQ ID NO: 92;

V, SEQ ID NO: 93;

pX, SEQ ID NO: 94;

VI, SEQ ID NO: 95;

Endoprotease, SEQ ID NO: 97;

100 kD, SEQ ID NO: 98;

22 kD, SEQ ID NO: 109;

VIII, SEQ ID NO: 99;

E3/12.5 K, SEQ ID NO: 100;

CR1-alpha SEQ ID NO: 110;

gp19K, SEQ ID NO: 101;

CR1-beta, SEQ ID NO: 102;

CR1-gamma, SEQ ID NO: 103;

CR1-delta, SEQ ID NO: 104;

RID-beta, SEQ ID NO: 105; and

E3/14.7K, SEQ ID NO: 111.

9. The adenovirus according to claim 1 , wherein said penton protein is the penton protein of SAdV-A1309 with the amino acids 1 to 540 of SEQ ID NO: 91.

10. A vector comprising at least one simian adenovirus nucleic acid sequence selected from the group consisting of:

(a) the open reading frame for the penton of SAdV-A1309, nucleotides 13897 to 15516 of SEQ ID NO: 86;

(b) the open reading frame for the hexon of SAdV-A1309, nucleotides 18348 to 21164 of SEQ ID NO: 86; and

(c) the open reading frame for the fiber protein of SAdV-A1309, nucleotides 32137 to 33411 of SEQ ID NO: 86;

said vector further comprising a heterologous gene operably linked to expression control sequences.
